Background: Simao pine is one of the primary economic tree species for resin and timber production in southwest China. The exploitation and utilization of Simao pine are constrained by the relatively lacking of genetic information. Construction a fine genetic linkage map and detecting quantitative trait locis (QTLs) for growth-related traits is a prerequisite section of Simao Pine's molecular breeding program. Results: In our study, a high-resolution Simao pine genetic map employed specific locus amplified fragment sequencing (SLAF-seq) technology and based on an F1pseudo-testcross population has been constructed. There were 11,544 SNPs assigned to 12 linkage groups (LGs), and the total length of the map was 2,062.85 cM with a mean distance of 0.37 cM between markers. According to the phenotypic variation analysis for three consecutive years, a total of seventeen QTLs for four traits were detected. Among 17 QTLs, there were six for plant height (Dh.16.1, Dh16.2, Dh17.1, Dh18.1–3), five for basal diameter (Dbd.17.1–5), four for needle length (Dnl17.1–3, Dnl18.1) and two for needle diameter (Dnd17.1 and Dnd18.1) respectively. These QTLs individually explained phenotypic variance from 11.0–16.3%, and the logarithm of odds (LOD) value ranged from 2.52 to 3.87.Conclusions: In our study, a fine genetic map of Simao pine applied the technology of SLAF-seq has been constructed for the first time. Based on the map, a total of 17 QTLs for four growth-related traits were identified. It provides helpful information for genomic studies and marker-assisted selection (MAS) in Simao pine.

The study of litter can provide an important reference for understanding patterns of forest nutrient cycling and sustainable management. Here, we measured litterfall (leaves, branches, etc.) from a wet, evergreen, broad-leaved forest in Ailao Mountains of southwestern China on a monthly basis for 11 years (2005–2015). We measured the total biomass of litter fall as well as its components, and estimated the amount of C, N, P, K, S, Ca, and Mg in the amount of litterfall. We found that: The total litter of evergreen, broadleaved forest in Ailao Mountains from 2005 to 2015 was 7.70–9.46 t/ha, and the output of litterfall differed between years. This provides a safeguard for the soil fertility and biodiversity of the area. The total amount of litterfall and its components showed obvious seasonal variation, with most showing a bimodal pattern (peak from March to May and October to November). The majority of litterfall came from leaves, and the total amount as well as its components were correlated with meteorological factors (wind speed, temperate and precipitation) as well as extreme weather events. We found that among years, the nutrient concentration was sorted as C > Ca > N > K > Mg > S > P. The nutrient concentration in the fallen litter and the amount of nutrients returned showed a decreasing trend, but the decreasing rate was slowed through time. Nutrient cycling was influenced by meteorological factors, such as temperature, precipitation, and wind speed, but the nutrient utilization efficiency is high, the circulation capacity is strong, and the turnover time is short. Our results showed that although there was nutrient loss in this evergreen, broad-leaved forest, the presence of forest litterfall can effectively curb potential ecological problems in the area.

Background: Grassland plays an important role in the ecosystem, but overgrazing harms the grassland system in many places. Grazing prohibition is an effective method to restore grassland ecosystems, and it plays a great role in realizing the sustainable development of grassland systems. Therefore, it is necessary to carry out research on the influence of regional grazing prohibition on the physical and chemical properties of different grassland systems. Methods: In Potatso National Park, Southwest China, we selected experimental plots in the artificial grazing meadow area to study the effects of grazing prohibition on plant and soil indexes in subalpine meadows and swamp meadows. We investigated the biomass and species diversity of grazing prohibition treatment and grazing treatment plots and sampled and tested the soil index. The variation percentage was used to remove the original heterogeneity and yearly variation, allowing us to compare differences in plant index and soil index values between grazing prohibition and grazing treatments. Results: Grazing prohibition increased the aboveground biomass, total biomass, total meadow coverage, average height, richness index, Shannon diversity index and evenness index and reduced the belowground biomass and root/shoot ratio in the subalpine meadow and swamp meadow. Additionally, grazing prohibition reduced the pH and soil bulk density and increased the soil total carbon, soil organic carbon, soil total nitrogen, soil hydrolyzable nitrogen, soil total phosphorus and soil available phosphorus in the subalpine meadow and swamp meadow. Nonmetric multidimensional scaling (NMDS) analysis showed that both plant indexes and soil indexes were significantly different between grazing and grazing prohibition treatments and between meadow types. Short-term grazing prohibition had a great impact on improving the fertility of meadow soil in the study area. We suggest that long-term and extensive research should be carried out to promote the restoration and sustainable development of regional grassland systems.

