The wound-healing process is a significant area of interest in the medical field, and it is influenced by both external and patient-specific factors. The aim of this review paper is to highlight the proven wound-healing potential of the biocompounds found in jellyfish (such as polysaccharide compounds, collagen, collagen peptides and amino acids). There are aspects of the wound-healing process that can benefit from polysaccharides (JSPs) and collagen-based materials, as these materials have been shown to limit exposure to bacteria and promote tissue regeneration. A second demonstrated benefit of jellyfish-derived biocompounds is their immunostimulatory effects on growth factors such as (TNF-α), (IFN-γ) and (TGF), which are involved in wound healing. A third benefit of collagens and polysaccharides (JSP) is their antioxidant action. Aspects related to chronic wound care are specifically addressed, and within this general theme, molecular pathways related to tissue regeneration are explored in depth. Only distinct varieties of jellyfish that are specifically enriched in the biocompounds involved in these pathways and live in European marine habitats are presented. The advantages of jellyfish collagens over mammalian collagens are highlighted by the fact that jellyfish collagens are not considered transmitters of diseases (spongiform encephalopathy) or various allergic reactions. Jellyfish collagen extracts stimulate an immune response in vivo without inducing allergic complications. More studies are needed to explore more varieties of jellyfish that can be exploited for their biocomponents, which may be useful in wound healing.

Skin health must be ensured at all times in the case of wounds when the skin is subjected to traumatic actions that require multiple wound-healing measures. Wound healing is a complex, multi-phase biological process critical for restoring skin integrity after trauma. This study investigates the development and evaluation of a novel composite hydrogel formulated from collagen peptides extracted from the jellyfish Rhizostoma pulmo and hydroethanolic extracts from the brown alga Cystoseira barbata, both sourced from the Romanian Black Sea coast. Throughout the work, the characteristics due to the biochemical compositions of the extracts from the brown alga C. barbata and from the jellyfish R. pulmo are highlighted as important, emphasizing the content of polysaccharides, proteins, and lipids. Total phenol content was analyzed for three extracts from natural products. The biochemical composition, antioxidant, antimicrobial, and in vitro wound-healing properties of the components and their composite (JPC-ALG) were assessed. The rheological behavior and optical microscopy studies of collagen hydrogels were prepared. The general mechanisms of wound healing with the involvement of polysaccharides and collagen peptides existing in all categories of extracts were highlighted. The study of the effects of JPC-ALG composites and individual extracts on fibroblast and keratocyte cell lines is also presented. Results demonstrated that the composite exhibited synergistic effects, enhancing fibroblast and keratinocyte migration and proliferation, key factors in wound closure. The findings support the potential application of this marine-derived bioactive composite as a promising biomaterial for wound-healing therapies.

Background/Objectives: Oral squamous cell carcinoma (OSCC) remains a major therapeutic challenge due to treatment-related toxicity and impaired oral tissue regeneration. This study aimed to develop and characterize a novel biocomposite based on bioactive compounds from Ganoderma lucidum incorporated into marine collagen derived from Rhizostoma pulmo and to evaluate its physicochemical properties, antioxidant and antimicrobial activities, and in vitro antitumor potential in the oral cavity. Methods: Hydroalcoholic extracts of G. lucidum and pepsin-soluble collagen peptides from R. pulmo jellyfish were prepared and combined to obtain two hydrogel biocomposites with different component ratios. Chemical and structural characterization was performed using HPLC-DAD, SDS-PAGE, FT-IR, circular dichroism, and spectrophotometric assays. Antioxidant activity was assessed by DPPH radical scavenging and reducing power assays, while antimicrobial activity was evaluated against oral pathogens using diffusion and MIC methods. In vitro biological activity was investigated using MTT viability and scratch migration assays on human OSCC cell lines (SCC-9 and HSC-3). Results: The biocomposites preserved the structural integrity of type I collagen and incorporated polysaccharides and polyphenols from G. lucidum. The combined formulations showed enhanced antioxidant and antimicrobial activities compared with collagen alone. In vitro assays demonstrated dose- and time-dependent reductions in OSCC cell viability and delayed cell migration, with effects comparable to those of G. lucidum extract. Conclusions: The G. lucidum–R. pulmo biocomposite exhibits favorable physicochemical properties and demonstrates antioxidant, antimicrobial, and in vitro antitumor activity. These findings support its potential as a multifunctional biomaterial for further investigation as an adjunct approach in oral cancer-related applications.

Wound healing is a complex, multi-stage process governed by tightly regulated molecular mechanisms. However, effective regenerative therapies remain with limitations. This study presents a novel marine-derived biocomposite, JPC-ALG-CT, designed to improve wound healing through synergistic bioactive mechanisms. The material incorporates collagen extracted from the jellyfish Rhizostoma pulmo, chitosan derived from the crab Pachygrapsus mormoratus, and polysaccharide-rich extracts from the green alga Cladophora vagabunda, all sourced from the Black Sea. The study is based on the biochemical analysis of these three marine-derived components, highlighting the collagen content of jellyfish, the polysaccharides present in algae, and the bioactive properties of chitosan. The biochemical and physico-chemical properties of each component were characterized, with particular emphasis on the structural features of jellyfish collagen and the functional bioactivity of chitosan and algal polysaccharides. The research findings are supported by the identification of the collagen type extracted from jellyfish, as well as by the characterization of chitosan and green algal extracts. The resulting composite demonstrated significant antioxidant and antimicrobial activities, indicating its potential to integrate key processes involved in wound repair, including inflammation control and microbial protection. In vitro studies using fibroblast and keratinocyte models showed that the JPC-CT-ALG biocomposite supported cell viability at lower tested concentrations and promoted scratch closure in cell monolayers, suggesting preliminary wound-relevant biological activity. These findings suggest that the combined marine-derived components interact to enchance wound healing at the cellular level. This work evidenced the potential of marine biomaterials as sources for next-generation regenerative therapies and supports further investigation into their molecular mechanisms and in vivo applications for improved wound care.

The Black Sea offers underutilized marine bioresources with potential for the development of materials intended for wound healing and skin tissue repair. In this study, new composite biomaterials were created using collagen peptides extracted from the jellyfish Rhizostoma pulmo (JPC) and hydroalcoholic extracts from the seaweeds Ulva lactuca (ALGU) and Enteromorpha intestinalis (ALGE). The marine-derived ingredients and resulting biocomposites were physicochemically and rheologically characterized and evaluated for antioxidant, antimicrobial, and biological activities on cell cultures relevant to wound healing. Two hydrogel biocomposites, B1 (JPC–ALGU 20%) and B2 (JPC–ALGE 20%), exhibited antioxidant activity of 65.2% and 63.5%, respectively, in the DPPH assay at a concentration of 400 mg/mL, and values of 1.26 μg/mg AAE and 1.15 μg/mg AAE, respectively, in the reducing power assay at a concentration of 800 µg/mL. Antibacterial activity was investigated against several Gram-positive and Gram-negative bacteria. The strongest effects were observed against Staphylococcus aureus. In vitro tests conducted on fibroblast and keratinocyte cultures revealed concentration-dependent cellular responses, while scratch assays confirmed the formulations’ potential to promote cell migration. These results support further investigation of jellyfish collagen–green algae biocomposites for wound-healing and skin tissue-repair applications, although additional dose optimization and in vivo validation are required.
