Total mercury (THg), methylmercury (MeHg), and selenium (Se) concentrations were measured in various commercially important fish species. The benefit–risk binomial associated with these chemicals was assessed in children through the probability of exceeding the provisional tolerable weekly intakes (PTWIs) of the contaminants and the Se recommended dietary allowance (RDA). The Se:Hg molar ratios, selenium health benefit values (HBVSe), and monthly consumption rate limits (CRmm) for each species were also calculated. THg and Se were analyzed by atomic absorption spectrophotometer (Shimadzu, Milan, Italy), while MeHg was determined by Trace Ultra gas chromatograph connected with a PolarisQ MS (Thermo Fisher Scientific, Waltham, MA, USA). None of the analyzed fish had Hg levels above the European Community regulatory limits, while most large predators had MeHg levels over the threshold concentration set by US EPA. The estimated weekly intakes of THg and MeHg exceeded in many cases the PTWIs and the Se estimated daily intakes were provided from 0.71% to 2.75% of the RDA. Se:Hg molar ratios above 1 and positive HBVSe index suggested that Se in fish could be enough to alleviate the potential toxic effect of Hg. However, high-risk groups as children should consume fish in moderation because a large consumption pattern, especially of swordfish and tunas, might be of concern for health.

The concentrations of nine trace elements (Hg, Cd, Pb, V, Ni, Co, Cr, Cu and Zn) were determined in sixteen fish species collected in commercial centers of South Italy. Additionally, the assessment of risk indices (Hazard Quotient, Hazard Index, Incremental Lifetime Cancer Risk) through the consumption of various fish species were estimated in different age groups. The concentrations fell under the maximum permissible levels (MPLs) for human consumption recommended by European Union, with an only exception regarding Cd, which exceeded the tolerable value in Atlantic bluefin tuna samples. The estimated weekly intakes of metals, all below Provisional Tolerable Weekly Intakes (PTWIs), were in the following decreasing order: children > teenagers > adults > elders. The estimated hazard quotient (HQ) of Hg being above or close to 1 indicated a high probability of experiencing non-carcinogenic health risks, while the noncarcinogenic threats associated with exposure to other metals were insignificant (< 1) for all age groups. The cumulative cancer risk (ΣILCR) in children (1.86 × 10−3) and teenagers (1.10 × 10−3) exceed the cancer benchmark by a greater margin than adults (7.53 × 10-4) and elders (6.43 × 10-4). These results impose a systematic monitoring of fish, especially of large predatory fish, so to ensure the continuing safety of this food safeguarding human health.

In order to investigate PCB and PCDD/F concentrations and potential human health risk a study has been conducted in meat samples (beef, pork, chicken and turkey) purchased from Italian supermarkets. PCBs were dominant (41.8-77.7 ng g-1 l.w.) with respect to PCDD/Fs (20.1-91.1 pg g-1 l.w.). The levels were variable and largely dependent upon the type of meat. Accumulation pattern showed a distribution typically reported for meat. PCBs tended to decrease from lowest to highest congeners, while for PCDD/Fs the concentration profile was dominated by highly chlorinated dioxins and furans. Concentrations of PCDD/Fs plus dl-PCBs and indicator PCBs exceeding the EU maximum permissible levels were found in 23.3% and 53.3% of the samples, respectively. Exposure estimates to PCDD/Fs plus dl-PCBs (0.08-4.16 pg WHO-TEQs kg-1 b.w. w.-1) were within the new limit proposed by the EFSA's expert panel, except for pork sausage, showing a twofold higher value. Concerning the exposure derived from indicator PCBs, the maximum level set by different European countries (10 ng kg-1 b.w. d.-1) was surpassed solely via consumption of pork sausage (17.22 ng kg-1 b.w. d.-1), thought also the estimated intake from hamburger consumption (6.88 ng kg-1 b.w. d.-1) merits attention being close to guideline limit.

