The characteristic chemical composition of Nigella seeds is directly linked to their beneficial properties. This study aimed to investigate the phytochemical composition of Nigella sativa seeds using a 100% ethanolic extract using HPLC-ESI-MS/MS. Additionally, it explored the potential biological effects of the extract on female rat reproduction. Follicle Stimulating Hormone (FSH), Luteinizing Hormone (LH), Estrogen (E2), and Progesterone (P4) hormone levels were also assessed, along with the morphological and histological effects of the extract on ovarian, oviductal, and uterine tissues. Molecular docking was performed to understand the extract’s activity and its role in regulating female reproduction by assessing its binding affinity to hormonal receptors. Twenty metabolites, including alkaloids, saponins, terpenes, flavonoids, phenolic acids, and fatty acids, were found in the ethanolic extract of N. sativa seeds through the HPLC-ESI-MS/MS study. The N. sativa seed extract exhibited strong estrogenic and LH-like activities (p < 0.05) with weak FSH-like activity. Furthermore, it increased the serum levels of LH (p < 0.05), P4 hormones (p < 0.001), and E2 (p < 0.0001). Molecular docking results displayed a strong interaction with Erβ, LH, GnRH, and P4 receptors, respectively. Based on these findings, N. sativa seeds demonstrated hormone-like activities, suggesting their potential as a treatment for improving female fertility.

Sonchus oleraceus L. is a leafy vegetable that is usually consumed in the area of the Mediterranean and is a frequently used traditional herb to treat a variety of ailments. Previous studies deduced the potent antioxidant and cytotoxic functions of the different extracts and isolated compounds from S. oleraceus. The current study represents the first instance of chemical profiling and bioactivities of the extracted essential oil (EO) of S. oleraceus. The present investigation set out to identify the chemical components of this EO by means of Gas Chromatography with Flame Ionization Detector (GC-FID) and Gas Chromatography-Mass Spectrometry (G004-MS) techniques; assess the oil’s antioxidant potencies through 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2′-azinobis-(3-ethylbenzothiazoline-6-sulfonate (ABTS) assays; and evaluate the oil’s cytotoxic impact against HepG2 cancer cell lines. The GC-MS chemical profiling revealed the identification of 23 components representing 97.43% of the total oil mass within abundant cyclic ketones (20.15%), nonterpenoidial hydrocarbons (28.77%), and sesquiterpenes (42.19%). The main components were n-nonadecane (28.77%), trans-caryophyllene (23.73%), trans-methyl dihydrojasmonate (19.55%), and cis-cadina-1,4-diene (9.44%). In a dose-dependent manner, this EO demonstrated antioxidant capacities on DPPH and ABTS, with IC50 values of 609.35 and 804.16 µg/mL, respectively, compared to ascorbic acid. Using doxorubicin as a reference therapy, the MTT assay findings revealed that this oil had remarkable inhibitory effects on the proliferation of HepG2 cancer cell lines, with an IC50 of 136.02 µg/mL. More studies were recommended for further investigation of new biological roles for this oil and its main components, along with the construction of action mechanisms based on chemical components.

Nanoparticle engineering via plants (green synthesis) is a promising eco-friendly technique. In this work, a green protocol was applied to the preparation of silver, zinc, and selenium nanoparticle solutions supported by the extracted aerial parts of Pulicaria undulata. The formation of nanoparticles in the solution was characterized using phytochemical analysis, and UV-visible, TEM, and zeta-potential spectroscopy. In addition, various biological activities were investigated for the extract of P. undulata and the produced nanoparticles (selenium, silver, and zinc), including antioxidant, antimicrobial, and cytotoxic activities. The volatile components of the extracted constitute verified the fact that twenty-five volatile components were characterized for the majority of abundant categories for the fatty acids, esters of fatty acids (59.47%), and hydrocarbons (38.19%) of the total area. The antioxidant activity of P. undulata extract and metal nanoparticles was assessed using DPPH assay. The results indicated reduced potency for the metal nanoparticles’ solutions relative to the results for the plant extract. The cytotoxicity of the investigated samples was assessed using an MTT assay against various tumor and normal cell lines with improved cytotoxic potency of the solutions of metal nanoparticles, compared to the plant extract. The antimicrobial activity was also estimated against various bacterial and fungal species. The results confirmed amended potency for inhibiting the growth of microbial species for the solutions of metal nanoparticles when compared to the extracted aerial parts of the plant. The present study showed that green synthetized nanoparticles using P. undulata have various potential bioactivities.

