Soilless cultivation has increased in China. Like vegetables or ornamental plants, tomatoes can be cultivated in soilless culture systems. Fusarium wilt (FW) is an economic tomato disease. The nature and amount of volatile organic compounds in Trichoderma asperellum PT-15 were investigated, and 6-pentyl-α-pyrone (6-PP) was detected. Furthermore, the effect of 6-PP on Fusarium oxysporum HF-26 was evaluated. Results revealed that 25 mg/L 6-PP was the optimal concentration inhibiting F. oxysporum HF-26 and that the content of fusaric acid decreased considerably compared to that of the control. FUB1, FUB4, and FUB10 toxin synthesis and transport genes were downregulated. Additionally, VelA, velB, and LaeA genes were downregulated, reducing F. oxysporum mycelial growth and hyphae formation. 6-PP was added to the soilless culture solution in a greenhouse experiment. The results showed that an antifungal–nutrient solution containing 25 mg/L 6-PP significantly suppressed FW with 70.71% efficacy and a 27.23% disease index (DI), which were higher efficacy and lower DI than that of the control. Furthermore, treatment with an antifungal–nutrient solution containing 6-PP increased the levels of defence enzymes 24 h post-inoculation (hpi) compared with those at other time periods. The relative expression levels of the PR1, NPR1, PR2, and PR5 genes were considerably upregulated at 24 hpi.

Potatoes are among the four most important staple crops worldwide. Verticillium wilt in potatoes caused by Verticillium dahliae is a devastating disease that is difficult to control. To identify potential avenues for disease control, the pathogenicity of 72 V. dahliae isolates was tested here. We also tested the resistance to the most virulent isolate (Vd-36) induced by the attenuated isolate Gibellulopsis nigrescens Vn-1 in potatoes. Induction of Verticillium wilt resistance in potatoes was strongest when using attenuated isolate Vn-1 to inoculate potatoes with a spore suspension concentration of 1 × 106 conidia mL−1, followed by infection with isolate Vd-36 at 5 d intervals. After incubation of potatoes with the attenuated isolate Vn-1 followed by isolate Vd-36, reactive oxygen species (ROS) and hydrogen peroxide (H2O2) were produced and accumulated in potato leaves 12 h post-inoculation. The changes in respective defense enzymes, except phenylalanine ammonia-lyase, were consistent with the changes in ROS and H2O2 levels. Furthermore, the content of salicylic acid (SA) in inoculated plants was higher than that in the control, and biosynthesis-related genes StNPR1, StPR1b, StPR2, StPR5 were activated. However, there was no significant difference in the jasmonic acid and ethylene (JA/ET) content between the treatment and control groups. These results demonstrated that the attenuated isolate Vn-1 enhanced resistance to Verticillium wilt by inducing the SA signalling pathway and weakly activating the JA/ET signalling pathways in potatoes.

(-)-Spirobrassinin, a glucosinolate compound from Brassicaceae plants, has shown inhibitory effects on weeds. However, its specific target sites are not well explored. This study used molecular docking, molecular dynamics simulations, and biological experiments to investigate (-)-Spirobrassinin’s target sites. The inhibitory effects of (-)-Spirobrassinin were observed on various enzymes crucial for plant metabolic pathways, including dihydroxyacid dehydrogenase, 4-hydroxyphenylpyruvate dioxygenase, protoporphyrinogen oxidase, and acetolactate synthase. Additionally, it disrupts the metabolism of vital phytohormones, namely abscisic acid and gibberellin. Molecular dynamics simulations revealed stable interactions between (-)-Spirobrassinin and specific residues (Phe270 and Phe261) of the photosystem II D1, involving electrostatic, hydrophobic, and van der Waals forces. This study provides valuable insights into (-)-Spirobrassinin’s mode of action, highlighting its potential as a natural herbicide from Brassicaceae plants.

Potato Fusarium Wilt is a soil-borne fungal disease that can seriously harm potatoes throughout their growth period and occurs at different degrees in major potato-producing areas in China. To reduce the use of chemical agents and improve the effect of biocontrol agents, the inhibitory effects of the fermentation broth of Bacillus subtilis ZWZ-19 (B) and Trichoderma asperellum PT-29 (T) on Fusarium oxysporum were compared under single-culture and co-culture conditions. Furthermore, metabolomic analysis of the fermentation broths was conducted. The results showed that the inhibitory effect of the co-culture fermentation broth with an inoculation ratio of 1:1 (B1T1) was better than that of the separately cultured fermentation broths and had the best control effect in a potted experiment. Using LC-MS analysis, 134 metabolites were determined and classified into different types of amino acids. Furthermore, 10 metabolic pathways had the most significant variations, and 12 were related to amino acid metabolism in the KEGG analysis. A correlation analysis of the 79 differential metabolites generated through the comprehensive comparison between B, T, and B1T1 was conducted, and the results showed that highly abundant amino acids in B1T1 were correlated with amino acids in B, but not in T.

The unreasonable use of chemical pesticides has caused serious damage to crops and the ecological environment. The botanical pesticide matrine has attracted attention as an environmentally friendly pesticide. Compared with traditional spraying methods, unmanned aerial vehicle (UAV) spraying has the advantages of safety, rapidity, uniform droplets, low dosages, and no terrain or crop restrictions. In this study, matrine OD was prepared according to the application requirements of flight prevention preparations using three different emulsifiers. The stability, wettability, particle size and distribution, and spraying performance of matrine OD were studied. The results indicated that when the amount of emulsifier was 8%, the three types of matrine OD had good stability. The stability, wettability, particle size and distribution, and spray performance of the suspension prepared using emulsifier VO/03 were better than the other two emulsifiers. Therefore, matrine OD prepared using 8% VO/03 could be used for ultra-low-volume sprays and aerial applications. In this study, we provide a theoretical basis and technical guidance to develop pesticide formulations for aerial applications.

Gibellulopsis nigrescens, previously classified in the Verticillium genus until 2007, is an attenuated pathogen known to provide cross-protection against Verticillium wilt in various crops. To investigate the potential mechanisms underlying its reduced virulence, we conducted genome sequencing, annotation, and a comparative genome analysis of G. nigrescens GnVn.1 (GnVn.1), an attenuated strain isolated from sunflower. The genome sequencing and annotation results revealed that the GnVn.1 genome consists of 22 contigs, with a total size of 31.79 Mb. We predicted 10,876 genes, resulting in a gene density of 342 genes per Mb. The pathogenicity gene prediction results indicated 1733 high-confidence pathogenicity factors (HCPFs), 895 carbohydrate-active enzymes (CAZys), and 359 effectors. Moreover, we predicted 40 secondary metabolite clusters (SMCs). The comparative genome analysis indicated that GnVn.1 contains more CAZys, SMCs, predicted effectors, and HCPF genes than Verticillium dahliae (VdLs.17) and Verticillium alfalfae (VaMas.102). The core–pan analysis results showed that GnVn.1 had more specific HCPFs, effectors, CAZys, and secreted protein (SP) genes, and lost many critical pathogenic genes compared to VdLs.17 and VaMs.102. Our results indicate that the GnVn.1 genome harbors more pathogenicity-related genes than the VdLs.17 and VaMs.102 genomes. These abundant genes may play critical roles in regulating virulence. The loss of critical pathogenic genes causes weak virulence and confers biocontrol strategies to GnVn.1.
