The NAC (NAM, ATAF1/2, and CUC2) gene family, one of the largest transcription factor families in plants, acts as positive or negative regulators in plant response and adaption to various environmental stresses, including cold stress. Multiple reports on the functional characterization of NAC genes in Arabidopsis thaliana and other plants are available. However, the function of the NAC genes in the typical woody mangrove (Kandelia obovata) remains poorly understood. Here, a comprehensive analysis of NAC genes in K. obovata was performed with a pluri-disciplinary approach including bioinformatic and molecular analyses. We retrieved a contracted NAC family with 68 genes from the K. obovata genome, which were unevenly distributed in the chromosomes and classified into ten classes. These KoNAC genes were differentially and preferentially expressed in different organs, among which, twelve up-regulated and one down-regulated KoNAC genes were identified. Several stress-related cis-regulatory elements, such as LTR (low-temperature response), STRE (stress response element), ABRE (abscisic acid response element), and WUN (wound-responsive element), were identified in the promoter regions of these 13 KoNAC genes. The expression patterns of five selected KoNAC genes (KoNAC6, KoNAC15, KoNAC20, KoNAC38, and KoNAC51) were confirmed by qRT-PCR under cold treatment. These results strongly implied the putative important roles of KoNAC genes in response to chilling and other stresses. Collectively, our findings provide valuable information for further investigations on the function of KoNAC genes.

Mangrove plant seedling cultivation is crucial for the protection, management, and restoration of the mangrove ecosystem. In this study, we focused on Kandelia obovata Sheue & al., a typical mangrove, and evaluated nursery cultivation with different combinations of three salinity levels (S1: 0 ppt, S2: 10 ppt, and S3: 20 ppt), three genealogies (EZD, JX, and YZ), and five growth media (M1: 100% loess, M2: 100% sandy, M3: 50% loess + 50% sandy, M4: 40% loess + 40% sandy + 20% peat, and M5: 40% loess + 40% sandy + 20% coir), by measuring the growth parameters such as mortality rate, seedling height, seedling diameter, and biomass partition. These growth indexes were significantly affected by salinity and medium, and genealogies also had significant effects on mortality rate and biomass accumulation. S2 or S3 both had lower mortality and higher growth indexes than S1. M1 was the medium that increased seedling height, diameter, and biomass the most and had the lowest death rate. EZD and JX were also at higher levels than YZ in these indicators, but the difference between them was not obvious. S3, M1, and EZD consistently performed well in fuzzy evaluation and quality assessment (Dickson quality index: 1.179, 1.478, and 1.089, respectively). Furthermore, combinations involving these treatments also produced highly favorable results. This indicates that the quality of seedlings produced under these conditions was high. These results furnish both a theoretical and practical foundation for advancing nursery cultivation techniques and germplasm breeding of K. obovata in mangroves.

Despite the accumulating evidence of the beneficial effects of diverse mixed species forests on ecosystem functioning and services, foresters in subtropical forest cultivation in China still prefer easily managed monocultures, which is also due to the complexity of mixed forests and the unknown underlying mechanisms related to relationships between biodiversity and forest growth. In a designed pot experiment, we selected two early-successional tree species (Pinus massoniana Lamb., Liquidambar formosana Hance.) and two late-successional tree species (Schima superba Champ., Elaeocarpus decipiens Hemsl.) and planted four saplings in one pot with regard to tree species diversity (monoculture, two species and four species mixtures), each combination replicated four times. In this three-year duration experiment, the effect of tree species diversity, tree identity, and functional traits on sapling growth (tree height, ground diameter, crown projection area), were analyzed. The results showed that the increments of ground diameter and crown projection area increased with tree species richness, whereas the mean tree height increment showed the opposite effect. This growth variation was species specific and related to functional traits (early or late succession), as the increments of the early successional tree species (P. massoniana Lamb. and L. formosana Hance.) had a positive correlation with tree species richness, while the late successional tree species (E. decipiens Hemsl. and S. superba Champ.) showed negative effects. In addition, our study provided evidence for the allometric differences between mixtures and monocultures, which have an important reference value on mixed-species forests.

Increasing studies have shown the importance of intraspecific trait variation (ITV) on ecological processes. However, the patterns and sources of ITV are still unclear, especially in the propagules of coastal vegetation. Here, we measured six hypocotyl traits for 66 genealogies of Kandelia obovata from 26 sites and analyzed how ITV in these traits was distributed across geography and genealogy through variance partitioning. We further constructed mixed models and structural equation models to disentangle the effects of climatic, oceanic, and maternal factors on ITV. Results showed that size‐related traits decreased along increasing latitudinal gradients, which was mainly driven by positive regulation of temperature on these traits. By contrast, ITV of shape trait was unstructured along latitudinal gradients and did not show any dependence among environmental variables. These findings indicate that propagule size mainly varied between populations, whereas propagule shape mainly varied between individuals. Our study may provide useful insights into the ITV in propagule from different functional dimensions and on a broad scale, which may facilitate mangrove protection in light of ITV.

Salt stress is common but detrimental to plant growth, even in mangroves that live in saline areas. Boron (B) is an essential micronutrient that performs an important role in many functions in plants; however, its protective role under salt stress is poorly understood, especially in long-lived woody plants. In this study, we conducted an indoor experiment under simulated tidal conditions with four treatments (10‰ salinity, 40‰ salinity, 40‰ salinity + 100 μM B, and 40‰ salinity + 500 μM B) and three mangrove species (Avicennia marina, Aegiceras corniculatum, and Bruguiera gymnorrhiza) to investigate the effects of exogenous B on salt tolerance in plant growth, morphology, physiology, and leaf anatomy. The results showed that exogenous low-concentration B treatment (100 μM B) improved the performance of mangrove species under high salinity stress, especially in terms of physiology and leaf anatomy, while high-concentration B treatment (500 μM B) had adverse effects. Additionally, we found that the response to exogenous B varied among species in physiology and leaf anatomy, such as proline, malondialdehyde, activity of antioxidant enzymes, palisade tissue, and spongy tissue, which may be related to the salt tolerance of different species. This study may provide useful insights into the alleviation of salt stress by B in mangrove growth and development, which may facilitate mangrove cultivation and afforestation in a saline environment.
