Foodborne illnesses pose a serious threat to public health, with increasing global incidence rates driven by factors such as rising meat consumption. Rapid detection of foodborne pathogens in meat is critical for preventing outbreaks. This study investigates the potential of gas chromatography-mass spectrometry (GC-MS) and Fourier-transform infrared spectroscopy (FTIR) for identifying biomarkers and spectral fingerprints indicative of foodborne pathogens in raw chicken meat. Raw broiler chicken meat samples were surface-sterilized and inoculated with foodborne pathogens. The samples were challenge inoculated with the specific pathogen and the physical quality parameters like pH, color, texture, drip loss, and water activity were assessed. GC-MS analysis identified 113 metabolites, including potential biomarkers like ureidopropionic acid, 5-sulfosalicylic acid, 11,14-eicosadienoic acid, methyl ester for E. coli O157:H7; 11-bromoundecanoic acid, neocurdione, glafenin, eicosanoic acid for Salmonella; azepan-1-yl-acetic acid, methyl ester, tramadol, cytarabine, dipipanone for Staphylococcus and cyclopentaneundecanoic acid, phosphonofluoridic acid, î-n-formyl-l-lysine for Pseudomonas. Pathway analysis revealed the involvement of fatty acid metabolism and amino acid degradation pathways. FTIR spectral data showed significant variances between control and spiked samples, particularly in the fatty acid spectral region. The identified metabolites and spectral patterns could serve as biomarkers for developing rapid pathogen detection methods, contributing to enhanced food safety protocols.

Background: Embryo production in vitro requires three steps: in vitro maturation (IVM), in vitro fertilization (IVF) and in vitro culture (IVC). The first step in the in vitro maturation of oocytes is germinal vesicle breakdown (GVBD), followed by completion of the 1st meiotic division and formation of the 1st polar body. These parameters are critical during ovary and oocyte selection. This study aimed to evaluate bovine ovary and oocyte collection for IVM. Methods: Ovaries collected from cows in a 0.9% NaCl saline solution at the central slaughterhouse were divided into two normal and abnormal ovaries according to their morphological appearance. The morphometric dimensions of bovine ovaries, such as weight and deamination (length, width and volume), were recorded. The oocyte was extruded and its deamination occurred before and after culture in maturation medium. Result: The mean ovary weight, volume, length and width were not significantly different between the two ovary types. Additionally, the dominant and subordinate follicle diameters in both ovary types showed no significant differences. Furthermore, the oocyte number per ovary of the normal and abnormal oocytes showed no significant differences. The mean cumulus oocyte complex before maturation showed no significant difference (52.73±7.23 mm for the normal ovary vs. 43.015±5.41 mm for the abnormal ovary). However, after maturation, a highly significant difference was found (P less than 0.001) between the normal ovaries before and after maturation (178.10±15.36 µm) and abnormal ovaries (10.45±7.99 µm). Additionally, data analysis of oocytes with or without 1st polar bodies revealed a highly significant difference (P less than 0.001) between oocytes of the normal ovary with (32.15±4.19) and without (10.95±1.59) 1st polar bodies and oocytes of the abnormal ovary with (7.0±078) and without 1st polar bodies (3.3±0.32). Thus, the critical point at which the normal ovary produces better in vitro matured follicles with good oocyte quality and produces the 1st polar body determines developmental competence. Therefore, the best selection of normal ovaries will enhance in vitro maturation for subsequent experiments, such as in vitro fertilization or cloning and in vitro embryo development.

Rumex vesicarius (R. vesicarius or RV) is an annual plant having rounded leaves, with flowers containing seeds. RV protects liver, resists cancer, and removes free radicals in cells. The aim of this study was to illustrate the effect of the R. vesicarius water seeds extract (RVWSE) treatment on mice before and during pregnancy, and its action on the liver and kidney histology and enzymes, aspartate aminotransferase and alanine aminotransferase, the blood urea nitrogen, the creatinine, and the uric acid. The seeds of R. vesicarius were collected, extracted, and its component analyzed via Gas chromatography mass spectrometry. Mice treated with 10 mg kg−1 RVWSE via feeding tube for 1 week before mating, or during pregnancy. The number of offspring or litter size was recorded and the blood sample was collected at the end of the experiment to test the kidney and liver enzymes, and their histology. Results showed that RVWSE contains different phytochemicals consisting of some hexane and chloroform compounds. The fertility rate of 1st treated pre-pregnancy group is 30% and the 2nd group is 35%. The offspring rate of 1st treated group showed higher new born rate (8 new born/female) than 2nd treated group (5.4 new born/female). The blood enzyme levels of the kidney and liver showed some variation between the two groups and their histology illustrates some non-significant variation between the treated and control groups of mice. The implications of the results of this study illustrate the safe use of the RVWSE, its effectiveness in improving mice fertility, and positive impact on biomarker of serum enzymes of liver and kidney with their histopathology.

