The search for new drugs to treat ulcerative colitis (UC) is ongoing, with the use of herbal extracts emerging as a current research focus in this field. This study primarily intended to investigate the antioxidant and anti‐inflammatory properties of the ethanolic extract of Thymus longicaulis subsp. chaubardii (TLC) in vitro. Secondly, we aimed to perform an acetic acid (AA)–induced rat UC model and assess the effects of TLC extract (200 mg/kg/orally/day, during three days) using ELISA test in terms of IL‐1 β , TNF‐ α , IL‐17, IL‐10, Na + /K + ‐ATPase, TLR‐9, MMP‐3 level, caspase‐3 and caspase‐9, luminol, lucigenin, and SOD. In addition, we accomplished macroscopic and histological evaluations. Our phytochemical investigation revealed TLC contained secondary metabolites including total phenolic, total flavonoid, and total triterpenes in quite concentrated amounts. The extract demonstrated anti‐2,2‐diphenyl‐1‐picrylhydrazyl (DPPH) radical scavenging activity that was 2.3 times more potent than ascorbic acid, anti‐2,2‐azino‐bis (3‐ethylbenzothiazoline‐6‐sulfonic acid) (ABTS) free radical scavenging activity that was 3.28 times less potent than Trolox, and antilipoxygenase Type V (LOX‐5) activity that was 1.31 times more potent than indomethacin in vitro. Due to colitis, increased luminol and lucigenin CL, TNF‐ α , IL‐1 β , IL‐17, TLR‐9, caspase‐3, caspase‐9, MMP‐3, and macroscopic and microscopic scores and decreased IL‐10, Na+/K + ‐ATPase levels in colon ameliorated with TLC and sulfasalazine (as reference drug) treatments. As a result, it can be said that TLC improves UC in rats, mainly on account of its free radical scavenging activity, anti‐inflammatory, and antiapoptotic effects.

Background/Objectives: Thymus longicaulis subsp. chaubardii (TL) (Rchb.f.) Jalas is widely used in traditional Turkish medicine for respiratory, digestive and uro-genital disorders. The aim of this study was to determine its phytochemical profile and to evaluate its cytotoxic, genotoxic and acute oral toxicity effects. Methods: The phenolic composition of the methanolic extract was determined by HPLC-DAD. Cytotoxicity and genotoxicity were evaluated in NIH3T3 cells using MTT, comet and micronucleus assays. Acute toxicity was evaluated in rats at doses of 300 and 2000 mg/kg body weight according to the OECD Guideline 420. Results: Rosmarinic acid (87.37 ± 5.39 µg/mg) was the major phenolic compound. TL extract showed >90% cell viability at 50–200 µg/mL, indicating no cytotoxicity. Comet assay revealed a slight increase in DNA damage at 100–200 µg/mL (p < 0.001), though significantly lower than the H2O2 group (p < 0.001). No significant (p > 0.05) effect was observed in the micronucleus assay between the treated groups. In rats, TL extract caused no mortality or behavioral changes over 14 days. No significant differences were observed in body or organ weights. Hematologically, platelet count increased (p < 0.001) and eosinophils decreased (p < 0.01 and p < 0.001). Biochemical tests showed lower ALT and AST levels (p < 0.01 and p < 0.05, respectively) and significantly decreased triglycerides in the high-dose group (p < 0.001). Histopathological examination showed no organ damage. Conclusions: The results of this study indicate that TL methanol extract is non-toxic up to 2000 mg/kg and exhibits no significant cytotoxic or genotoxic effects. These findings support its safe use and traditional medicinal value.

This comprehensive study delves into the profound impact of low frequency whole-body vibrations (WBV) on health, focusing on the utilization of rat models for understanding this complex phenomenon. It highlights the adverse effects of low frequency vehicle vibrations on human health, encompassing musculoskeletal discomfort, fatigue, concentration deficits, potential gastrointestinal issues, hearing impairment, and psychological stress. WBV's influence extends to physiological and cognitive consequences, affecting multiple systems. Prolonged WBV exposure, particularly in the lumbar region, is associated with spinal disorders. To navigate ethical challenges in studying WBV in humans, rat models are crucial tools. These models, with customized parameters, offer insights into various health aspects, including bone density, muscle strength, hormonal responses, cardiovascular parameters, and more. Advantages and disadvantages of using low-frequency vibrations in rat models are discussed. While demonstrating WBV's potential effect in research, further exploration is essential to optimize parameters and applications, always prioritizing ethical considerations and regulations. The article concludes by proposing measures to mitigate vehicle vibration impacts on drivers, emphasizing the collaboration of manufacturers, drivers, and regulators for safer and healthier driving experiences.
