CRISPR-Cas9 has emerged as a revolutionary tool that enables precise and efficient modifications of the genetic material. This review provides a comprehensive overview of CRISPR-Cas9 technology and its applications in genome editing. We begin by describing the fundamental principles of CRISPR-Cas9 technology, explaining how the system utilizes a single guide RNA (sgRNA) to direct the Cas9 nuclease to specific DNA sequences in the genome, resulting in targeted double-stranded breaks. In this review, we provide in-depth explorations of CRISPR-Cas9 technology and its applications in agriculture, medicine, environmental sciences, fisheries, nanotechnology, bioinformatics, and biotechnology. We also highlight its potential, ongoing research, and the ethical considerations and controversies surrounding its use. This review might contribute to the understanding of CRISPR-Cas9 technology and its implications in various fields, paving the way for future developments and responsible applications of this transformative technology.

Context: The COVID-19 outbreak is caused by the transmission and infection of SARS-CoV-2 at the end of 2019. It has led many countries to implement lockdown policies to prevent the viral spreading. Problems arise in a COVID-19 patient because of viral infection that leads to a systemic response in the immune system, specifically due to cytokine storm. Moreover, the antiviral drugs that have not been found. Indonesia had a variety of traditional medicines, such as is ‘jamu’. It consists of a mixture of natural ingredients such as Moringa oleifera Lam. and Curcuma longa L. Aims: To identify the activity of dual inhibitors as antiviral and anti-inflammatory agents from herbal combination compounds. Methods: Sample was collected from PubChem (NCBI, USA) and Protein Data Bank (PDB), then drug-likeness analysis using Lipinski rule of five in SCFBIO web server and bioactive probability analysis of bioactive compounds were conducted by PASS web server. Furthermore, the blind docking method was performed using PyRx 0.8 software to determine the binding activity and molecular interaction by PoseView web server and PyMol software v2.4.1 (Schrödinger, Inc, USA). Results: Cryptochlorogenic acid and curcumin have been computationally proven as dual inhibitors for antivirals by inhibiting Mpro SARS-CoV-2 and as anti-inflammatory through inhibition of NFKB1 activity. However, the results are merely computational so that it must be validated through a wet lab research. Conclusions: The combination of Moringa oleifera Lam. and Curcuma longa L. is predicted to have antiviral and anti-inflammatory activity through dual inhibitor mechanism played by cryptochlorogenic acid and curcumin.

Crimean-Congo hemorrhagic fever virus (CCHFV) is a tick-borne virus, and zoonosis, and affects large regions of Asia, Southwestern and Southeastern Europe, and Africa. CCHFV can produce symptoms, including no specific clinical symptoms, mild to severe clinical symptoms, or deadly infections. Virus isolation attempts, antigen-capture enzyme-linked immunosorbent assay (ELISA), and reverse transcription polymerase chain reaction (RT-PCR) are all possible diagnostic tests for CCHFV. Furthermore, an efficient, quick, and cheap technology, including biosensors, must be designed and developed to detect CCHFV. The goal of this article is to offer an overview of modern laboratory tests available as well as other innovative detection methods such as biosensors for CCHFV, as well as the benefits and limits of the assays. Furthermore, confirmed cases of CCHF are managed with symptomatic assistance and general supportive care. This study examined the various treatment modalities, as well as their respective limitations and developments, including immunotherapy and antivirals. Recent biotechnology advancements and the availability of suitable animal models have accelerated the development of CCHF vaccines by a substantial margin. We examined a range of potential vaccines for CCHF in this research, comprising nucleic acid, viral particles, inactivated, and multi-epitope vaccines, as well as the present obstacles and developments in this field. Thus, the purpose of this review is to present a comprehensive summary of the endeavors dedicated to advancing various diagnostic, therapeutic, and preventive strategies for CCHF infection in anticipation of forthcoming hazards.Graphical Abstract

Silver nanoparticles have been successfully synthesized using a chemical reduction method at room temperature using glucose as a reducing agent, silver nitrate as a source of nitrate, polyvinyl alcohol (PVA) as a stabilizing agent. The purpose of this research was to determine the effect of the mole ratio of reducing agent Glu / Ag and percentage (%) PVA (b/v) in the silver nanoparticle synthesis of the resulted product. Product characterization was analyzed by UV-Vis Spectrophotometers, XRD, and TEM. The Analysis of the UV-Vis spectra showed that the most stable particles were silver nanoparticles used PVA 3% and Glu / Ag. 1: 8 on λmax 415 to 417 nm. TEM’s characterization showed that silver nanoparticles were spherical with a dominant size range of 12,28 nm – 38,45 nm nm. The newly synthesized silver nanoparticles may have a potential for antibacterial applications.

Rendangis a kind of food that has become one of cultural heritages of Minangkabau tribe. It is not only a local food that has been crowned the best food in the world by CNN, but it also serves as the cultural identity. Most of people think that rendang is only made from beef of buffalo meat. In fact, rendang has many varieties that relate to the types of the area and the local natural resources where it is cooked. West Sumatra consists of mountainous and coastal areas. One of the popular rendang varieties in coastal areas isRendang Lokan—it is not made from beef but oyster (lokan). Rendang Lokan is a kind of food for people who live in this area. They use resources of the sea for their foods. Rendang, for Minangkabau tribe, is not only considered as a traditional food but also as a symbol of cultural identity from a certain area. This article is aimed at discussing the history of using Oyster as the main ingredient for rendang as one of the rendang varieties coming from the coastal areas in West Sumatra. The results show that rendang lokan is a representation of the characteristics of Minangkabau people who have the ability to adapt themselves to the local environmental conditions in maintaining their cultural identity. Rendang lokan was initially created as the adjustment of the Minangkabau people who live in the coastal areas to the local natural resources and their economic conditions.
