Populus ussuriensis Kom. is a valuable forest regeneration tree species in the eastern mountainous region of Northeast China. It is known that diploid P. ussuriensis (CK) performed barely satisfactorily under salt stress, but the salt stress tolerance of polyploid (i.e., triploid (T12) and tetraploid (F20)) P. ussuriensis is still unknown. In order to compare the salt stress tolerance and salt stress response mechanism between diploid and polyploid P. ussuriensis, phenotypic observation, biological and biochemistry index detections, and transcriptome sequencing (RNA-seq) were performed on CK, T12, and F20. Phenotypic observation and leaf salt injury index analysis indicated CK suffered more severe salt injury than T12 and F20. SOD and POD activity detections indicated the salt stress response capacity of T12 was stronger than that of CK and F20. MDA content, proline content and relative electric conductivity detections indicated CK suffered the most severe cell-membrane damage, and T12 exhibited the strongest osmoprotective capacity under salt stress. Transcriptome analysis indicated the DEGs of CK, T12, and F20 under salt stress were different in category and change trend, and there were abundant WRKY, NAM, MYB and AP2/ERF genes among the DEGs in CK, T12, and F20 under salt stress. GO term enrichment indicated the basic growth progresses of CK, and F20 was obviously influenced, while T12 immediately launched more salt stress response processes in 36 h after salt stress. KEGG enrichment indicated the DEGs of CK mainly involved in plant–pathogen interaction, ribosome biogenesis in eukaryotes, protein processing in endoplasmic reticulum, degradation of aromatic compounds, plant hormone signal transduction, photosynthesis, and carbon metabolism pathways. The DEGs of T12 were mainly involved in plant–pathogen interaction, cysteine and methionine metabolism, phagosomes, biosynthesis of amino acids, phenylalanine, tyrosine and tryptophan biosynthesis, plant hormone signal transduction, and starch and sucrose metabolism pathways. The DEGs of F20 were mainly involved in plant hormone signal transduction, plant–pathogen interaction, zeatin biosynthesis, and glutathione metabolism pathways. In conclusion, triploid exhibited stronger salt stress tolerance than tetraploid and diploid P. ussuriensis (i.e., T12 > F20 > CK). The differences between the DEGs of CK, T12, and F20 probably are the key clues for discovering the salt stress response signal transduction network in P. Ussuriensis.

In order to determine suitable traits for selecting high-wood-yield Korean pine materials, eleven morphological characteristics (tree height, basal diameter, diameter at breast height, diameter at 3 meter height, stem straightness degree, crown breadth, crown height, branch angle, branch number per node, bark thickness, and stem volume) were investigated in a 38-year-old Korean pine clonal trial at Naozhi orchard. A statistical approach combining variance and regression analysis was used to extract appropriate traits for selecting elite clones. Results of variance analysis showed significant difference in variance sources in most of the traits, except for the stem straightness degree, which had a p-value of 0.94. Moderate to high coefficients of variation and clonal repeatability ranged from 10.73% to 35.45% and from 0.06% to 0.78%, respectively. Strong significant correlations on the phenotypic and genotypic levels were observed between the straightness traits and tree volume, but crown breadth was weakly correlated to the volume. Four principal components retaining up to 80% of the total variation were extracted, and stem volume, basal diameter, diameter at breast height, diameter at 3 meter height, tree height, and crown height displayed high correlation to these components (r ranged from 0.76 to 0.98). Based on the Type III sum of squares, tree height, diameter at breast height, and branch number showed significant information to explain the clonal variability based on stem volume. Using the extracted characteristics as the selection index, six clones (PK105, PK59, PK104, PK36, PK28, and K101) displayed the highest Qi values, with a selection rate of 5% corresponding to the genetic gain of 42.96% in stem volume. This study provides beneficial information for the selection of multiple traits for genetically improved genotypes of Korean pine.

