Microorganisms of plant phyllosphere play an important role in plant health and productivity and are influenced by abiotic and biotic factors. In this study, we investigated the phyllosphere bacterial communities of three cigar tobacco varieties cultivated in Guangcun (GC) and Wuzhishan (WZS), Hainan, China. Metagenomic DNA was extracted from tobacco leaf samples and sequenced by 16S rDNA amplicon sequencing. Our results showed that bacterial communities of cigar tobacco phyllosphere in GC exhibited remarkably higher alpha diversity than that in WZS. There was slight effect of tobacco genotype variations on the alpha diversity in both cultivation sites, and beta diversity and structure of bacterial community were not influenced significantly by the cultivation sites and tobacco varieties. Statistical analyses of species diversity unraveled that the dominant species in bacterial communities of cigar tobacco phyllosphere among all these samples were phylogenetically affiliated to Proteobacteria and Cyanobacteria. At the genus level, the most abundant microorganism was Limnobacter, followed by Brevundimonas, unidentified_Cyanobacteria, and Pseudomonas. Additionally, environmental conditions except for humidity were negatively correlated with the relative abundance of bacterial genera. Further analyses revealed that influence of site‐specific factors on tobacco bacterial community was relatively higher than genotype‐specific factors. In short, this study may contribute to the knowledge base of practical applications of bacterial inoculants for tobacco leaf production.

Water and nitrogen are the key factors affecting the yield and nitrogen recovery efficiency of double-cropping rice, but information about optimizing nitrogen fertilizer and irrigation management to achieve high yield is still limited. The purpose was to study the effects of different nitrogen application rates (D1, D2, D3, D4 (0, 112.5, 150.0, and 187.5 kg ha−1 for early-season rice and 0, 135, 180, and 225 kg ha−1 for late-season rice)) and irrigation conditions (G1, Alternate wetting and drying irrigation, G2, flooding irrigation) on rice yield and nitrogen recovery efficiency. Field experiments were carried out in the early and late seasons of the subtropical environment in Heshan County, Hunan Province, China in 2018 and 2019. The results showed that the yield was increased by the comprehensive action of reasonable irrigation mode and nitrogen management. Under G1D4 and G1D3 treatments, the maximum grain yield of early-season rice was 7.42 t ha−1, which was 0.8~35.9% higher than other treatments, and that of late-season rice was 8.20 t ha−1, which was 13.3~67.0% higher than other treatments. The increase of yield in G1D4 and G1D3 treatments was due to the increase in dry matter accumulation, effective Panicles number, and Spikelets per panicle, whose increase was due to an increase in photosynthesis and nitrate reductase activity. Compared with other treatments, late-season rice G1D3 treatment achieved a higher yield with less nitrogen fertilizer, while early-season rice G1D4 treatment achieved the highest yield, but the nitrogen recovery efficiency decreased. We should focus on more effective nitrogen fertilizer and irrigation management to further improve the yield potential of double-cropping rice.

It is important to ensure the nutritional quality and safe production of rice. Here, plot experiments were used to analyze the effects of three soil amendments—10 t ha−1 of biochar (BC), 1.5 t ha−1 of lime (LM), and 2.25 t ha−1 of silicon–calcium fertilizer (SC)—on the soil characteristics, rice yield and quality of double-cropping rice grown in mildly cadmium-polluted paddy fields. Compared with the control treatment (CK), the BC and SC treatments significantly improved rice processing, appearance and nutritional quality, but reduced cooking quality. All three soil amendments significantly reduced cadmium (Cd) content in brown rice. Soil amendments could significantly increase soil pH and reduce soil available Cd content. The application of the BC and SC treatments increased the content of each nutrient index in the soil (SOM, NN, AP, AK). Correlation analysis showed that the improvement in rice processing, appearance, and nutritional quality was mainly affected by the comprehensive effects of soil SOM, NN, AP and AK; the hygiene quality was mainly affected by soil pH and available Cd. In terms of benefit analysis combined with cost, the SC treatment had the highest benefit effect. Taken together, in mildly cadmium-polluted paddy fields, the application of silicon–calcium fertilizer improved the soil quality, thereby increased the yield and quality of rice, and had the best effect on increasing income.

Rooting ability of rice seedling for mechanical transplanting has a large impact on grain yield. This study explored the relationship between endogenous soluble sugar content and rooting ability of rice seedlings. We placed 15-day-old rice seedlings in controlled environment cabinets with stable light and sampled after 0, 3, 6, 9, 12, and 24 hours of light to measure their soluble sugar content, nitrate content, starch content, soluble protein content and rooting ability. The soluble sugar content of the rice seedlings before rooting increased rapidly from 65.1 mg g-1 to 126.3 mg g-1 in the first 9 hours of light and then tended to stabilize; however, few significant changes in the other physiological indices were detected. With the light exposure time increasing from 3 hours to 12 hours, the rooting ability measured with fresh weight, dry weight, total length, and number of new roots increased by 91.7%, 120.0%, 60.6% and 30.3%, respectively. Rooting ability was related more closely to soluble sugar content than to nitrate-nitrogen content of rice seedlings before rooting and their correlation coefficients were 0.8582–0.8684 and 0.7045–0.7882, respectively. The stepwise regression analysis revealed that the soluble sugar content before rooting explained 73.6%–75.4% of the variance, and the nitrate-nitrogen content explained an additional 7.3%–14.2% of the variance in rooting ability, indicating that compared with nitrate-nitrogen content, soluble sugar content of rice seedlings before rooting was more dominant in affecting rooting ability. This study provides direct evidence of the relationship between the rooting ability and endogenous soluble sugar content of rice seedlings.

Soil contamination by heavy metals has become a prevalent topic due to their widespread release from industry, agriculture, and other human activities. Great progress has been made in elucidating the uptake and translocation of cadmium (Cd) accumulation in rice. However, there is still little known about corresponding progress in maize. In the current study, we performed a comparative RNA-Seq-based approach to identify differentially expressed genes (DEGs) of maize immature kernel related to Cd stress. In total, 55, 92, 22, and 542 DEGs responsive to high cadmium concentration soil were identified between XNY22-CHS-8 vs. XNY22-YA-8, XNY22-CHS-24 vs. XNY22-YA-24, XNY27-CHS-8 vs. XNY27-YA-8, and XNY27-CHS-24 vs. XNY27-YA-24, respectively. The weighted gene co-expression network analysis (WGCNA) categorized the 9599 Cd stress-responsive hub genes into 37 different gene network modules. Combining the hub genes and DEGs, we obtained 71 candidate genes. Gene Ontology (GO) enrichment analysis of genes in the greenyellow module in XNY27-YA-24 and connectivity genes of these 71 candidate hub genes showed that the responses to metal ion, inorganic substance, abiotic stimulus, hydrogen peroxide, oxidative stress, stimulus, and other processes were enrichment. Moreover, five candidate genes that were responsive to Cd stress in maize kernel were detected. These results provided the putative key genes and pathways to response to Cd stress in maize kernel, and a useful dataset for unraveling the underlying mechanism of Cd accumulation in maize kernel.