Polychlorinated biphenyls (PCBs) and polychlorinated dibenzo-furans (PCDD/Fs) were measured in Mediterranean bluefin tuna (Thunnus thynnus) to verify the compliance with the EU regulations for food commercialization. The estimated intakes were also evaluated. The analyses were performed by gas chromatography-ion trap tandem mass spectrometry (GC-MS-MS). The PCBs were dominant (1132.0 ng g−1 l.w.), followed by PCDFs (23.2 pg g−1 l.w.) and PCDDs (8.5 pg g−1 l.w.). The pollutant levels (dl-PCBs: 0.7 pg TEQ/g w.w.; PCDD/Fs: 1.9 pg TEQ/g w.w.) and their sum expressed as TEQ values (2.6 pg TEQ/g w.w.) remained below the limits for human consumption proposed by the European Union. On the contrary, the sum of the six indicator non-dioxin-like PCBs (84.2 ng g−1 w.w.) was slightly above the maximum level fixed by the in-force legislation. The estimated dietary intakes for PCDD/Fs plus dl-PCBs were below the toxicological reference values (TRVs) set by various international bodies, while non-cancer and cancer risk assessment revealed a safety concern. Additionally, the estimated intake of ndl-PCBs exceeded the maximum levels set by different European countries. These findings suggest caution in tuna consumption together with an active and frequent surveillance of the chemical quality of its flesh.

Trace element (Hg, Pb, Cd, Zn, Cu, Ni, and Cr) occurrence was determined in the muscle tissue of swordfish collected in the Mediterranean Sea to assess whether the intakes complied with the recommended levels for essential metals and permissible levels for toxic elements. Metals were analyzed by an atomic absorption spectrophotometer (Shimadzu AA 7000). The methodology of Target Hazard Quotient (THQ) was also evaluated. The ranking order of toxic metal concentration was Hg > Cd > Pb, while for essential elements the distribution pattern followed the sequence Zn > Cu > Ni > Cr. The Estimated Weekly Intakes (EWI) as well as THQ for Cd and Pb indicated that swordfish consumption did not pose a risk to human health, whereas the major concern was for Hg. Fish size-related changes in Hg concentrations resulted in high EWI and THQ values relative to larger fish consumption, implying a potential risk to human health. For consumer protection, catches of swordfish approximately above 44 kg should be avoided as these fish have a higher risk of containing toxic levels of Hg.

The aim of this study was to assess the growth and survival of Escherichia coli O157:H7 during the manufacturing and ripening of Cacioricotta goat cheese. Goat milk was artificially contaminated with E. coli O157:H7 and the bacterial load was monitored from production up to 90 days of ripening. Goat milk was inoculated with 102 cfu ml-1 of E. coli O157:H7 and the bacterial count of the curd at time zero was 2.31 log10 cfu g-1. During the first day of ripening, the bacterial load has increased to 5.73 log10 cfu g-1 to more than 6.20 log10 cfu g-1 during the first week. The bacterial load remained constant up to 28 days and then slightly decreased until the end of ripening, with values of aw and pH of 0.88 and 5.41 respectively. The results of this study highlighted that E. coli O157:H7 is able to survive the manufacturing process and they suggest that the 90-day period of ripening alone is insufficient to remove E. coli O157:H7 in contaminated Cacioricotta goat cheese. Moreover, these results support the assumption that the presence of a low contamination of milk with E. coli O157:H7 could represent a potential source of infection and a threat to consumers.

The objectives of this study were to determine and compare the concentrations of Hg, Cd, Pb, Zn, Cu, Ni and Se in the liver of macrourid fish as Trachyrinchus scabrus, Nezumia sclerorhynchus and Coelorhynchus coelorhynchus from the Mediterranean Sea, Italy. It was also carried out to evaluate the relationship between metal concentration and fish size and to explore selenium/mercury molar ratio. The highest concentrations were in T. scabrus, followed by N. sclerorhynchus and C. coelorhynchus. In all species, any element displayed significant correlation between metal body burden and fish size, except Hg. The mean selenium/mercury ratios were greater than one in all fish species indicating that Se antidotal effect in counteracting Hg occurred. This report represents one of the few surveys providing information on trace metal in deep-sea fish from Mediterranean Sea constituting, thus, an essential baseline work with which future levels may be compared.