Pinus koraiensis, as a keystone tree species, possesses immense economic and ecological value. However, the present cultivation of high-quality seedlings in Pinus koraiensis plantations remains hindered by prohibitively high costs and inadequate technological advancements. Additionally, the species’ prolonged growth cycle and low yield, when compounded by issues such as excessive harvesting, may result in supply constraints. Plant growth regulators (PGRs), a class of naturally occurring or synthetically derived chemical compounds, are capable of modulating plant development and physiology. These regulators exert notable effects by enhancing root proliferation, facilitating lignification, influencing plant architecture, and augmenting yield. Owing to their operational simplicity and relatively low cost, PGR applications hold substantial promise for cultivating Pinus koraiensis seedlings with superior traits. In this study, four-year-old Pinus koraiensis seedlings were employed to evaluate the impacts of three PGRs (paclobutrazol, chlormequat chloride, and diethyl aminoethyl hexanoate), alongside varied application methods (dosage and frequency), on the growth, physiological, and photosynthetic parameters of the seedlings. The findings revealed that treatment with 1.5 g/L paclobutrazol produced the most pronounced effects across a range of indicators. Specifically, this treatment markedly enhanced growth traits (e.g., branch diameter, new shoot length, lateral branch length, aboveground fresh and dry weights, root fresh and dry weights, lateral root dry weight, and number of second-order roots), physiological attributes (e.g., increased superoxide dismutase and peroxidase activities, elevated lignin content, and reduced relative conductivity and malondialdehyde levels), and photosynthetic metrics (e.g., elevated net photosynthetic rate, stomatal conductance, transpiration rate, and maximum net photosynthetic rate), thereby constituting the optimal treatment combination.

Juglans mandshurica is an economically and ecologically valuable species that is used for various construction purposes, making luxurious furniture, as food and sources of medicinal substances and landscaping because of its excellent wood, edible fruits and rich in various types of chemical compounds. In the past few decades, several genetic improvements of J. mandshurica were made, with a focus on the selection of improved varieties and on breeding technology. Many elite provenances and families were selected based on growth traits or wood properties. In recent years, with the increasing demand for high-quality seedlings in Chinese forestry production, the breeding goals of genetic improvement for J. mandshurica were redefined to include other traits, such as fruit yield and contents of medicinal component. However, the improvement processes were still slow due to the long breeding cycle and the limited use of advanced breeding technologies, resulting in the selection of fewer improved varieties. In this review, we summarized the research progresses on genetic improvements of J. mandshurica and other related works, and discussed research gaps and suggested future directions for genetic improvement of the species. The review provides valuable insight for the selection of improved varieties and production of excellent germplasms.

Lonicera caerulea L. has high nutritional and health value, and it is an important emerging small berry tree species. In this study, the morphology and nutrient composition of 60 cultivars were used to analyze and evaluate the diversity of the genus. Morphological analysis showed that the phenotypic traits of different cultivars had significant differences (p < 0.01). The phenotypic coefficient of variation (PCV) of each trait was 12.42%~84.06%, and the coefficient of genetic variation (GCV) was between 7.07%~71.72%. The analysis of nutrient content showed significant differences among the cultivars (p < 0.01). The PCV of each trait was 3.95%~96.10%, and the GCV was 0.13%~32.83%. Based on breeding objectives, cultivars with excellent growth and leaf quantitative traits, fruit quantitative traits and nutrient contents were selected through the method of comprehensive analysis of multiple characters. Traits of the selected varieties were all above average, and specific genetic gain was higher. At the same time, the selection of varieties was carried out according to flowering and fruiting phenology, which provided an indication for the breeding of improved varieties. In this study, growth, leaf and fruit quantitative traits, phenological period and nutrient components of different cultivars provided valuable information for the breeding of improved varieties.

Korean pine (Pinus koraiensis) is an economically valuable species owing to its excellent timber quality and nuts useful for various purposes. But few studies have been made on growth performance, and aspects combining the genetic gain and classification method on phenotypic similarity in the selection process of superior families. Thus, the present study aimed at analyzing the genetic variation and highlight suitable morphological traits for family selection; establishing trait correlations and families’ ordination based on similarities in phenotypic characters, and selecting elite families and suitable parent trees. Full-sib families from 28 crosses established in randomized complete block design from Naozhi orchard in Northeast China were used, and 11 morphological traits were investigated. Significant differences were observed among families for all traits. The traits coefficients of variation ranged from 6.07 to 56.25 % and from 0.029 to 15.213 % in phenotype and genotypic variation, respectively. A moderate level of inherited genetic control was observed (broad sense heritability H2, varied from 0.155 to 0.438). Traits related to stem growth were highly positively correlated to each other whereas crown traits showed a weak correlation with stem traits (Pearson correlation r, ranged from -0.161 to 0.956). Based on multi-trait comprehensive analysis, we selected six elite families and six parents, which resulted in a genetic gain of 5.6 %, 16.9 %, and 36.4 % in tree height, diameter at breast height, and volume, respectively. These results make a theoretical basis for selecting excellent families and establish orchards of Korean pine from improved seeds.
