Yellow rust is a worldwide disease that poses a serious threat to the safety of wheat production. Numerous studies on near-surface hyperspectral remote sensing at the leaf scale have achieved good results for disease monitoring. The next step is to monitor the disease at the field scale, which is of great significance for disease control. In our study, an unmanned aerial vehicle (UAV) equipped with a hyperspectral sensor was used to obtain hyperspectral images at the field scale. Vegetation indices (VIs) and texture features (TFs) extracted from the UAV-based hyperspectral images and their combination were used to establish partial least-squares regression (PLSR)-based disease monitoring models in different infection periods. In addition, we resampled the original images with 1.2 cm spatial resolution to images with different spatial resolutions (3 cm, 5 cm, 7 cm, 10 cm, 15 cm, and 20 cm) to evaluate the effect of spatial resolution on disease monitoring accuracy. The findings showed that the VI-based model had the highest monitoring accuracy (R2 = 0.75) in the mid-infection period. The TF-based model could be used to monitor yellow rust at the field scale and obtained the highest R2 in the mid- and late-infection periods (0.65 and 0.82, respectively). The VI-TF-based models had the highest accuracy in each infection period and outperformed the VI-based or TF-based models. The spatial resolution had a negligible influence on the VI-based monitoring accuracy, but significantly influenced the TF-based monitoring accuracy. Furthermore, the optimal spatial resolution for monitoring yellow rust using the VI-TF-based model in each infection period was 10 cm. The findings provide a reference for accurate disease monitoring using UAV hyperspectral images.

In total, 13 commercial wheat cultivars around China and four races of Puccinia striiformis f. sp. tritici (namely, CYR32, CYR33, G22-9, and G22-14) were employed for a test of relative parasitic fitness (RPF) using the drop method. The RPF values were measured, including the urediniospore germination rate, the latent period, the uredinial length, the uredinial density, the infection area, the sporulation intensity, the lesion expansion speed, and the sporulation period. The results indicated that the parameters of relative parasitic fitness of the four P. striiformis f. sp. tritici races on the 13 wheat cultivars were significantly different (P = 0.00) in sporulation intensity, lesion expansion speed, uredinial length, sporulation period, uredinial density, and latent period. The urediniospore germination rates of the four P. striiformis f. sp. tritici races for the test were significantly different (P = 0.00), whereas no correlation with the different cultivars was observed (P = 1.00). The infection areas of the tested races on the different cultivars were significantly different (P = 0.00) but there were no obvious manifestations among the various races (P = 0.20). Principal component analysis (PCA) showed that the sporulation intensity represented sporulation capacity and scalability, the latent period indicated infection ability, and the urediniospore germination rate represented urediniospore vigor, all of which fully contributed to the RPF in the interaction of the four races and 13 wheat cultivars, which was calculated by the following formula: RPF = (sporulation intensity × urediniospore germination rate)/latent period. The sporulation and infection of G22-9 on the 13 large-scale cultivated cultivars were the highest, and the RPF of G22-9 was higher than that of the predominant races, CYR32 and CYR33. This result suggested that G22-9 could become a new predominant race and potentially cause epidemics of wheat stripe rust in China. To prevent potential epidemics, susceptible wheat cultivars should be withdrawn from production and breeding programs should reduce the use of Yr10 and Yr26 and use other more effective resistance genes in combination with nonrace-specific resistance for developing wheat cultivars with durable resistance to stripe rust.

Wheat stripe (yellow) rust, caused by Puccinia striiformis f. sp. tritici (Pst), is a serious fungal disease worldwide, especially in the Huang-Huai-Hai region, a main wheat production area in China. Gene postulation, molecular testing, and pedigree analysis were conducted to determine the presence of stripe rust resistance genes to 15 Pst races in 66 selected commercial wheat cultivars released from 2000 to 2016. In addition, races CYR32, CYR33, and CYR34 were used to evaluate resistance to Pst at the adult-plant stage of wheat in the field. Four Yr genes (Yr9, Yr10, Yr26, and Yr32) were postulated in 24 wheat cultivars either singly or in combination. Thirty-six cultivars might contain unknown Yr genes, whereas no identified Yr gene was postulated in six cultivars. Yr9 was detected at a frequency of 28.8%, and no cultivars carried Yr5, Yr15, or Yr18. Ten cultivars (15.2%) exhibited adult-plant resistance in the field tests with three predominant races. Three cultivars (Langyan 43, Xinong 889, and Yunfeng 139) had all-stage resistance. These results are useful to growers selecting cultivars and to breeders aiming to use more resistance genes to develop new cultivars with effective resistance in order to reduce stripe rust damage.

Puccinia triticina (Pt), the causal agent of wheat leaf rust, is one of the most destructive fungal pathogens threatening global wheat cultivations. The rational utilization of leaf rust resistance (Lr) genes is still the most efficient method for the control of such diseases. The Lr47 gene introgressed from chromosome 7S of Aegilops speltoides still showed high resistance to the majority of Pt races collected in China. However, the Lr47 gene has not been cloned yet, and the regulatory network of the Lr47-mediated resistance has not been explored. In the present investigation, transcriptome analysis was applied on RNA samples from three different wheat lines (“Yecora Rojo”, “UC1037”, and “White Yecora”) carrying the Lr47 gene three days post-inoculation with the epidemic Pt race THTT. A comparison between Pt-inoculated and water-inoculated “Lr47-Yecora Rojo” lines revealed a total number of 863 upregulated (q-value < 0.05 and log2foldchange > 1) and 418 downregulated (q-value < 0.05 and log2foldchange < −1) genes. Specifically, differentially expressed genes (DEGs) located on chromosomes 7AS, 7BS, and 7DS were identified, ten of which encoded receptor-like kinases (RLKs). The expression patterns of these RLK genes were further determined by a time-scale qRT-PCR assay. Moreover, heatmaps for the expression profiles of pathogenesis-related (PR) genes and several transcription factor gene families were generated. Using a transcriptomic approach, we initially profiled the transcriptional changes associated with the Lr47-mediated resistance. The identified DEGs, particularly those genes encoding RLKs, might serve as valuable genetic resources for the improvement of wheat resistance to Pt.

Wheat yellow rust, caused by Puccinia striiformis f. sp. tritici (PST), is one of the most important wind-borne diseases in all wheat-growing regions, and its occurrence can lead to devastating yield losses in wheat. In China, the wheat fields in Gansu act as an inoculum reservoir during summer periods and provide PST spores for wheat in the fall. The wheat fields in Qinghai provide large amounts of oversummering inocula. The exchange and migration of spores between the two regions are essential to ensure the persistence of PST. To confirm this relationship, we studied the genetic diversity, seasonal population dynamics, role of recombination, and gene flow between PST populations in different oversummering areas of Gansu and Qinghai using molecular markers combined with a spatiotemporal sampling strategy. Shared genotypes provide molecular evidence of migration between the pathogen populations in the two regions. The distribution of genotypic frequencies indicates that the pathogen mainly flows from Qinghai to Gansu in the autumn, whereas the opposite direction of movement occurs in the spring. The inoculum source from spring wheat can be directly transmitted to autumn seedlings, not necessarily through volunteer wheat. Therefore, the bridging effect of spring wheat may play a more important role than the off-season pathogen surviving on volunteer wheat plants. Furthermore, linkage disequilibrium tests indicate that sexual recombination continues throughout the year in the Tianshui and Dingxi regions of Gansu.