The cloning between different animals known as interspecific somatic cell nuclear transfer (iSCNT) was carried out for endangered species. The iSCNT has been characterized by a poor success rate due to several factors that influence the formation of the SCNT in various cytoplasms. The cell cycle of the transferred somatic cell, the passage number of the cultured somatic cell, the mitochondria oocytes, and their capabilities are among these factors. This study investigates the role of the passage number of the Arabian Oryx somatic cell culture when transplanted to an enucleated domestic cow oocyte and embryo developmentin vitro. The fibroblast somatic cell of the Arabian Oryx was cultured for several passage lanes (3–13). The optimal passage cell number was found to be 10–13 Oryx cell lines that progressed to various cell stages up to the blastula stage. There was some variation between the different passage numbers of the oryx cell line. The 3–9 cell line did not show a good developmental stage. These could be attributed to several factors that control the iSCNT as stated by several investigators. More investigation is needed to clarify the role of factors that affect the success rate for the iSCNT.

This study aimed to evaluate the effects of marjoram extract on oocyte maturation and gene expression in sheep oocytes and embryos. The first experiment studied the effect of the extract as an antioxidant to improve the in vitro maturation media used for sheep oocytes; the oocytes were matured in a TCM199 medium supplemented with 1 or 10 µg/mL of marjoram extract or the control, 0 µg, for 24 hr. Then, the maturation was estimated, and the gene expression was measured by using qPCR. The second experiment studied the effect of the extract on the development of sheep embryos produced in vitro; the fertilized oocytes were cultured in a SOF medium supplemented with 1 or 10 µg/mL of marjoram extract or the control, 0 µg, for 7 days. Then, the gene expression was measured using qPCR. The results showed that the marjoram extract did not improve nuclear maturation or the blastocyst rate. There was a significant increase in the level of GDF-9 gene expression in mature oocytes in the treatment groups. An increase in the expression of BCL-2 and EGR-1 genes was observed for the blastocysts in the 10 µg/mL group. We concluded that the marjoram extract did not improve nuclear maturation, but it did affect the expression of some genes in sheep oocytes and embryos.

Background: The utilization of in vitro fertilization (IVF) and in vitro embryo production (IVEP) has increased significantly as an effective tool of assisted reproductive technology. Sperm capacitation is a process of membrane maturation allowing the acrosome reaction to take place. Ionomycin is a calcium ionophore that induces the acrosome reaction by increasing calcium cycling across phospholipid membranes. The aim of this study was to enhance the cleavage rate and development of bovine embryos. Methods: Bovine ovaries were collected from the central slaughter house. Oocytes were obtained from follicles and were let to mature for 24 h. Frozen bull semen thawed in 37°C water and transferred to a discontinuous percoll gradient (Sigma GE17-0891-01). The spermatozoon pellet was separated into four groups after resuspension in bovine in vitro fertilization media (BO-IVF): control without additives; with DMSO and with ionomycin (25 and 50 nM) and kept in an incubator for three hours. The oocytes matured under commonly used media (TCM-199) for 24 h. The mature oocytes were then fertilized for 18-24h. The embryos were cultured in SOF medium until the seventh day to study cleavage rate and blastocyst rate. Result: The results indicated that 50 nM ionomycin supplementation considerably reduced cleavage while DMSO and 25 nM ionomycin supplementation had no effect on cleavage percentage. Moreover, when compared to the other groups, ionomycin at 25 nM significantly increased blastocyst development. In conclusion, the highest rate of blastocyst development was observed in the capacitation medium supplemented with 25 nM ionomycin, apparently due to the importance of calcium and metabolism in spermatozoa for fertilization and embryo development.
