Consumers are becoming increasingly interested in food containing high concentration of polyunsaturated fatty acids (PUFA). PUFA are considered as functional ingredients to prevent cardiovascular disease. The present study aimed to evaluate the effects of Clostridium butyricum on antioxidant properties, meat quality and fatty acid composition of broilers. A total of 320 one-day-old Arbor Acres male chicks were randomly assigned to one of five treatments with eight replicates and fed a antibiotic-free basal corn-soybean meal diet (control) or the basal diet supplemented with either 2.5 × 108 (CB1), 5 × 108 (CB2) or 1 × 109 (CB3) cfu of C. butyricum/kg or150 mg of aureomycin/kg (antibiotic) for 42 days. The results showed that chicks fed diets supplemented with C. butyricum had higher (P < 0.05) superoxide dismutase activity and lower (P < 0.05) malondialdehyde concentration in liver compared with those in the control group. Broilers had lower (P < 0.05) cholesterol content of serum in either CB2 or CB3 treatment at day 21 and in the C. butyricum-supplemented groups at day 42 than those in the control group. Chicks fed CB3 diet had lower (P < 0.05) percentage of abdominal fat and higher (P < 0.05) breast muscle yield than those in the control and antibiotic groups. The supplementation of C. butyricum increased (P < 0.05) the concentrations of C20:1n-9, C20:2n-6, C20:3n-6, C20:3n-3, C20:4n-6, C20:5n-3, C22:6n-3 and total PUFA as well as ratio of PUFA to saturated fatty acids in breast muscle and the contents of C18:2 t-9, t-12, C20:3n-6, C20:3n-3 and C20:5n-3 in thigh muscle. Supplementation of C. butyricum promotes hepatic antioxidant status, decreases cholesterol content of serum and percentage of abdominal fat, and improves meat quality and fatty acid composition of broiler birds. The results from the present study indicate that the increased PUFA concentrations in meat of broilers fed C. butyricum might be attributable to enhanced antioxidant activity.

The degree of hydrolysis (DH) plays important roles in the characteristics of food proteins. Herein, in order to explore the effects of partial hydrolysis on the structural, functional and antioxidant characteristics of hydrolysates, oat protein isolate was partially hydrolyzed with alcalase at different DHs (2%, 4%, 6%, 8%, 12%, and 16%). Our results showed that hydrolysis could induce significant structural changes in oat protein, mainly reflecting at the amino acid pattern, molecular weight profile and protein conformation. Alcalase hydrolysis also resulted in hydrolysates with the emulsifying activity index of at least 19.83 m2 g−1, and the highest emulsion stability was observed in the hydrolysate with a DH of 6%, possibly due to its suitable molecular weight, exposed hydrophobic amino acid residues and high surface net charge. Besides, all hydrolysates exerted excellent DPPH radical scavenging activity with an IC50 value ranging from 19.23 to 30.32 μg mL−1, which was closely correlated with DH. The oat protein isolate with moderate alcalase hydrolysis (DH 6%) exhibited the strongest metal ion-chelating activity and possessed the maximum amount of hydrophilic amino acids. More importantly, the oat protein hydrolysate with a DH of 6% not only prolonged the induction period of sunflower oil, but also improved the stability of the sunflower oil-in-water emulsion, as evidenced by the reduced TBARS production and the homogeneous droplet size. Therefore, partial hydrolysis can be advantageous for improving the functional and antioxidant characteristics of oat protein isolate, particularly the hydrolysate with a DH of 6%.

Localized intestine inflammation could induce short-term increases in colonic oxygenation and leads to increases in the aerobic bacteria population and reduction in the anaerobic bacteria population by changing the intestinal environment. However, the mechanisms involved and the associated functions of intestinal anaerobes in gut health still remain unclear. Here, we found that early-life depletion of gut microbiota exacerbated later colitis, while mid-life microbiota depletion showed partially reduced colitis. Notably, we observed that early-life gut microbiota depletion confers susceptibility to ferroptosis in colitis. In contrast, restitution of early-life microbiota conferred protection against colitis and inhibited ferroptosis triggered by gut microbiota dysbiosis. Similarly, colonization with anaerobic microbiota from young mice suppressed colitis. These results may attribute to high abundance of plasmalogen-positive (plasmalogen synthase [PlsA/R]-positive) anaerobes and plasmalogens (one of the common ether lipids) in young mice but reduced abundance in the development of inflammatory bowel disease. Early-life anaerobic bacteria elimination also resulted in the aggravation of colitis, while this aggravation phenotype was reverted by plasmalogen administration. Interestingly, plasmalogens inhibited ferroptosis triggered by microbiota dysbiosis. We further find that the alkenyl-ether group of plasmalogens was critical to colitis prevention and ferroptosis inhibition. These data point to one of the mechanisms by which the gut microbiota controls susceptibility to colitis and ferroptosis early in life via microbial-derived ether lipids.

