Podophyllotoxin, along with its various derivatives and congeners are widely recognized as broad-spectrum pharmacologically active compounds. Etoposide, for instance, is the frontline chemotherapeutic drug used against various cancers due to its superior anticancer activity. It has recently been redeveloped for the purpose of treating cytokine storm in COVID-19 patients. Podophyllotoxin and its naturally occurring congeners have low bioavailability and almost all these initially discovered compounds cause systemic toxicity and development of drug resistance. Moreover, the production of synthetic derivatives that could suffice for the clinical limitations of these naturally occurring compounds is not economically feasible. These challenges demanded continuous devotions towards improving the druggability of these drugs and continue to seek structure-optimization strategies. The discovery of renewable sources including microbial origin for podophyllotoxin is another possible approach. This review focuses on the exigency of innovation and research required in the global R&D and pharmaceutical industry for podophyllotoxin and related compounds based on recent scientific findings and market predictions.

Development of antibiotic resistance in bacteria is one of the major issues in the present world and one of the greatest threats faced by mankind. Resistance is spread through both vertical gene transfer (parent to offspring) as well as by horizontal gene transfer like transformation, transduction and conjugation. The main mechanisms of resistance are limiting uptake of a drug, modification of a drug target, inactivation of a drug, and active efflux of a drug. The highest quantities of antibiotic concentrations are usually found in areas with strong anthropogenic pressures, for example medical source (e.g., hospitals) effluents, pharmaceutical industries, wastewater influents, soils treated with manure, animal husbandry and aquaculture (where antibiotics are generally used as in-feed preparations). Hence, the strong selective pressure applied by antimicrobial use has forced microorganisms to evolve for survival. The guts of animals and humans, wastewater treatment plants, hospital and community effluents, animal husbandry and aquaculture runoffs have been designated as “hotspots for AMR genes” because the high density of bacteria, phages, and plasmids in these settings allows significant genetic exchange and recombination. Evidence from the literature suggests that the knowledge of antibiotic resistance in the population is still scarce. Tackling antimicrobial resistance requires a wide range of strategies, for example, more research in antibiotic production, the need of educating patients and the general public, as well as developing alternatives to antibiotics (briefly discussed in the conclusions of this article).

Biofilm formation on surfaces via microbial colonization causes infections and has become a major health issue globally. The biofilm lifestyle provides resistance to environmental stresses and antimicrobial therapies. Biofilms can cause several chronic conditions, and effective treatment has become a challenge due to increased antimicrobial resistance. Antibiotics available for treating biofilm-associated infections are generally not very effective and require high doses that may cause toxicity in the host. Therefore, it is essential to study and develop efficient anti-biofilm strategies that can significantly reduce the rate of biofilm-associated healthcare problems. In this context, some effective combating strategies with potential anti-biofilm agents, including plant extracts, peptides, enzymes, lantibiotics, chelating agents, biosurfactants, polysaccharides, organic, inorganic, and metal nanoparticles, etc., have been reviewed to overcome biofilm-associated healthcare problems. From their extensive literature survey, it can be concluded that these molecules with considerable structural alterations might be applied to the treatment of biofilm-associated infections, by evaluating their significant delivery to the target site of the host. To design effective anti-biofilm molecules, it must be assured that the minimum inhibitory concentrations of these anti-biofilm compounds can eradicate biofilm-associated infections without causing toxic effects at a significant rate.

Rhizospheric plant–microbe interactions have dynamic importance in sustainable agriculture systems that have a reduced reliance on agrochemicals. Rhizosphere signaling focuses on the interactions between plants and the surrounding symbiotic microorganisms that facilitate the development of rhizobiome diversity, which is beneficial for plant productivity. Plant–microbe communication comprises intricate systems that modulate local and systemic defense mechanisms to mitigate environmental stresses. This review deciphers insights into how the exudation of plant secondary metabolites can shape the functions and diversity of the root microbiome. It also elaborates on how rhizosphere interactions influence plant growth, regulate plant immunity against phytopathogens, and prime the plant for protection against biotic and abiotic stresses, along with some recent well-reported examples. A holistic understanding of these interactions can help in the development of tailored microbial inoculants for enhanced plant growth and targeted disease suppression.

The booming demand for energy across the world, especially for petroleum-based fuels, has led to the search for a long-term solution as a perfect source of sustainable energy. Lignocellulosic biomass resolves this obstacle as it is a readily available, inexpensive, and renewable fuel source that fulfills the criteria of sustainability. Valorization of lignocellulosic biomass and its components into value-added products maximizes the energy output and promotes the approach of lignocellulosic biorefinery. However, disruption of the recalcitrant structure of lignocellulosic biomass (LCB) via pretreatment technologies is costly and power-/heat-consuming. Therefore, devising an effective pretreatment method is a challenge. Likewise, the thermochemical and biological lignocellulosic conversion poses problems of efficiency, operational costs, and energy consumption. The advent of integrated technologies would probably resolve this problem. However, it is yet to be explored how to make it applicable at a commercial scale. This article will concisely review basic concepts of lignocellulosic composition and the routes opted by them to produce bioenergy. Moreover, it will also discuss the pros and cons of the pretreatment and conversion methods of lignocellulosic biomass. This critical analysis will bring to light the solutions for efficient and cost-effective conversion of lignocellulosic biomass that would pave the way for the development of sustainable energy systems.

The increase in the industrial revolution, urbanization, and deficit in crude oil resources urged scientists to look for sustainable and renewable energy resources. Microbial fuel cells (MFCs) are bio-electrochemical devices that convert the chemical energy of bio mass into electrical energy utilizing microorganisms as biocatalysts. MFC is an enthralling technology, and it has many important applications. There are different designs of MFC currently in use, each having their pros and cons; however, the stacked MFC is so far generating the highest power output. Different factors, discussed in this review, contribute toward the efficiency of the process. An ample amount of research has been done to convert the theoretical framework of MFC into a practical application; however, the later still remains a big challenge. A plethora of technical problems is in the way for MFC to be driven toward up-scaling and real-world applications. This review discusses electron transfer mechanisms in MFCs and important factors affecting the performance of MFC. Reactor design is a main limiting factor in MFC technology, and this review also focuses on recent improvements in reactor designs and modifications. Moreover, some main applications of MFC technology, limitations, recent advancements, and future prospects are discussed. GRAPHICAL ABSTRACT
