Chitosan is a promising environment friendly active polymer packaging material due to its biodegradability, exceptional film forming capacity, great mechanical strength, appropriate barrier property along with intrinsic antioxidant and antimicrobial features. Bifunctional reagent was used for producing water insoluble chitosan films. Biopolymeric films crosslinked by Genipin (Gp), which is a reagent of natural origin, should have high potential in food packaging. The influence of the ratio of functional groups in the chitosan-Gp system on film absorption in the visible and ultraviolet regions of the spectrum, sorption, physical, and mechanical properties of the films has been studied. The degree of chitosan crosslinking in the films obtained from solutions containing Gp was estimated using the experimental data on film swelling and water vapor sorption isotherms. It is demonstrated that crosslinking with genipin improves swelling, water resistance, and mechanical properties of the films.
The objectives of the study were as follows: (1) to develop two methods for the preparation of macroporous composite chitosan/hyaluronic acid (Ch/HA) hydrogels based on covalently cross-linked Ch and low molecular weight (Mw) HA (5 and 30 kDa); (2) to investigate some properties (swelling and in vitro degradation) and structures of the hydrogels; (3) to evaluate the hydrogels in vitro as potential biodegradable matrices for tissue engineering. Chitosan was cross-linked with either genipin (Gen) or glutaraldehyde (GA). Method 1 allowed the distribution of HA macromolecules within the hydrogel (bulk modification). In Method 2, hyaluronic acid formed a polyelectrolyte complex with Ch over the hydrogel surface (surface modification). By varying compositions of the Ch/HA hydrogels, highly porous interconnected structures (with mean pore sizes of 50–450 μm) were fabricated and studied using confocal laser scanning microscopy (CLSM). Mouse fibroblasts (L929) were cultured in the hydrogels for 7 days. Cell growth and proliferation within the hydrogel samples were studied via MTT-assay. The entrapment of low molecular weight HA was found to result in an enhancement of cell growth in the Ch/HA hydrogels compared to that in the Ch matrices. The Ch/HA hydrogels after bulk modification promoted better cell adhesion, growth and proliferation than the samples prepared by using Method 2 (surface modification).
Biomaterials based on hydroxyapatite with controllable composition and properties are promising in the field of regenerative bone replacement. One approach to regulate the phase composition of the materials is the introduction of biopolymer-based additives into the synthesis process. The purpose of present study was to investigate the formation of hydroxyapatite-based hybrid materials in the presence of 6–24% platelet-poor plasma (PPP) additive, at a [Ca2+]/[PO43−] ratio of 1.67, pH 11, and varying maturing time from 4 to 9 days. The mineral component of the materials comprised 53% hydroxyapatite/47% amorphous calcium phosphate after 4 days of maturation and 100% hydroxyapatite after 9 days of maturation. Varying the PPP content between 6% and 24% brought about the formation of materials with rather defined contents of amorphous calcium phosphate and biopolymer component and the desired morphology, ranging from typical apatitic conglomerates to hybrid apatite-biopolymer fibers. The co-precipitated hybrid materials based on hydroxyapatite, amorphous calcium phosphate, and PPP additive exhibited increased solubility in SBF solution, which defines their applicability for repairing rhinoplastic defects.
Silk fibroin (SF) holds promise for the preparation of matrices for tissue engineering and regenerative medicine or for the development of drug delivery systems. Regenerated fibroin from Bombyx mori cocoons is water-soluble and can be processed into scaffolds of various forms, such as fibrous matrices, using the electrospinning method. In the current study, we studied the correlation between concentrations of fibroin aqueous solutions and their properties, in order to obtain electrospun mats for tissue engineering. Two methods were used to prevent solubility in fibroin-based matrices: The conversion of fibroin to the β-conformation via treatment with an ethanol solution and chemical cross-linking with genipin (Gp). The interaction of Gp with SF led to the appearance of a characteristic blue color but did not lead to the gelation of solutions. To speed up the cross-linking reaction with Gp, we propose using chitosan-containing systems and modifying fibrous materials via treatment with a solution of Gp in 80% ethanol. It was shown that the composition of fibroin with chitosan contributes to an improved water resistance, reduces defective material, and leads to a decrease in the diameter of the fibers. The electrospun fiber matrices based on regenerated fibroin modified by cross-linking with genipin in water–alcohol solutions were shown to promote cell adhesion, spreading, and growth and, therefore, could hold promise for tissue engineering.
Applying of blood biopolymers to regulate the phase composition is promising in designing the hydroxyapatite-based hybrid biomaterials with controllable resorbability. Hybrid materials based on hydroxyapatite and platelet-poor plasma (PPP) were formed in conditions of chemical precipi-tation at pH 11, [Ca2+] / [PO43–] ratio 1.67, PPP volume fraction of 6–24%, maturing time of 4–9 days. Mineral component of the materials was represented as 53% hydroxyapatite / 47% amor-phous calcium phosphate after 4 days of maturation, and 100% hydroxyapatite after 9 days of maturation. Varying PPP content from 6% to 24% provided forming of materials with rather de-fined content of amorphous calcium phosphate and biopolymer component, having desired mor-phology ranging from typical apatitic conglomerates to hybrid apatite-biopolymer fibers. Co-precipitated hybrid materials based on hydroxyapatite and PPP are promising for bone regen-eration in osteoplastic and maxillofacial applications.
Silk fibroin (SF) extracted from Bombyx mori cocoons is a biocompatible and biodegradable polymer shows potential as matrices for tissue engineering and regenerative medicine, or con-trolled drug delivery systems. Regenerated fibroin, obtained by removing sericin from natural silk, is water-soluble. Its aqueous solutions can be processed into a variety of materials, for example, fibrous matrices, using the electrospinning method. In this article, the concentration dependences of the properties of aqueous solutions of regener-ated fibroin, that determine the possibility of obtaining electrospun mats for tissue-engineering applications, are studied. A slowdown in the growth of viscosity after a concentration of fibroin of 30% and a decrease in the transparency of the solution were found; this may be due to the on-set of conformational rearrangements: the transition of fibroin from the random coils, charac-teristic for dilute solutions, to the anisotropic conformation of the helix and the appearance of β-pleated sheets. Fibrous mats from regenerated fibroin were obtained from solutions with a concentration of 10 and 20%. Two methods were used to prevent solubility of fibroin-based matrices: conversion of fibroin to the β-conformation by treatment with an ethanol solution and chemical cross-linking with gen-ipin (Gp). The structures of the fibrous samples were studied by AFM and confocal laser scan-ning microscopy. The composition of the moulding solution for the electrospinning of regener-ated fibroin fibrous matrices and the conditions for their modification have been optimised. It has been shown that the composition of fibroin with chitosan contributes to a decrease in the solubility of electrospun fibrous matrices by increasing the number of amino groups available for modification by Gp, and also leads to a decrease in the diameter of the fibers. Electrospun fiber matrices based on regenerated SF, modified by cross-linking with Gp in water-alcohol solutions, promote cell growth and proliferation and may be promising for tissue engineering.
