Acute liver injury occur after intraperitoneal administration of lipopolysaccharide (LPS). Oxidative stress and release of pro-inflammatory cytokines are both implicated in the pathogenesis of LPS-induced acute liver injury. This study investigated the ameliorative effect of fermented rooibos (Aspalathus linearis) extract on LPS-induced acute liver injury. Major phenolic compounds in the fermented rooibos extract by HPLC-DAD, as well as the in vitro antioxidant capacity were quantified before the start of the experiment. Male Wistar rats were randomized into 4 groups (n = 10 per group) and given either water or fermented rooibos extract for 4 weeks before LPS injection. Hepatic function markers, including aminotransferases and lactate dehydrogenase, lipid peroxidation markers, antioxidant enzymes, glutathione redox status, as well as cytokine levels were monitored in the rats. Injection of LPS significantly increased serum levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST) and lactate dehydrogenase (LDH). Oxidative stress, evidenced by increased thiobarbituric acid reactive substances (TBARS) measured as malondialdehyde (MDA) in plasma and liver, and decreased glutathione redox status (GSH: GSSG ratio) in whole blood and liver was induced in LPS-challenged rats. Furthermore, hepatic levels of pro-inflammatory response markers TNF-α, IL-1β and IL-6 were increased significantly. Pre-feeding the fermented rooibos extract for 4 weeks decreased LPS-induced elevated levels of serum AST and LDH (significantly, p < 0.05) as well as ALT marginally. Consuming rooibos caused an attenuation of the observed increase in plasma and hepatic MDA, decrease in whole blood and liver GSH:GSSG ratio, as well as the changes noted in various antioxidant enzymes. The elevation in TNF-α and IL-6 was significantly suppressed, indicating an inhibition of the induced inflammatory response by rooibos. Overall, our data showed that aqueous rooibos extract attenuated LPS-induced liver injury possibly by modulating oxidative stress and suppressing pro-inflammatory cytokines formation.

The effect of different sources of Zinc (Zn) in the diets of laying birds was investigated in a 10 weeks trial. Diets were formulated to include Zinc Oxide (ZnO), Zinc Sulphate (ZnSO4), Zinc Carbonate (ZnCO3) and Zinc Proteinate (ZnP) in which Zn in each diet was supplied at 140 mgKg-1 diet. Responses measured included performance, some serum biochemistry, Zn residues in tibia bone, liver, excreta and egg shell. Significant (p<0.05) differences were recorded among the treatment means in final body weight, feed intake, egg production and feed conversion ratio. Birds fed control diet significantly (p<0.05) consumed more feed than the birds on ZnP and other inorganic Zn sources. Birds on ZnP supplemented diet recorded significantly (p0.05) different due to Zn sources except HU values. Serum glucose, SGPT and creatinine concentrations did not vary (p>0.05) due to Zn sources while serum protein, uric acid, SGOT and serum Zn concentrations were consistently lowered (p<0.05) in the control group. Birds on ZnP supplemented diet showed marked significant (p<0.05) difference in the value of Zn residue in tibia bone, liver and excreta of the chickens. The values of the stress indicators were consistently more pronounced in the control group than birds on Zn supplemented diets. For better laying performance, higher Zn retention and alleviation of stress, 140 ppm of Zn in bioplex form (Zn proteinate) was recommended for laying chickens in the tropics.

Diabetes mellitus is a metabolic disease that can lead to high morbidity, mortality and long-term complications. Available treatment strategies, which are mainly based on treating hyperglycemia, with insulin and other pharmacological agents are not completely efficient and can even lead to development of unwanted side effects. Scientific evidence suggests that bioactive compounds from teas and other plant-based foods, which are known source of natural antioxidants, could be an attractive strategy to preferentially treat and manage type 2 diabetes mellitus (T2DM) and thus, have significant therapeutic implications. In this review, we attempt an in-depth analysis and discussion of the current progress in our understanding of the antidiabetic potential of two commercialized South Africa herbal tisanes—Rooibos and Honeybush and their polyphenols.

