The aim of this study is to conduct a comparative assessment of the effectiveness of neural network models—U-Net, DeepLabV3+, SegNet and Mask R-CNN—for the semantic segmentation of micrographs of human mesenchymal stem cells (MSCs). A dataset of 320 cell micrographs annotated by cell biology experts was created. The models were trained using a transfer learning method based on ImageNet pre-trained weights. As a result, the U-Net model demonstrated the best segmentation accuracy according to the metrics of the Dice coefficient (0.876) and the Jaccard index (0.781). The DeepLabV3+ and Mask R-CNN models also showed high performance, although slightly lower than U-Net, while SegNet exhibited the least accurate results. The obtained data indicate that the U-Net model is the most suitable for automating the segmentation of MSC micrographs and can be recommended for use in biomedical laboratories to streamline the routine analysis of cell cultures.

This scoping review explores the application of artificial intelligence (AI) methods for analyzing mesenchymal stem cells (MSCs) images. The aim of this study was to identify key areas where AI-based image processing techniques are utilized for MSCs analysis, assess their effectiveness, and highlight existing challenges. A total of 25 studies published between 2014 and 2024 were selected from six databases (PubMed, Dimensions, Scopus, Google Scholar, eLibrary, and Cochrane) for this review. The findings demonstrate that machine learning algorithms outperform traditional methods in terms of accuracy (up to 97.5%), processing speed and noninvasive capabilities. Among AI methods, convolutional neural networks (CNNs) are the most widely employed, accounting for 64% of the studies reviewed. The primary applications of AI in MSCs image analysis include cell classification (20%), segmentation and counting (20%), differentiation assessment (32%), senescence analysis (12%), and other tasks (16%). The advantages of AI methods include automation of image analysis, elimination of subjective biases, and dynamic monitoring of live cells without the need for fixation and staining. However, significant challenges persist, such as the high heterogeneity of the MSCs population, the absence of standardized protocols for AI implementation, and limited availability of annotated datasets. To advance this field, future efforts should focus on developing interpretable and multimodal AI models, creating standardized validation frameworks and open-access datasets, and establishing clear regulatory pathways for clinical translation. Addressing these challenges is crucial for accelerating the adoption of AI in MSCs biomanufacturing and enhancing the efficacy of cell therapies.

This study evaluates the effectiveness of transfer learning with pre-trained convolutional neural networks (CNNs) for the automated binary classification of surgical suture quality (high-quality/low-quality) using photographs of three suture types: interrupted open vascular sutures (IOVS), continuous over-and-over open sutures (COOS), and interrupted laparoscopic sutures (ILS). To address the challenge of limited medical data, eight state-of-the-art CNN architectures—EfficientNetB0, ResNet50V2, MobileNetV3Large, VGG16, VGG19, InceptionV3, Xception, and DenseNet121—were trained and validated on small datasets (100–190 images per type) using 5-fold cross-validation. Performance was assessed using the F1-score, AUC-ROC, and a custom weighted stability-aware score (Scoreadj). The results demonstrate that transfer learning achieves robust classification (F1 > 0.90 for IOVS/ILS, 0.79 for COOS) despite data scarcity. ResNet50V2, DenseNet121, and Xception were more stable by Scoreadj, with ResNet50V2 achieving the highest AUC-ROC (0.959 ± 0.008) for IOVS internal view classification. GradCAM visualizations confirmed model focus on clinically relevant features (e.g., stitch uniformity, tissue apposition). These findings validate transfer learning as a powerful approach for developing objective, automated surgical skill assessment tools, reducing reliance on subjective expert evaluations while maintaining accuracy in resource-constrained settings.

The global burden of ischemic stroke requires a search for new factors that affect its risk and outcomes. Recent studies have shown that triiodothyronine could potentiate favorable stroke outcomes, but the reason for this is still unclear. To clarify the effects of the free triiodothyronine (fT3) level on stroke outcomes, we adopted a Mendelian randomization (MR) approach to evaluate their causal relationship. The genetic variants associated with the free triiodothyronine (fT3) level were obtained from the ThyroidOmics Consortium. Thirteen single-nucleotide polymorphisms, genetically predicting the fT3 level with a significance of p < 10−7, were adopted for MR analysis. Summary-level data for ischemic stroke outcomes (reported as a continuous variable, namely the modified Rankin score 3 months after stroke) was obtained from the GISCOME network. MR analyses were performed using the TwoSampleMR framework. The inverse-variance weighting method of MR analysis showed that a genetically predicted increase in fT3 level is associated with a reduction in ordinal Modified Rankin Scale scores (OR = 0.581, 95% CI 0.37–0.92, p = 0.0183). This study showed that higher fT3 levels could be causally associated with more favorable ischemic stroke outcomes and provides more evidence for the possibility of using thyroid hormone (TH) analogs to improve stroke outcomes.
