Modern studies have shown that adaptogens can non-specifically enhance the resistance of human body under a wide range of external stress conditions with a multi-targeted and multi-channel network-like manner, especially by affect the immune-neuro-endocrine system and the hypothalamic–pituitary–adrenal axis. This review article draws the attention to the relationships of adaptogens, tonics from traditional Chinese medicine (TCM) and ginseng-like herbs worldwide, which all have similar plant sources and clinical applications. To clarify the sources and pharmacological mechanisms of these plant-originated adaptogens, which will provide useful information for the utilization of adaptogens to improve the human health. Meanwhile, the TCMs and the world-wide ginseng-like herbs from each region’s ethnopharmacology will be beneficial modernization and globalization.

The CRISPR/Cas system is now being used extensively in nucleic acid detection applications, particularly after the trans-cleavage activity of several Cas effectors was found. A CRISPR/Cas system combined with multiple signal-readout techniques has been developed for various molecular diagnostics applications. Fluorescence is now a widely utilized dominant read-out technique in CRISPR biosensors. An in-depth understanding of various fluorescence readout types and variables affecting the fluorescence signals can facilitate better experimental designs to effectively improve the analytical performance. There are the following two commonly used types of CRISPR/Cas detection modes: the first is based on binding activity, such as Cas9 and dCas9; the second is based on cleavage activity, such as Cas12a, Cas12b, Cas13, and Cas14. In this review, fluorescence signal-readout strategies from the last 5 years based on the binding activity and cleavage activity of the CRISPR/Cas system with fundamentals and examples are fully discussed. A detailed comparison of the available fluorescent reporter sequences and design principles is summarized. Current challenges and further applications of CRISPR-based detection methods will be discussed according to the most recent developments.

Our facial skin hosts millions of microorganisms, primarily bacteria, crucial for skin health by maintaining the physical barrier, modulating immune response, and metabolizing bioactive materials. Aging significantly influences the composition and function of the facial microbiome, impacting skin immunity, hydration, and inflammation, highlighting potential avenues for interventions targeting aging-related facial microbes amidst changes in skin physiological properties. We conducted a multi-center and deep sequencing survey to investigate the intricate interplay of aging, skin physio-optical conditions, and facial microbiome. Leveraging a newly-generated dataset of 2737 species-level metagenome-assembled genomes (MAGs), our integrative analysis highlighted aging as the primary driver, influencing both facial microbiome composition and key skin characteristics, including moisture, sebum production, gloss, pH, elasticity, and sensitivity. Further mediation analysis revealed that skin characteristics significantly impacted the microbiome, mostly as a mediator of aging. Utilizing this dataset, we uncovered two consistent cutotypes across sampling cities and identified aging-related microbial MAGs. Additionally, a Facial Aging Index (FAI) was formulated based on the microbiome, uncovering the cutotype-dependent effects of unhealthy lifestyles on skin aging. Finally, we distinguished aging related microbial pathways influenced by lifestyles with cutotype-dependent effect. Together, our findings emphasize aging’s central role in facial microbiome dynamics, and support personalized skin microbiome interventions by targeting lifestyle, skin properties, and aging-related microbial factors.

Crosslinked liquid crystalline polymers (CLCPs) containing azobenzene (AZO-CLCPs) are a type of promising material due to their significance in the design of light-driven smart actuators. Developing AZO-CLCP composites by incorporating AZO-CLCPs with other materials is an effective way of enhancing their practicability. Herein, we report an AZO-CLCP/CNT nanocomposite prepared by the in situ polymerization of diacrylates containing azobenzene chromophores on carbon nanotube (CNT) sheets. The liquid crystal phase structure of CLCP matrix was evidenced by the two-dimensional X-ray scattering. The prepared pure AZO-CLCP films and AZO-CLCP/CNT nanocomposite films demonstrated strong reversible photo-triggered deformation under the irradiation of UV light at 366 nm of wavelength, as a result of photo-induced isomerization of azobenzene moieties in the polymer network. But compared to pure AZO-CLCP films, the AZO-CLCP/CNT nanocomposite films could much more rapidly return to their initial shapes after the UV light irradiation was removed due to the elasticity effect of CNT sheets. The deformation behavior of AZO-CLCP/CNT nanocomposite films under the light irradiation was also different from that of the pure AZO-CLCP films due to the interfacial interaction between a polymer network and CNT sheet. Furthermore, incorporation of a CNT sheet remarkably increased the mechanical strength and robustness of the material. We also used this AZO-CLCP/CNT nanocomposite as a microvalve membrane actuator, which can be controlled by light, for a conceptual device of a microfluidic system. The results showed that this AZO-CLCP/CNT nanocomposite may have great potential in smart actuator applications for biological engineering, medical treatment, environment detection and microelectromechanical systems (MEMS), etc.
