The aim of the study was to investigate the plasma and muscle pharmacokinetic of enrofloxacin (ENR) and its active metabolite ciprofloxacin (CIP) in Nile tilapia (Oreochromis niloticus) following single intravascular (IV), intraperitoneal (IP), or oral (PO) administration at 30 ± 1 °C. In this study, 234 healthy Nile tilapia (120–150 g) were used. The fish received a single IV, IP, or PO treatment of ENR at a dose of 10 mg/kg. The plasma and muscle tissue concentrations of ENR and CIP were measured using high–performance liquid chromatography with fluorescence detection and were evaluated using non–compartmental analysis. The elimination half–life, volume of distribution at steady state, and total body clearance of ENR were 21.7 h, 2.69 L/kg, and 0.09 L/h/kg, respectively. The peak plasma concentrations of ENR after IP or PO administration were 6.11 and 4.21 µg/mL at 0.25 and 2 h, respectively. The bioavailability of ENR for IP or PO routes was 78% and 86%, respectively. AUC(0–120)muscle/AUC(0–120)plasma ratios following the IV, IP, or PO administrations were 1.43, 1.49, and 1.07, respectively. CIP was detected after all routes, but the AUC0–last ratios of CIP to ENR were <1.0% for plasma and muscle. ENR was detected up to 120 h following the IV, IP, or PO administrations. The long residence time of ENR after single IV, IP, or PO administration ensured the plasma concentration was ≥1 × MIC for bacteria with threshold MIC values of 0.92, 0.72, and 0.80 μg/mL over the whole 120 h observed. However, further studies are necessary to determine the optimum pharmacokinetic/pharmacodynamics data of ENR for the treatment of infections caused by susceptible bacteria in tilapia.

A 45-day feeding study was carried out to assess the immune-stimulatory effects of capsicum oleoresin when added to rainbow trout diets. A total of 450 fish (mean weight: 155.20 ± 1.96 g) were distributed into 400 L tanks (30 fish/tank) across five experimental groups: control (CT, 0%), C7 (0.7%), C14 (1.4%), C21 (2.1%), and C28 (2.8%). Each group consisted of three replicate tanks. At the end of this period, hemato-biochemical parameters, innate immune responses, and immune-related gene expression levels were evaluated, and a histological examination of head kidney and liver sections was conducted. Finally, fish in all groups were challenged with Lactococcus garvieae and observed for an additional 20 days. The results revealed that oleoresin supplementation enhanced the immune responses of the treated fish, which was evidenced by the increased globulin, total protein, respiratory burst activity, and total immunoglobulin levels. The highest expression levels of the il-8, il-1β, TGF-β, and SAA genes was noticed in the C7 group, as compared with the results for the other groups. The IgT gene expression levels were higher in all experimental groups than in the CT group, and this increase was at the highest level in the C28 group. Following the bacterial challenge, all experimental groups displayed higher survival rates compared to that of the CT group. These values were 75.93, 72.22, 46.30, 33.33, and 29.63% in the C7, C14, C21, C28, and CT groups, respectively, with the C7 group displaying the highest survival rate among the groups. The histological examination of liver and head kidney tissues revealed that higher doses (in the C21 and C28 groups) showed an increase in cytoplasmic vacuolization, which causes adverse effects on fish health. However, the C7 group displayed normal histological structure in both tissues. Taken together, the most favorable immune responses were achieved in the C7 group, suggesting that 0.7% oleoresin could be applied to rainbow trout to boost immunity and protect the fish from diseases.

This study was aimed at determining the effects of ethanolic extracts of nettle ( Urtica dioica ) and green olive tree ( Phillyrea latifolia ), used as feed additives, on innate immune responses in gilthead seabream ( Sparus aurata ). Ethanolic extracts from leaves of both plants were incorporated into feeds in two separate experimental setups, and the fish received 0, 50, 100, 200, and 400 mg/kg nettle or green olive tree extracts for 60 days. Growth performance was evaluated on the 30th and 60th days, and hematological parameters as well as the immune responses were assessed on the 60th day. Moreover, the nettle group was challenged with Vibrio anguillarum at the end of the 60th day. Supplementation at different doses increased lysozyme and myeloperoxidase activities significantly in both trials ( p < 0.05). Respiratory burst activity also significantly increased after nettle supplementation ( p < 0.05). It was determined that nettle supplementation resulted in 96% survival rate after the challenge test. In both trials, expression levels of IL-1β , IL-6 , IL-10 , IL-18 , and TNF-α genes were analyzed and evaluated alongside other parameters. Consequently, the findings indicated that nettle, particularly at 100 mg/kg feed, provided the strongest support for immunostimulation and disease resistance in gilthead seabream. In contrast, green olive tree extract showed preliminary immunomodulatory effects that require further confirmation by deeper phytochemical standardization/quantitative characterization and a valid pathogen challenge.
