Insulin-like growth factor 1 (IGF-1) regulates dairy cow reproduction, while the paracrine IGF system locally influences fertility. In both systems, IGF-1 bioactivity is regulated through binding proteins (IGFBPs) inhibiting IGF-1 binding to its receptor (IGF1R). This study aimed to investigate a possible transfer between this endocrine and paracrine system. Therefore, blood and follicular fluid (FF) from postpartum dairy cows were analysed for ß-hydroxybutyrate (BHB), IGF-1, IGFBP-2, -3, -4, -5, and an IGFBP fragment in two study parts. The mRNA expression of IGFBP-2, IGFBP-4, IGF1R, and the pregnancy-associated plasma protein A (PAPP-A) in granulosa cells was measured. The results showed correlations between plasma and FF for IGF-1 (r = 0.57, p < 0.001) and IGFBP-2 (r = −0.57, p < 0.05). Blood BHB negatively correlated with IGF-1 in blood and FF and IGFBP-3, -5 and total IGFBP in blood (IGF-1 plasma: r = −0.26, p < 0.05; FF: r = −0.35, p < 0.05; IGFBP-3: r = −0.64, p = 0.006; IGFBP-5: r = −0.49, p < 0.05; total IGFBP: r = −0.52, p < 0.05). A negative correlation was found between IGFBP-2 expression and IGF-1 concentration in FF (r = −0.97, p = 0.001), while an IGFBP fragment positively correlated with IGF1R-mRNA in FF (r = 0.82, p = 0.042). These findings suggest a transfer and local regulation between the somatotropic axis and the follicular IGF system, linking the metabolic status with local effects on dairy cow fertility.

The present study aimed to evaluate the effects of SOD (superoxide dismutase)-rich melon feed supplement on some performance parameters, serum biochemical and antioxidant indexes, and meat quality characteristics of weaned Tuj lambs. An independent measures design (between groups) was used to determine these effects of treatment. After one week of the adaptation period, twenty-four weaned lambs at the age of 60 ± 5.0 days with a body weight of 23.14 ± 0.5 kg were divided into two groups, i.e., the control group (CON) fed basal diet and experimental group (EXP) fed with basal diet + SOD-rich melon (n = 12 per group). The results revealed a decrement in the (p < 0.05) feed efficiency ratio (5.88 ± 0.40 vs. 6.59 ± 0.86 kg weight gain/kg feed) and higher carcass yield (61.76 ± 0.80 vs. 60.11 ± 1.07%) in the EXP group as compared to the CON group. Additionally, the EXP group showed a significant increase (p < 0.05) in serum glucose and high-density lipoprotein levels, while there was a reduction in cholesterol, triglyceride, and low-density lipoprotein levels when compared to the CON group. The serum malondialdehyde was lowered (5.53 ± 0.47 vs. 5.98 ± 0.79 mmol/L) significantly (p < 0.05), while glutathione concentration was higher (p < 0.05) in the EXP group (17.82 ± 1.51 mmol/L) when compared to the CON group (16.54 ± 1.59 mmol/L). The cooking loss was also significantly (p < 0.05) lower in the EXP group when compared to the CON group. In conclusion, the results indicate that SOD-rich melon supplement (30 g/ton of the concentrate feed) can considerably improve carcass yield, some serum biochemical parameters, and meat quality characteristics in Tuj lambs. Thus, the supplementation of lamb diets with a SOD-rich melon additive may be used as an effective nutritional approach to improve their performance and health.

In vitro fertilization is used to produce embryos from high-genetic-merit cattle. However, these embryos often exhibit inferior quality than those derived in vivo, possibly due to increased oxidative stress. This study investigates whether adding antioxidant polyphenols (resveratrol (RV), chlorogenic acid (CA), ellagic acid (EA)) to the in vitro maturation (IVM) medium at 0.25, 0.5, and 1 μM could improve embryo development. Oxygen consumption and gene expression were evaluated at the blastocyst stage following treatment with 1 μM of each polyphenol. Embryo development (cleavage, blastocyst, and hatched blastocyst rates) and oxygen consumption were not significantly affected by polyphenols. However, RV significantly upregulated the mRNA expression of the antioxidant enzyme glutathione peroxidase 4 (GPX4), while GPX4 expression was significantly downregulated by EA. Expression of other gene markers related to antioxidant defense, apoptosis, development, and metabolism was not significantly affected. The results indicate that applying RV, CA, and EA during bovine oocyte IVM does not enhance in vitro embryo development at the tested concentrations. Given the opposing effects of RV and EA on the expression of GPX4, the effects of those polyphenols regarding the protection of embryos from oxidative stress and potential long-term effects on the offspring remain to be elucidated.

In this project, ejaculates (n=34) were collected from three Brown Swiss bulls using the artificial vagina method, then divided into seven equal parts and diluted to 60×10⁶ spermatozoa/mL using the following extenders: Control, Control+1mM carnosic acid (CR1), Control+3mM carnosic acid (CR3), Control+2.5mM glutamine (G2.5), Control+7.5mM glutamine (G7.5), Control+CR1+G7.5, and Control+CR3+G2.5. All diluted aliquots were frozen after 3 hours of equilibration. Thawed samples were analyzed for motility, plasma membrane and acrosome integrity (PMAI), mitochondrial membrane potential (MMP), and DNA integrity. The Control, G7.5, and G2.5 groups showed the highest total and progressive motility. The highest PMAI value was observed in the Control group, while the lowest was in the CR3 group. The PMAI values of G2.5, G7.5, and G7.5+CR1 were significantly higher than those of CR1, CR3, and CR3+G2.5. There were no statistically significant differences in MMP among the groups. The lowest DNA damage was observed in the Control group and the highest in CR3+G2.5. Control, G2.5, G7.5, and CR1+G7.5 groups had statistically similar kinematic values (VAP, VSL, VCL) and outperformed the others. Although 2.5 mM and 7.5 mM glutamine showed a synergistic interaction, they did not significantly improve spermatological parameters. Carnosic acid appeared to act as an antagonistic antioxidant.
