A total of 636 1-day-old male Cobb chicks were randomly assigned to seven treatments. The chicks were offered feed and water ad libitum throughout the experimental period. The first three groups included different stocking densities of broiler birds (low stocking density, LSD: 23 kg/m2; medium stocking density, MSD: 34 kg birds/m2; and high stocking density HSD: 39 kg birds/m2). The LSD group was considered a control group. The other four groups included MSD or HSD broiler birds supplemented with either Vit E (100 mg/kg DM diet; MSDVE and HSDVE) or Zn (100 mg/kg DM diet; MSDZn and HSDZn) in their basal diet. The main findings indicated that HSD and MSD negatively affected (p < 0.05) all variables under investigation compared with LSD. Compared with LSD, broiler birds in the MSD and HSD groups had lower body weights and higher feed conversion ratios, higher concentrations of blood plasma hormones (triiodothyronine thyroxine and corticosterone), and downregulated expression levels of hepatic growth hormone and insulin-like growth factor-l. In addition, broiler birds stocked at medium or high densities resulted in less economic return and profit. Vit E or Zn supplementation to broiler birds stocked at medium or high densities significantly reversed all adverse effects of HSD (> 23 kg/m2) on growth performance, hormones, and gene expression. It could be recommended that adding Zn at a level of 100 mg/kg per DM diet allows increasing the stocking density of broiler birds from 23 kg/m2 to 34 birds/m2 while maintaining the birds, welfare and economic profit.

Wheat germ oil (WGO) is a well-known product with anti-inflammatory and antioxidant properties. The current study aimed to investigate the impacts of WGO against ethanol-induced liver and kidney dysfunction at the serum, anti-inflammatory, antioxidants and anti-apoptotic signaling pathways. Rats received saline orally as a negative control or WGO in a dose of 1.5 mL/kg (1400 mg/kg body weight orally) for 15 days. The affected group received ethanol 50% v/v 10 mL/kg (5 g/kg) body weight orally once a day for consecutive 15 days to induce hepatorenal injuries in ethanolic non-treated group. The protective group received WGO daily 1 h before ethanol administration. Serum (1.5 mL) from blood was extracted and examined for the changes in biochemical assessments in serum alkaline phosphatase (ALP), alanine aminotransferase (ALT), bilirubin, serum γ-glutamyl transpeptidase (GGT), total protein, serum albumin, butyrylcholinesterase (BChE), total cholesterol (TC), total triglyceride (TG), urea, creatinine, uric acid, potassium (K+), Beta-2 microglobulin (β2M), malondialdehyde (MDA), catalase (CAT), reduced glutathione (GSH), superoxide dismutase (SOD) and aspartate aminotransferase (AST). Kidney and liver homogenate was used to measure MDA, GSH and catalase activities. Quantitative real time PCR (qRT-PCR) was used to express Nrf2 and HO-1 in liver, and NF-kB and kidney injury molecule (KIM-1) in kidneys, which are correlated with oxidative stress and inflammation. Capase-3 and Bcl2 genes were examined using immunohistochemical analysis in the kidney and liver. Ethanol administration induced significant alteration in examined liver and kidney markers (AST, ALT, GGT, ALP, total proteins, urea, creatinine and uric acid). Moreover, alcohol administration decreased antioxidant activities at serum and hepatorenal tissues (GSH, catalase and SOD), while MDA was increased as a tissue degradation marker. Inflammatory cytokines, together with genes of oxidative stress markers (Nrf2 and HO-1), were all affected. At cellular levels, apoptotic marker caspase-3 was upregulated, while antiapoptotic marker B-cell lymphoma 2 (Bcl2), was down regulated using immunohistochemical analysis. Of interest, pretreatment with WGO improved the side effects induced by ethanol on hepatic, renal biomarkers and reversed its impact on serum and tissue antioxidant parameters. Nrf2/HO-1 were upregulated, while NFk-B and KIM-1 were downregulated using real time PCR. Immune reactivities of caspase-3 and Bcl2 genes were restored in the protective group. In conclusion, WGO ameliorated ethanol-induced hepatic and renal dysfunction at the biochemical, molecular and cellular levels by regulating some mechanisms that controls oxidative stress, apoptosis, inflammation and anti-apoptotic pathways.

Background and Aim: Artemisia annua (AA), used as a growth promoter in poultry, lowers feed costs and enhances economic efficiency. This study aimed to assess the impact of varying AA concentrations on broiler chicken growth, gene expression, and profitability. Materials and Methods: Two hundred 1-day-old male Cobb chicks were randomly allocated into four treatment groups, each containing five replicates and 10 birds. The experimental groups consisted of G1 (basal diet), G2 (basal diet with 0.3% AA), G3 (basal diet with 0.6% AA), and G4 (basal diet with 0.9% AA). The birds had continuous access to feed and water throughout the study. The experiment lasted for 42 days. we measured the growth performance (Feed intake, Life weight), carcass traits (weight after slaughter, dressed carcass, heart, gizzard, spleen, giblet and thymus weight), liver and spleen antioxidants (CAT, GSH, SOD), and gene expression of anti-inflammatory and immune- related genes. Results: The primary findings revealed that the addition of 0.6% AA had a positive impact (p < 0.05) on all investigated variables compared with the control and other groups. Dietary supplementation with 0.6% AA led to increased breast, giblet, skeleton, and total yield, and net return compared with the control group. Supplementation with AA exhibited antioxidant, anti-inflammatory, and immunological effects through improved levels of antioxidant superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GSH-Px) in tissue homogenates of the liver and spleen. It also upregulated the relative messenger RNA levels of anti-inflammatory interleukin (IL)-10, SOD, CAT, and GSH-Px, whereas IL-1β and tumor necrosis factor-alpha were downregulated. Conclusion: The study found that AA is a promising replacement for antibiotics in poultry farming as a growth promoter for chickens. 0.6% AA in the broiler diet yielded the best results, striking a balance between superior performance and robust economic benefits. Keywords: Artemisia annua, broiler, carcass traits, economics, immunity, mRNA gene expression.

This research was carried out to evaluate the difference in the mRNA genetic expression of the liver genes as adenine nucleotide translocase (ANT), cytochrome oxidase subunit III (COX III), Insulin like growth factor binding protein 2 (IGFBp2) and Avian uncoupling protein (Av UCP), as well as growth performance include (body weight and body weight gain ) between Cobb broilers and Fayoumi native breeds. 120 females of one-day-old Cobb and Fayoumi chicks had beenused. Chicks were distributed equally in three replicates. the results showed that, expression of adenine nucleotide translocase (ANT) had non-significant difference between cobb and Fayoumi (P > 0.05) but there was significant difference in COX III, IGFBp2 , Av UCP gene expressions and body weight, body weight gain between cobb and Fayoumi breeds.(P<.05). From the obtained results it could be concluded that, variations in the genetic expression levels of the mitochondrial genes (ANT, COX III, Av UCP) and growth-related gene (IGFBp2) related to differences in growth performance of chickens are good biomarkers for selection and improvement of growth traits in chickens.