Pangasius hypophthalmus fillets, classified according to different frozen technologies, were subjected to microbiological (Psychrophilic Total Aerobic Count = PTAC, Enterobacteriacea, Staphylococcus aureus, Salmonella spp. and Listeria monocytogenes) and chemical analyses (Hg, Pb, Cd and six “indicator” congeners of polychlorinated biphenyls=PCB6). Mean count of psychrophilic aerobic total bacteria, Enterobacteriaceae and S. aureus in all samples examined was 4.44 log CFU/g, 2.16 log CFU/g and 1.07 log CFU/g, respectively. Any sample showed the presence of Salmonella, while L. monocytogenes was encountered in 2.1% of samples tested. The microbiological counts were different among fillet categories, but with values always lower than the limit of ICMSF. Also the chemical quality of processed products is satisfactory being the concentrations of all metals and six “indicator” PCBs below safety levels for human consumption. This study has contributed to addressing the current paucity in literature on the microbial and chemical status of frozen P. hypophthalmus fillets. Practical Applications: The present research reveals a good chemical and microbiological quality of the frozen Pangasius fillets, although the samples examined show a low incidence of Listeria monocytogenes and Staphylococcus aureus. Thus, it would be desirable a greater attention to the operator's personal hygiene practices during the processing procedures, appropriate preservation techniques and sanitation programs in the processing plants to avoid cross-contamination. The data contained in this study may also be useful for national and international food safety regulatory authorities

Mercury (Hg) and cadmium (Cd) were quantified in fish, cephalopods and crustaceans from Italian supermarkets. Sample compliance with European dietary standards as well as human health risks according to provisional tolerable weekly intake (PTWI) and the methodology of target hazard quotient (THQ) were evaluated. Both element levels were under European legal limits, except for some fish having Hg and Cd contents exceeding or equal to critical values. Estimated weekly intakes (Hg: fish = 0.07–1.44 µg kg−1 bw week–1; cephalopods = 0.05–0.15 µg kg−1 bw week–1; crustaceans = 0.04–0.08 µg kg−1 bw week–1; and Cd: fish = 0.04–0.32 µg kg−1 bw week–1; cephalopods = 0.07–0.27 µg kg−1 bw week–1; crustaceans = 0.05–0.11 µg kg−1 bw week–1) as well as THQ < 1 were within safe limits. Although there seems to be no important risks associated with seafood consumption, Hg exposure was in some cases close to safety margins and thus levels of this metal should be under frequent surveillance.

PCBs including “indicator” and “dioxin-like” congeners were measured in a marine gastropod, Hexaplex trunculus, from Mediterranean areas exposed to anthropogenic pressure, to evaluate their compliance with European Union (EU) legislation. Total PCB concentrations showed location-dependent variation, and were higher in samples from Ionian Sea-Taranto (1001 ng g−1 lipid weight) compared with those from Adriatic Sea-Bari (519 ng g−1 lipid weight) and Adriatic-Sea-Brindisi (253 ng g−1 lipid weight). All sample concentrations were compliant with EU regulation limits for the sum of six “indicator” PCBs (Ionian Sea-Taranto: 40.7 ng g−1 wet weight; Adriatic Sea-Bari: 24.0 ng g−1 wet weight; Adriatic Sea-Brindisi: 11.0 ng g−1 wet weight). Also “dioxin-like” PCBs content expressed as toxic equivalent (TEQ) values, in samples from Adriatic Sea (Adriatic Sea-Bari: 2.48 pg WHO-TEQ/g wet weight; Adriatic Sea-Brindisi: 2.14 pg WHO-TEQ/g wet weight), were below the limit, while the specimens from the Ionian Sea (Ionian Sea-Taranto: 3.53 pg WHO-TEQ/g wet weight) showed levels exceeding the maximum threshold

