Plants of the genus Ilex are widespread throughout the world, with its best-known representative being Ilex paraguraiensis from South America. The European species Ilex aquifolium shows similarities in its terpenoid, sugar and phenolic acid profiles. Using aqueous extracts of Ilex aquifolium as a supplement in Wistar rats showed that, despite the lack of caffeine, it had strong hypocholesterolemic effects. In addition, a reduction in oxidative lipid degradation and a decrease in hepatic steatosis in histopathological studies were observed. The results of this study suggest that extracts from the European species Ilex aquifolium may have potential as an alternative treatment for hyperlipidemia.

Progesterone biotransformation is worth studying because of the high industrial value of its derivatives. This study investigated the catalytic ability of the entomopathogenic filamentous fungus strain Isaria farinosa KCh KW1.1 to transform progesterone derivatives: 11α-hydroxyprogesterone, 17α-hydroxyprogesterone, 16α,17α-epoxyprogesterone and pregnenolone. In the culture of Isaria farinosa KCh KW1.1, 11α-hydroxyprogesterone was effectively transformed into only one product: 6β,11α-dihydroxyprogesterone. Transformation of 17α-hydroxyprogesterone gave three hydroxy derivatives: 6β,17α-dihydroxyprogesterone, 12β,17α-dihydroxyprogesterone and 6β,12β,17α-trihydroxyprogesterone. Two products: 6β-hydroxy-16α,17α-epoxyprogesterone and 6β,11α-dihydroxy-16α,17α-epoxyprogesterone, were obtained from the 16α,17α-epoxyprogesterone transformation. We isolated two compounds from the biotransformation medium with pregnenolone: 11α-hydroxy-7-oxopregnenolone and 5α,6α-epoxy-3β,11α-dihydroxypregnan-7,20-dione. In this study, we observed only mono- and dihydroxy derivatives of the tested substrates, and the number of obtained products for each biotransformation did not exceed three.

A rapid ultra-high-performance liquid chromatography method with diode-array (DAD) and tandem mass spectrometric (MS/MS) detection was developed and, for the first time, applied for the determination of potassium iodide in ophthalmic formulations. The approach is based on an indirect derivatization reaction in which iodide is oxidized by hydrogen peroxide to electrophilic iodine species that react with 4-hydroxybenzoic acid, forming 3-iodo-4-hydroxybenzoic acid as a stable and quantifiable product. Key reaction parameters, including oxidant concentration, temperature, and incubation time, were optimized to ensure selective mono-iodination and consistent analytical response. Chromatographic separation was performed on a C18-DE RP column using gradient elution with aqueous ammonium acetate and acetonitrile, while MS/MS detection was carried out in MRM mode under ESI(−) conditions. Both UHPLC–DAD and UHPLC–MS/MS methods were validated according to ICH Q2(R2), demonstrating linearity with r2 ≥ 0.998, recovery values of 97.5–107.1%, and intraday/interday RSD values up to 3.7%. UHPLC–MS/MS provided higher sensitivity (LOD 37.7 ng/mL; LOQ 114 ng/mL), whereas UHPLC–DAD reached LOD and LOQ values of 24.9 and 75.4 µg/mL. Comparative analysis showed that DAD is suitable for routine quantification, while MS/MS allows lower detection limits and improved selectivity. The developed method offers a practical and reliable tool for the quality control of potassium iodide in ophthalmic formulations.
