Transcriptional regulators are crucial in adipocyte differentiation. We now show that the homeodomain-containing transcription factor Prep1 is a repressor of adipogenic differentiation since its down-regulation (DR) in both ex vivo bone marrow-derived mesenchymal stromal cells (MSC) and in vitro 3T3-L1 preadipocytes significantly increases their adipogenic differentiation ability. Prep1 acts at a stage preceding the activation of the differentiation machinery because its DR makes cells more prone to adipogenic differentiation even in the absence of the adipogenic inducers. Prep1 DR expands the DNA binding landscape of C/EBPβ (CCAAT enhancer binding protein β) without affecting its expression or activation. The data indicate that Prep1 normally acts by restricting DNA binding of transcription factors to adipogenic enhancers, in particular C/EBPβ.

Lung cancer is the leading cause of cancer deaths. Lethal pulmonary adenocarcinomas (ADC) present with frequent mutations in the EGFR. Genetically engineered murine models of lung cancer expedited comprehension of the molecular mechanisms driving tumorigenesis and drug response. Here, we systematically analyzed the evolution of tumor heterogeneity in the context of dynamic interactions occurring with the intermingled tumor microenvironment (TME) by high-resolution transcriptomics. Our effort identified vulnerable tumor-specific epithelial cells, as well as their cross-talk with niche components (endothelial cells, fibroblasts, and tumor-infiltrating immune cells), whose symbiotic interface shapes tumor aggressiveness and is almost completely abolished by treatment with Unesbulin, a tubulin binding agent that reduces B cell–specific Moloney murine leukemia virus integration site 1 (BMI-1) activity. Simultaneous magnetic resonance imaging (MRI) analysis demonstrated decreased tumor growth, setting the stage for future investigations into the potential of novel therapeutic strategies for EGFR-mutant ADCs. Significance: Targeting the TME is an attractive strategy for treatment of solid tumors. Here we revealed how EGFR-mutant landscapes are affected at the single-cell resolution level during Unesbulin treatment. This novel drug, by targeting cancer cells and their interactions with crucial TME components, could be envisioned for future therapeutic advancements.

Osimertinib (Osi) resistance remains a significant challenge in EGFR mutant non-small-cell lung cancer (NSCLC). This study investigates the metabolic reprogramming associated with Osi resistance, identifying key metabolic vulnerabilities that may be targeted for therapeutic intervention. Employing the EGFR-mutant H1975 parental (Par) cell line and its Osi-resistant (OsiR) counterpart, we integrated transcriptomics, metabolomics, nuclear and mitochondrial genomics, functional assays and bioanalytical techniques, as well as advanced 3D imaging to comprehensively define the resistant phenotype. We found that OsiR cells exhibit mitochondrial dysfunction, including impaired oxidative phosphorylation (OXPHOS), mitochondrial DNA mutations, and altered mitochondrial gene expression. To describe this systems-level characterization, we introduce the concept of mitochondromics , a comprehensive profiling of mitochondrial genomic, transcriptomic, structural, and functional changes contributing to therapeutic resistance. Metabolomic profiling revealed a significant accumulation of glycolytic intermediates (lactate, pyruvate, acetate, and acetaldehyde) in the extracellular medium, indicating a shift toward glycolysis and activation of alternative metabolic pathways, including the Warburg effect. Notably, we identified the pyruvate-acetaldehyde-acetate (PAA) pathway as a functionally repurposed metabolic route that facilitates NADPH production, which is critical for antioxidant defense and anabolic processes in OsiR cells. Additionally, although the pentose phosphate pathway (PPP) is not the primary source of NADPH in OsiR cells, it plays a supporting role in biosynthesis, contributing to the production of amino acids, nucleotides, and vitamins. Altered expression of enzymes involved in glycolysis, the TCA cycle, and both oxidative and non-oxidative arms of the PPP further supports an adaptive metabolic network promoting cell growth and resistance to Osi. This study reveals a complex metabolic reprogramming in Osi-resistant EGFR-mutant NSCLC, where a newly identified role for the PAA pathway, alongside integrated mitochondromic alterations emerges as key driver of resistance. These insights uncover potential metabolic vulnerabilities of Osi-resistant tumors and provide a foundation for developing therapeutic strategies to counteract resistance and improve osimertinib efficacy. Targeting these metabolic pathways may offer promising avenues for overcoming resistance in clinical settings.

Molecular mechanisms governing cell fate decision events in bone marrow mesenchymal stromal cells (MSC) are still poorly understood. Herein, we investigated the homeobox gene Prep1 as a candidate regulatory molecule, by adopting Prep1 hypomorphic mice as a model to investigate the effects of Prep1 downregulation, using in vitro and in vivo assays, including the innovative single cell RNA sequencing technology. Taken together, our findings indicate that low levels of Prep1 are associated to enhanced adipogenesis and a concomitant reduced osteogenesis in the bone marrow, suggesting Prep1 as a potential regulator of the adipo-osteogenic differentiation of mesenchymal stromal cells. Furthermore, our data suggest that in vivo decreased Prep1 gene dosage favors a pro-adipogenic phenotype and induces a “browning” effect in all fat tissues.

New drugs and technologies are continuously developed to improve the efficacy and minimize the critical side effects of cancer treatments. The present investigation focuses on the development of a liposomal formulation for Idelalisib, a small-molecule kinase inhibitor approved for the treatment of lymphoid malignancies. Idelalisib is a potent and selective antitumor agent, but it is not indicated nor recommended for first-line treatment due to fatal and serious toxicities. Herein, liposomes are proposed as a delivery tool to improve the therapeutic profile of Idelalisib. Specifically, PEGylated liposomes were prepared, and their physicochemical and technological features were investigated. Light-scattering spectroscopy and cryo-transmission electron microscopy revealed nanosized unilamellar vesicles, which were proved to be stable in storage and in simulated biological fluids. The cytotoxicity of the liposome formulation was investigated in a human non-Hodgkin's lymphoma B cell line. Idelalisib was able to induce death of tumor cells if delivered by the nanocarrier system at increased efficacy. These findings suggest that combining Idelalisib and nanotechnologies may be a powerful strategy to increase the antitumor efficacy of the drug.

Small activating RNAs are short double-stranded RNAs designed to upregulate transcription of target genes. By this virtue, they can be used to restore expression of genes frequently silenced in cancer. AW1-51 (also referred to as CEBPA-51), the first small activating RNA therapeutic to enter clinical evaluation, has demonstrated biological activity and safety in Phase II trials for hepatocellular carcinoma, both as monotherapy and in combination with sorafenib, and in Phase 1a/1b in combination with pembrolizumab for patients with advanced solid tumors. It targets the master regulator CCAAT enhancer-binding protein alpha, abnormally silenced by DNA methylation in a wide range of hematological and non-hematological malignancies. However, the molecular events enabling this mechanism are only partially elucidated. In this study, we uncovered the molecular basis for AW1-51-induced transcriptional reactivation of CCAAT enhancer-binding protein alpha demonstrating that by directly promoting DNA demethylation of its promoter restores its expression, protein synthesis, and consequently cell differentiation. These findings unveil AW1-51 as a prototype for RNA-based precision medicine enabling conditional expression of CCAAT enhancer-binding protein alpha in diseases characterized by aberrant gene silencing and extending its potential therapeutic impact beyond cancer.