Concentrations of 11 polycyclic aromatic hydrocarbons (PAHs) and toxic metals (Hg, Cd and Pb) were measured in three fish species [(Raja miraletus (brown ray), Lepidorhombus whiffiagonis (megrim) and Lophius piscatorius (angler)] from Mediterranean Sea (Adriatic Sea). No significant difference in PAHs and metal concentrations was encountered among the fish species examined. Total PAH concentrations ranged from 209.9ngg-1 wet weight to 227.2ngg-1 wet weight. Low-molecular weight (two and three rings) PAHs were observed dominating over the high molecular weight (from four to six rings) PAHs. With regard to metals, Hg exhibited the highest concentrations (0.68-0.98μgg-1 weight wet), followed by Pb (0.08-0.12μgg-1 weight wet) and Cd (0.05-0.10μgg-1 weight wet). The health risks by consumption of these species were assessed and did not present threat to public health concerning PAH, Pb and Cd intakes. However, the estimated exposure from Hg illustrates the importance of limiting the dietary consumption of larger-sized fish

Concentrations of 7 polychlorinated dibenzo-p-dioxins (PCDDs), 10 polychlorinated dibenzofurans (PCDFs) and 22 polychlorinated biphenyls (PCBs), including 12 dioxin like-PCBs (non- and mono-ortho PCBs) were measured in 80 sheep milk samples from farms located in an industrialized area of Sardinia, Italy. PCDDs and PCDFs mean concentrations were 2.45 and 3.69 pgg(-1) fat basis, respectively. The mean dl-PCB concentration was 2.01 ngg(-1) fat basis, while cumulative ndl-PCB levels ranged from 1.02 to 20.42, with a mean of 4.92 ngg(-1) fat. The results expressed in pg WHO-TEQ/g fat showed that contamination level of milk was below the limit values for human consumption established by EC legislation. In the same way, all the investigated milk exhibited PCDD/Fs concentrations below EU action levels, while dl-PCBs concentrations exceeded the action level of 2.0 pg WHO-TEQ/g fat. These findings point to the need to continue to conduct general monitoring programmes, including also milk samples from areas not close to the contaminant-emitting industries, in order to better evaluate the impact of industrial activities on surrounding environment.

The most popular brands of salted anchovies (Engraulis encrasicolus) from the Mediterranean Sea and Atlantic Ocean were purchased from several Italian supermarkets and grocery stores. Heavy metal (Hg, Cd, and Pb) and polychlorinated biphenyl (PCB) levels were determined and assessed by comparing the concentrations in these samples with the maximum permissible limits set by the European Union (Reg EC 629/2008 and Reg EC 1881/2006 [Off. J. Eur. Union L 173:3–9 and 364:5–24, respectively]). The Hg and Cd levels were higher than those of Pb in all samples examined. For Hg and Pb, the concentrations recorded in this study were below the authorized limits, while an appreciable percentage of samples from both locations (Mediterranean Sea, 35%, and Atlantic Ocean, 25%) showed Cd levels exceeding the threshold recommended for human consumption. Concerning PCBs, the results of principal component analysis showed that samples from the two different marine areas appeared to be discriminate, with Mediterranean anchovies more contaminated than the others, in spite of their lower lipid content. However, anchovy samples from both locations had dioxinlike-PCB levels (Mediterranean Sea, 0.011 pg World Health Organization toxic equivalency [WHO-TEQ] g−1, wet weight, and Atlantic Ocean, 0.007 pg WHO-TEQ g−1, wet weight) that were below the WHO-TEQ maximum concentration set by European regulation. The results of this study will help in generating data needed for the assessment of heavy metal and PCB intake from this food.

Liver of blue shark (Prionace glauca) specimens from the South-Eastern Mediterranean Sea were analyzed for the presence of polychlorinated biphenyls (PCBs), including coplanar congeners, polychlorinated dibenzo-p-dioxins (PCDDs) and dibenzofurans (PCDFs). PCBs were the dominant chemicals, followed by PCDFs and PCDDs. The pattern of PCB congener concentrations in the hepatic tissue was dominated by higher chlorinated compounds. The specific profile of toxic PCDD/F congeners was characterized mainly by 2,3,7,8-TCDF and 2,3,7,8-TCDD, followed by 1,2,3,6,7,8-HxCDD and 2,3,4,6,7,8-HxCDF. The total 2,3,7,8-TCDD toxic equivalent (TEQs) was 149 pg g⁻¹ lipid wt. The profile of TEQ shows that PCDDs present the greatest risk to this species contributing to total toxicity with a percentage approximately of 60%, while the contribution of PCDFs and DL-PCBs is almost the same being 22.4% and 21.6%, respectively. Further investigations are urgently needed to characterize the PCDD/Fs contamination levels not only in elasmobranch fish but in all Mediterranean marine biota.