Urtica urens extract was applied for Se/CuO NPs synthesis with investigation of its biological activities and bio-stimulant qualities for Zea mays L. More antimicrobial activity was documented using Se/CuO NPs than U. urens extract against C. albicans, E. coli, S. typhi, B. subtilis, and S. aureus. Moreover less MIC and MBC values of Se/CuO NPs compared to U. urens extract against examined bacteria were recorded. Excellent antioxidant activity of Se/CuO NPs and U. urens extract was documented, with IC50 4.2±0.41 and 5.12 ±0.19 µg/mL, respectively. Anti-hemolysis of U. urens extract was higher than Se/CuO NPs with IC50 4.1±0.12 and 12.26±0.22 µg/mL, respectively. Anticoagulant potential of Se/CuO NPs was better than the U. urens extract via partial thromboplastin time (PPT) and prothrombin time (PT). Se/CuO NPs at 10 ppm caused the highest significant improvement in chlorophyll a and b of maize plants, but the highest value of carotenoids appeared at 20 ppm compared to the control and other treatments. The increase of shoot and root lengths was shown at 10 ppm of Se/CuO NPs by increasing 32.6 and 17.9%, respectively, compared to the control. Thus, novel bimetallic Se/CuO NPs synthesized using U. urens extract displayed antimicrobial, antioxidant, anticoagulant, anti-hemolysis, and plant biostimulant effects.

Essential oils (EOs) are advised by traditional medical systems for the treatment of a variety of disorders worldwide. In many ancient medical systems around the world, Polygonum herbs have been employed as remedies including P. equisetiforme Sm. The EO profile of P. equisetiforme and its bioactivities have yet to be discussed in depth. As a result, the current study aims to investigate the chemical profile and free radical scavenging capacity of P. equisetiforme EO. Hydrodistillation was used to obtain the EO from P. equisetiforme, and gas chromatography–mass spectrometry (GC-MS) was used for analysis. A total of forty-three compounds, including terpenes and sesquiterpenes as the main components (76.13% and 69.06%, respectively), were identified in the oil using the GC-MS analysis. The main constituents of the oil were hexahydrofarnesyl acetone (29.45%), 7-epi-selinene (14.45%), isospathulenol (8.35%), and n-docosane (6.79%). The chemosystematic significance of the plant was established via multivariate assessing, comprising principal component analysis (PCA), hierarchical clustering, and constellation plot, of the EO principal components of the various Polygonum plants. The P. equisetiforme exhibited different associations with the studied Polygonum spp. Then, the scavenging of the free radicals 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2′-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) was used to evaluate the radical scavenging abilities of EO compared with those of vitamin C, a reference antioxidant. P. equisetiforme EO exhibited the scavenging capacity of the DPPH and the ABTS free radical with respective IC50 values of 470.01 and 113.74 mg L−1 compared with vitamin C, and with IC50 values of 39.06 and 26.09 mg L−1, respectively. The in silico studies revealed that the oxygenated sesquiterpenes, especially ar-turmerone, hexahydrofarnesyl acetone, and 5E,9E-farnesyl acetone, exhibited the best fitting with hematopoietic cell kinase (Hck) and human Peroxiredoxin 5 proteins with ΔG values of −6.14 and −4.93, −6.83 and −5.34, and −7.08 and −5.47 kcal/mol, respectively. The major components’ combined or individual effects may be responsible for the antioxidant properties. Therefore, additional extensive studies are advised to characterize the essential compounds as radical scavenger agents, either individually or in combination.