Xylan is a major component of plant cells and the most abundant hemicellulose. Xylanases degrade xylan into monomers by randomly cleaving β-1,4-glycosidic bonds in the xylan backbone, and have widespread potential applications in various industries. The purpose of our study was to clone and express the endoxylanase gene xynA of Thermobifida fusca YX in its native form and with a C-terminal histidine (His) tag in Pichia pastoris X-33. We analyzed and compared these two forms of the protein and examined their potential applications in various industries. The xynA gene from T. fusca YX was successfully cloned and expressed using P. pastoris X-33. We produced a recombinant native form of the protein (rXyn11A) and a C-terminal His-tagged form of the desired protein (rXyn11A-(His)6). The specific activities of rXyn11A and rXyn11A-(His)6 in culture supernatants approached 149.4 and 133.4 U/mg, respectively. These activities were approximately 4- and 3.5-fold higher than those for the non-recombinant wild-type Xyn11A (29.3 U/mg). Following purification, the specific activities of rXyn11A and rXyn11A-(His)6 were 557.35 and 515.84 U/mg, respectively. The specific activity of rXyn11A was 8% higher than that of rXyn11A-(His)6. Both recombinant xylanases were optimally active at 80°C and pH 8.0, and exhibited greater than 60% activity between pH 6–9 and 60–80°C. They exhibited similar pH stability, while rXyn11A exhibited better thermostability; N-glycosylation enhanced the thermostability of both recombinant xylanases. The products of beechwood xylan hydrolyzed by both xylanases included xylobiose, xylotriose, xylotetraose and xylopentaose. The C-terminal His tag had adverse effects when added to the Xyn11A protein. The thermostability of both recombinant xylanases was enhanced by N-glycosylation. Their stabilities at a high pH and temperature indicate their potential for application in various industries.

Aerated emulsions have widespread applications in the food industry. However, the poor stability of aerated emulsions remains a major challenge due to their inherent thermodynamic instability. Herein, a novel strategy to improve the foam stability of aerated emulsions using natural waxes was developed. Natural waxes including beeswax (BW), candelilla wax (CLW), carnauba wax (CW) and rice bran wax (RW), the bio-based and sustainable materials, displayed a high efficiency in improving the interfacial behavior, structural properties and foam stabilization of aerated emulsions. Compared with emulsions containing pure palm kernel stearin (PKS), the presence of waxes enhances the elastic modulus (E′) of air–liquid interfacial films and the viscosity (η) of the continuous phase. Interestingly, cryo-scanning electron microscopy (cryo-SEM) images revealed that the fat droplet shape was controlled by waxes. These were related to that waxes were conducive to the formation of larger and rigid fat crystals in fat droplets and facilitated effective piercing of the interface, promoting the fat coalescence. Waxes promoted the formation of partial coalescence in the plateau borders and the interface of the aerated emulsions, thus improving the foam stability. Furthermore, whipping experiments showed that the addition of 1.0–2.0 wt% waxes resulted in a shorter optimum whipping time and a higher overrun of aerated emulsions. The microstructure images showed that aerated emulsions with waxes displayed more numerous and uniform air bubbles. Finally, rheology experiments strongly supported that waxes strengthened the interface and network structure of aerated emulsions. Our observations revealed the mechanism of interaction between the natural wax-tuned interfacial behavior and the structure of aerated emulsions.

Two commercial plant proteases namely ficin and bromelain, were acquired to hydrolyze mung bean protein over 300 min hydrolysis, and the physicochemical and antioxidative properties of the obtained hydrolysates were investigated. Bromelain-treated mung bean protein hydrolysates presented a higher degree of hydrolysis in comparison with ficin-treated hydrolysates, further modifying their physicochemical and emulsifying properties. All mung bean protein hydrolysates exhibited 50% scavenging of DPPH radical (IC50) in the concentration range from 8.67 to 16.22 μg mL−1. Our results also showed that strong metal ion-chelating activity was found in the ficin- (higher activity) and bromelain-treated protein hydrolysates. In addition, oxidative stability of linoleic acid was significantly enhanced by two selected protein hydrolysates, particularly the bromelain-treated hydrolysate with the highest inhibition effect of linoleic acid oxidation (94.55 ± 0.10%). Interestingly, both of these two hydrolysates could effectively retard lipid oxidation of sunflower oil and sunflower oil-in-water emulsion, while the ficin-treated hydrolysate showed slightly better performance. Therefore, mung bean protein hydrolysates showed potential to inhibit lipid oxidation, which could be advantageous in the food industry for producing fortified food.