PCB and DDT concentrations were determined in the adipose tissue of cats and dogs from Southern Italy. In cats p,p′-DDE was the most abundant DDT component (95.0%), while in dogs these compounds were absent, except in two specimens. PCB concentrations were higher in cats (199.02 ng g− 1 lipid weight) than in dogs (41.61 ng g− 1 lipid weight). Also there were inter-specific differences in the contribution of the different congeners to PCBs, although PCB 138, PCB 153 and PCB 180 were the most representative congeners in both species. Animals from one location, Taranto City, had significantly elevated concentrations of dioxin-like PCBs compared to the other locations. Consequently the estimated mean 2,3,7,8-tetrachlorodibenzo-p-dioxin toxic equivalents (TEQs) of coplanar PCBs were higher in these animals (cats: 0.65 pg g− 1 lipid weight; dogs 0.29 pg g− 1 lipid weight) than in the other ones (cats: 0.12 pg g− 1 lipid weight; dogs: 0.001 pg g− 1 lipid weight).

Polybrominated diphenyl ethers (PBDEs) were investigated in 15 composite liver samples from two deep-sea fish species, the hollowsnout grenadier (Trachyrinchus trachyrinchus, TT) and the roughsnout grenadier (Coelorhynchus coelorynchus, CC). Mean concentrations of sum tri- to hepta-BDEs were higher in CC species than in the TT species (16.9 vs 4.5 ng/g lipid weight (lw), respectively). BDE 47 and BDE 100 were the major congeners in both species. Methoxylated PBDEs (MeO-PBDEs), reported as being naturally produced in marine environments, were also measured in all samples. Similar to those of PBDEs, mean concentrations of sum MeO-PBDEs (sum congeners 2′-MeO-BDE 68 and 6-MeO-BDE 47) were higher in the CC species (28.9 vs 6.6 ng/g lw, respectively). Interestingly, polybrominated hexahydroxanthene derivatives (PBHDs) had the highest contribution to the total load of organobromines in the deep-sea fish samples. Contrary to those of PBDEs and MeO-PBDEs, the mean concentrations of PBHDs were higher in the TT species (7040 vs 530 ng/g lw, respectively). Furthermore, an unusual profile was seen in the TT species, for which a tetrabrominated BHD isomer was the predominant isomer (up to 98% of the sum PBHDs). This differs from the profiles of PBHDs dominated by a tribrominated BHD isomer reported until now in the literature. The mixed halogenated monoterpene (MHC-1) and 2,4,6-tribromoanisole (TBA) were detected as well, albeit at lower concentration (<5 ng/g lw), supporting the hypothesis that these two natural products are rather abundant in shallow waters.

This study investigated the subcellular distribution of Cd, Cu and Zn in liver and kidney of green turtles (Chelonia mydas) stranded along the Italian coast of the South Adriatic Sea (Eastern Mediterranean). Cd and Zn mean concentrations did not differ significantly between liver (4.26microgg(-1) and 34.53microgg(-1), respectively) and kidney (5.06microgg(-1) and 26.39microgg(-1), respectively), whereas the levels of Cu were significantly higher in liver (32.75microgg(-1)) than in kidney (8.20microgg(-1)) (p<0.009). Most of Cd, Cu and Zn was present in hepatic and renal cytosol, and their concentrations increased with total levels in both organs, indicating that cytosol has a crucial role in metal accumulation. Cd and Cu in hepatic and renal cytosol were present mostly in metallothionein fractions (MTs), whereas Zn was fractionated into MTs and high-molecular-weight-substances (HMWS). The comparison with the results of other investigations on individuals of the same species collected in different marine areas shows good agreement relatively to essential metals. For Cd our data are comparable with those encountered in specimens from the Mediterranean Sea (Cyprus) confirming the homogeneity of the area comprising the south-eastern basin of the Mediterranean Sea from an ecological point of view.