Background: Soil respiration (RS) plays an important role in the concentration of atmospheric CO2 and thus in global climate patterns. Due to the feedback between RS and climate, it is important to investigate RS responses to climate warming. Methods: A soil warming experiment was conducted to explore RS responses and temperature sensitivity (Q10) to climate warming in subtropical forests in Southwestern China, and infrared radiators were used to simulate climate warming. Results: Warming treatment increased the soil temperature and RS value by 1.4 °C and 7.3%, respectively, and decreased the soil water level by 4.2% (%/%). Both one- and two-factor regressions showed that warming increased the Q10 values by 89.1% and 67.4%, respectively. The effects of water on Q10show a parabolic relationship to the soil water sensitivity coefficient. Both RS and Q10 show no acclimation to climate warming, suggesting that global warming will accelerate soil carbon release.

Background Simao pine (Pinus kesiya Royle ex Gordon var. langbianensis (A. Chev.) Gaussen) is one of the most important tree species in the production of timber and resin in China. However, the genetic diversity of the natural populations has not been assessed to date. In this study, sequence related amplified polymorphism (SRAP) markers were used to investigate the genetic composition of natural Simao pine populations. Method The SRAP markers were applied and their efficiency was compared using various statistical multivariate methods, including analysis molecular of variance (AMOVA), the unweighted pair group method with arithmetic mean (UPGMA), and Principal coordinate analysis (PCoA). Results The 11 populations revealed a high level of genetic diversity (PPB = 95.45%, H = 0.4567, I = 0.6484) at the species level. A moderately low level of genetic differentiation (Gst = 0.1701), and a slightly high level of gene flow (Nm = 2.4403) were observed among populations using AMOVA. Eleven populations of Simao pine were gathered into four distinct clusters based on molecular data, and the results of UPGMA and PCoA also illustrated that assignment of populations is not completely consistent with geographic origin. The Mantel test revealed there was no significant correlation between geographic and genetic distance (r = 0.241, p = 0.090). Discussion The SRAP markers were very effective in the assessment of genetic diversity in Simao pine. Simao pine populations display high levels of genetic diversity and low or moderate levels of genetic differentiation due to frequent gene exchange among populations. The low genetic differentiation among populations implied that conservation efforts should aim to preserve all remaining natural populations of this species. The information derived from this study is useful when identifying populations and categorizing their population origins, making possible the design of long term management program such as genetic improvement by selective breeding.

Docynia indica (Wall.) Decne. (Duo-Yi) is a high economic value for exploitation and utilization wild fruit tree species with edible and medicinal values in southwest China. We have sequenced the chloroplast genome to facilitate genetic improvement of this species and to assess phylogenetic relationships among major lineages. The result showed that the total chloroplast genome size of Duo-Yi was 159,546 bp in length, containing a pair of inverted repeats (IRs) of 26,369 bp, which were separated by a large single copy (LSC) and small single copy (SSC) of 87,650 bp and 19,158 bp, respectively. The overall guanine-cytosine (GC) content of the chloroplast genome was 36.6%. There were 125 genes in the chloroplast genome, which including 82 protein-coding genes, 35 transfer RNA genes, and 8 ribosomal RNA genes. Among these genes, there were 14 genes with one intron and 3 genes have two introns. The result of phylogenetic analysis indicated that the Duo-Yi was closely related to the genera of Malus doumeri.
