Biphenyls are unique phytoalexins de novo synthesized in plants in response to pathogen attack. These compounds are found in Maloideae, a subfamily of the Rosaceae. The anti-microbial activities of biphenyls have been reported in a number of studies and they appear to represent an important defense strategy against pathogens common in the Maloideae, such as species in Malus, Pyrus, Sorbus, and Chaenomeles. Here, cell suspension cultures of Sorbus pohuashanensis were established to study biphenyl phytoalexins formation after yeast extract (YE) treatment. An ultra-performance liquid chromatography (UPLC) method coupled with quadrupole time of flight mass spectrometry (Q-TOF-MS) LC−MS/MS was applied to determine the time course of these biphenyl biomarkers accumulation in YE-treated S. pohuashanensis suspension cells. The results of quantitative analyses show the content of Noraucuparin, 2′-Hydroxyaucuparin, and their glycosides initially increased, then decreased over time. The Noraucuparin content reached its highest (225.76 μg·g−1) at 18 h after treatment, 6 hours earlier than that of Noraucuparin 5-O-β-d-glucopyranoside. The content of 2′-Hydroxyaucuparin reached its highest (422.75 μg·g−1) at 30 h after treatment, also earlier than that of its glycoside. The understanding of phytoalexin metabolism in this study may provide a basis for improving Maloideae resistance to pathogens.

Notopterygium incisum is an important Chinese medicinal plant. Its mature seeds have underdeveloped embryos and are physiological dormant. We found the seeds with full developed embryos can germinate after treated by fluridone (FL), an inhibitor of abscisic acid (ABA). In order to understand the molecular mechanisms underlying seed dormancy release by FL, we compared the transcriptomic changes in dormancy release induced by two different methods, FL and cold stratification (CS) in N. incisum. We further analyzed the gene expression patterns involved in seed germination and dormancy using quantitative reverse-transcription PCR. RNA-sequence analysis revealed more dramatic changes in the transcriptomes of FL than those in CS, particularly for genes involved in the biosynthesis and regulation of gibberellins (GAs) and ABA. The down-regulation of ABA biosynthesis genes and the dramatic up-regulation of NiCYP707As, an ABA catabolic gene, contributed to the reduced ABA levels in FL. The increased GA3 levels in CS-treated seeds were due to the up-regulation of NiGA3OX. Both NiABI5 (a positive ABA regulator) and NiGAI (a negative regulator of GA) were down-regulated in FL and CS. The upregulation of strigolactones (SLs; the metabolites with the same precursor as ABA) biosynthesis and regulatory genes in both FL- and CS-treated seeds indicates that SLs contribute positively to seed dormancy release in N. incisum. Our results indicated that FL- and CS-seed dormancy release possibly depends on two totally different mechanisms: alleviation of the effects of ABA and potentiation of the effects of GA, respectively. However, NiABI5 and NiGAI probably function as common factors integrating the effects of ABA and GA on seed dormancy release.

Salvia miltiorrhiza (S. miltiorrhiza) Bunge is one of the most economically important medicinal crops in China. In traditional Chinese medicine, its root is used as an important ingredient in formulas for treatment of atherosclerosis-related disorders. The continuous cropping of S. miltiorrhiza increases the proportion of dried seedlings and decreases the biomass of the shoots and roots and the contents of active components. In this study, three field experiments were conducted to investigate the effects of Glomus mosseae (G. mosseae) inoculation on the growth and contents of active ingredients and nutrients in continuously cropped S. miltiorrhiza. The results showed that inoculation with G. mosseae increased the shoot biomass of S. miltiorrhiza by 48.1% and the root biomass by 39.2%, and decreased the dried seedling rate by nearly 75%. Inoculation with G. mosseae also increased the salvianolic acid B concentration by 21.9% in the shoots and 9.2% in the roots of S. miltiorrhiza, and also significantly increased Mn concentration in the roots and shoots (by 65.1% and 93.4%, respectively) and Fe concentration in the roots (by 75%). The accumulation of salvianolic acid B, Mn, and Fe in G. mosseae inoculated S. miltiorrhiza may be a mechanism that imparts tolerance to continuous cropping. Inoculation of S. miltiorrhiza with G. mosseae can serve as an effective approach of biocontrol to improve the performance of continuously cropped S. miltiorrhiza.

Four new diterpenoids, isodosins A–D (1–4), together with nine known compounds (5–13) were isolated and identified from the aerial parts of Isodon serra (Maxim.) Hara. The structures of the new diterpenoids were elucidated based on the analysis of HR-ESI-MS data, 1D/2D-NMR-spectroscopic data, and electronic circular dichroism (ECD) calculations. Cytotoxicities of compounds 2, 3, 5, 6, and 9 against the HepG2 and H1975 cell lines were evaluated with the MTT assay. As a result, compounds 2, 3, and 6 revealed higher levels of cytotoxicity against HepG2 cells than against H1975 cells. Moreover, compund 6 demonstrated the most efficacy in inhibiting the proliferation of HepG2 cells, with an IC50 value of 41.13 ± 3.49 μM. This effect was achieved by inducing apoptosis in a dose-dependent manner. Furthermore, the relationships between the structures and activities of these compounds are briefly discussed.

Background: Cannabidiol (CBD), a unique secondary metabolite of Cannabis sativa L. (cannabis), is widely used in medicine, food, cosmetics, and forage. Elucidation of the transcriptional regulation mechanism can provide a theoretical basis for molecular breeding with higher CBD content. However, the regulatory network for cannabidiol biosynthesis in cannabis is largely unknown. Methods: Transcriptome analysis of four cannabis tissues was conducted to screen differentially expressed genes. Short time-series expression miner (STEM) analysis combined with strain-specific expression correlation analysis was performed to filter core transcription factors (TFs) associated with CBD biosynthetic enzyme genes. Yeast one-hybrid and yeast two-hybrid assays were applied to verify the binding and protein interaction relationships of key TFs, and subcellular localization assay was performed in tobacco leaf. Results: A total of 1314 genes specifically upregulated in female flowers were identified by transcriptome analysis of four different tissues with distinct CBD contents and clustered into 19 profiles using short time-series expression miner (STEM) analysis. Profile 8, including 16 TFs and 6 enzyme genes related to the CBD biosynthesis pathway, demonstrated similar trends in the CBD contents of the four tissues. Among the 16 TFs, 7 exhibited similar strain-specific expression profiles and their expression levels positively correlated with those of the six enzyme genes. Moreover, a yeast one-hybrid assay found that CsMYB6 and CsMYB29 bind to the promoters of four and two CBD biosynthetic enzyme genes, respectively. Meanwhile, a yeast two-hybrid assay revealed that CsMYB6 interacts with two regulators of CBD biosynthesis, CsMYB1 and CsWRKY1, while CsMYB29 interacts with CsWRKY1. In addition, CsMYB29 and CsMYB6 were localized in the nuclei. Conclusions: In summary, the two TFs, CsMYB6 and CsMYB29, identified in this study provide valuable insight into the complex regulatory mechanisms of CBD production in cannabis and offer a genetic resource for the genetic improvement of CBD content.
