Object: Glial scar and cystic formation greatly contribute to the inhibition of axonal regeneration after spinal cord injury (SCI). Attempts to promote axonal regeneration are extremely challenging in this type of hostile environment. The objective of this study was to examine the surgical methods that may be used to assess the factors that influence the level of scar and cystic formation in SCI. Methods: In the first part of this study, a complete transection was performed at vertebral level T9–10 in adult female Sprague-Dawley rats. The dura mater was either left open (control group) or was closed using sutures or hyaluronic acid. In the second part of the study, complete or subpial transection was performed, with the same dural closure technique applied to both groups. Histological analysis of longitudinal sections of the spinal cord was performed, and the percentage of scar and cyst formation was determined. Results: Dural closure using sutures resulted in significantly less glial scar formation (p = 0.0248), while incorporation of the subpial transection surgical technique was then shown to significantly decrease cyst formation (p < 0.0001). Conclusions: In this study, the authors demonstrated the importance of the vasculature in cyst formation after spinal cord trauma and confirmed the importance of dural closure in reducing glial scar formation.

In this study we investigated the effect of neurotrophin-3 (NT-3) and knockdown of NG2, one of the main inhibitory chondroitin sulfate proteoglycans (CSPG), in the glial scar following spinal cord injury (SCI). Short hairpin (sh) RNA were designed to target NG2 and were cloned into a lentiviral vector (LV). A LV was also constructed containing NT-3. LV expressing NT-3, shRNA to NG2 or combinations of both vectors were injected directly into contused rat spinal cords 1 week post-injury. Six weeks post-injection of LV, spinal cords were examined by histology for changes in scar size and by immunohistochemistry for changes in expression of CSPG, NT-3, astrocytes, neurons and microglia/macrophages. Motor function was assessed using the Basso, Beattie and Bresnahan (BBB) locomotor scale. Animals that received the combination treatment of LV shNG2 and LV NT-3 showed reduced scar size. These animals also showed an increase in levels of neurons and NG2, a decrease in levels of astrocytes and a significant functional recovery as assessed using the BBB locomotor scale at 2 weeks post-treatment. The improvement in locomotor recovery and decrease in scar size shows the potential of this gene therapy approach as a therapeutic treatment for SCI.

This article presents a performance evaluation of a flexible manufacturing system (FMS), in which the system and its components can have different performance levels ranging from perfect functioning to total failure. An original multistate transition process for the FMS’s main component, Computerized Numerical Control (CNC) machine tools, proposed in the current article and treated as an aging process because of its increasing failure rate is investigated. The Markov reward approach is utilized to analyze the availability of CNC machine tools and sensitivity analysis of each non-operational state to the machine’s downtime. To overcome the “state explosion problem” in the subsequent FMS performance evaluation process, an application of the Lz-transform method is proposed to determine the performance evaluation and availability of such aging multistate FMS. An illustrated case is presented to demonstrate the performance evaluation process. The results show that the suggested methods can be implemented in engineering decision making and construction of various aging multistate FMS related to demands, availability, and output performance.

Considering the influence of performance degradation on a product’s traumatic failure, under the condition that only degradation data are observed and no traumatic failure data are observed, this paper proposes a subdistribution competing risk model to achieve a motorized spindle life assessment. This paper assumes that the failure rate ratio of the tested products does not change with time under different stress levels. Basic reliability with zero traumatic failure data is modeled by a unilateral confidence limit method under a two-parameter Weibull distribution. Performance degradation data are taken as covariates. The regression coefficients of the covariates are calculated by SPSS software. Then, a subdistribution competing risk model is constructed, which reflects the dependency relationship between reliability and performance degradation, and the product’s reliability life can be evaluated accordingly. The correctness and advantages of the model built in this paper are verified by a case analysis combined with the performance degradation information of a motorized spindle.