This study was designed to investigate the in vivo effects of dietary copper supplementation on the oxidative and antioxidant systems in the erythrocyte and liver of broiler chickens. Birds were exposed to commercially prepared diet (control group, n= 20) or commercial diet supplemented with 250 mg CuSO4/kg diet (experimental group, n= 20) for 6 weeks. The copper burdens, oxidative lipid peroxidation (LPO), glutathione (GSH) concentration, activity of antioxidant enzymes, superoxide dismutase (SOD) and catalase (CAT) were evaluated in the erythrocyte and hepatic tissues using well established techniques. Oral exposure to copper for a period of 6 weeks significantly (p<0.05) increased the copper burden and lipid peroxidation respectively in the blood and liver of treated birds compared with control. Concomitantly, there was a significant reduction (p<0.05) in the activities of SOD, CAT, and the concentration of GSH in the exposed birds compared with control. It is concluded from this study that dietary copper supplementation (250 mg CusO4/kg diet) induced oxidative stress in the erythrocyte and liver of broiler chickens.

Red palm oil (RPO) and rooibos have been shown to exhibit cardioprotective properties. RPO is rich in essential fatty acids and fat soluble antioxidants while rooibos contains polyphenolic compounds with a unique composition of flavonoids. They exert their biological effects in different cellular compartments. Therefore the combination of these two natural food compounds has the potential to enhance the spectrum of available dietary antioxidants in different cellular compartments, which could result in an enhanced protection against certain pathological conditions such as inflammation. Male Wistar rats weighing 150-200 g were supplemented with RPO, rooibos or their combination for 28 days. The Langendorff system and the lipoposaccharide (LPS)-induced inflammatory model were used to establish if RPO and rooibos, when supplemented alone or in combination, will reverse the negative effects of LPS on cardiac function at baseline. The effect of dietary intervention was also investigated on modulation of pro-inflammatory and anti-inflammatory cytokines in plasma and myocardial tissue. The LPS resulted in induction of systemic inflammation as evidenced by increased levels of IL-1β in plasma of LPS-treated rats compared to their non-treated control counterparts. Dietary supplementation and LPS treatment did not have an effect on baseline cardiac functional parameters. However, the elevation of IL-1β levels in plasma of LPS-induced rats consuming either RPO or rooibos alone were paralleled with increased levels of the anti-inflammatory cytokine, IL-10. The combination of rooibos and RPO was associated with enhanced endogenous production of myocardial IL-10 in LPS-induced rats. The results of this study indicate that RPO and rooibos when supplemented individually showed anti-inflammatory effect at systemic level while their combination exhibited an enhanced anti-inflammatory effect in the myocardial tissue. Therefore, the findings in the current study argue that the combination of these two natural food substances could be beneficial in clinically relevant conditions where inflammation plays a role.

The possible protective effects of an aqueous rooibos extract (Aspalathus linearis), red palm oil (RPO) (Elaeis guineensis), or their combination ontert-butyl-hydroperoxide-(t-BHP-)induced oxidative hepatotoxicity in Wistar rats were investigated.tert-butyl hydroperoxide caused a significant (P<0.05) elevation in conjugated dienes (CD) and malondialdehyde (MDA) levels, significantly (P<0.05) decreased reduced glutathione (GSH) and GSH : GSSG ratio, and induced varying changes in activities of catalase, superoxide dismutase, glutathione peroxidase, and glutathione reductase in the blood and liver. This apparent oxidative injury was associated with histopathological changes in liver architecture and elevated levels of serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), and lactate dehydrogenase (LDH). Supplementation with rooibos, RPO, or their combination significantly (P<0.05) decreased CD and MDA levels in the liver and reduced serum level of ALT, AST, and LDH. Likewise, changes observed in the activities of antioxidant enzymes and impairment in redox status in the erythrocytes and liver were reversed. The observed protective effects when rooibos and RPO were supplemented concomitantly were neither additive nor synergistic. Our results suggested that rooibos and RPO, either supplemented alone or combined, are capable of alleviatingt-BHP-induced oxidative hepatotoxicity, and the mechanism of this protection may involve inhibition of lipid peroxidation and modulation of antioxidants enzymes and glutathione status.
