(1) Background: To further improve the rapid reproduction and large-scale application of Catalpa bignonioides. (2) Methods: With young softwood cuttings from a 3-year-old C. bignonioides mother plant used as materials, the effects of indole-3-acetic acid(IAA), indolebutyric acid(IBA) and rhizogenic powder-1(ABT-1) growth regulators at different concentrations on cutting indexes and the dynamic changes in endogenous hormone contents during the rooting of the C. bignonioides cuttings were studied. (3) Results: The rooting of C. bignonioides cuttings could be divided into five stages. There were three types of rooting of adventitious roots. IBA treatment resulted in a high rooting rate and beneficial root morphology. The morphological indexes of the cutting roots after treatment with 1000 mg·L−1 IBA had the best overall quality, which was significantly higher than that of the roots in the control (CK) group (p < 0.05). Although the average longest root length (20.51 cm) under ABT-1 was the longest, its overall average rooting rate was slightly lower than that under IBA. The rooting effect under IAA was generally lower than that under IBA and ABT-1. The endogenous hormone content of the cuttings was found to be closely related to rooting; the IAA and zeatin nucleoside (ZR) content was high, and the ratios of IAA/ABA and IAA/ZR were high. The contents of gibberellin3 (GA3) and abscisic acid (ABA) were low, which had a promoting effect on the rooting of the cuttings. (5) Conclusions: All three kinds of auxin can promote rooting and, of the three treatment groups, the rooting effect of cuttings in the IBA treatment group was the strongest, with 1000 mg·L−1 being the optimum concentration.

The AP2/ERF gene family, referring to an exclusive class of transcription factors unique to plants, is involved in various biological processes, including plant growth and responses to environmental stresses like high salt and drought. In this study, the AP2/ERF gene in M. notabilis was comprehensively identified and bioinformatically analyzed based on the genomic data of M. notabilis. 106 members in the MnAP2/ERF gene family were identified in the M. notabilis genome and were categorized into five subfamilies: ERF, AP2, DREB, RAV, and Soloist, with the ERF subfamily representing 80.19% of the total. The MnAP2/ERF gene family was observed to be distributed on six chromosomes of M. notabilis. Members in the MnAP2/ERF gene family exhibited obvious differences in amino acid number, molecular weight, isoelectric point, and other properties. Approximately 68.87% of the MnAP2/ERF proteins were acidic, all exhibiting hydrophilic characteristics. Differences in conserved sequences and arrangement of AP2 domains were observed among distinct subfamilies, with genes in the same subfamily sharing similar conserved domain compositions. There were 47 genes without untranslated regions and 44 genes with two untranslated regions. The upstream functions of promoters were concentrated on light reactions and plant hormones. Evolutionarily, significant structural differences were identified, and 28 MnAP2/ERF gene family proteins could interact with each other. Moreover, 35 family genes were involved in 22 fragment repeat events, and 55 MnAP2/ERF and 84 AtAP2/ERF genes showed collinearity. The expression of the MnAP2/ERF gene family was significantly different in different parts, indicating that these gene family members were involved in different physiological activities. These results established a theoretical foundation for investigating the functional and evolutionary aspects of AP2/ERF gene family genes in M. notabilis, as well as exploring the root morphogenesis of M. notabilis. Additionally, this study contributes to a basis for the improvement of cultivar stress resistance of M. notabilis.

Pears are popular table fruits, grown and consumed worldwide for their excellent color, aroma, and taste. Volatile aroma is an important factor affecting fruit quality, and the fatty acid metabolism pathway is important in synthesizing volatile aromas. Most of the white pear varieties cultivated in China are not strongly scented, which significantly affects their overall quality. Panguxiang is a white pear cultivar, but its aroma has unique components and is strong. The study of the mechanisms by which aroma is formed in Panguxiang is, therefore, essential to improving the quality of the fruit. The study analyzed physiological and transcriptome factors to reveal the molecular network behind volatile aroma formation in Panguxiang. The samples of Panguxiang fruit were collected in two (fruit development at 60, 90, 120, and 147 days, and fruit storage at 0, 7, 14, 21, and 28 days) periods. A total of nine sample stages were used for RNA extraction and paired-end sequencing. In addition, RNA quantification and qualification, library preparation and sequencing, data analysis and gene annotation, gene co-expression network analysis, and validation of DEGs through quantitative real-time PCR (qRT-;PCR) were performed in this study. The WGCNA identified yellow functional modules and several biological and metabolic pathways related to fatty acid formation. Finally, we identified seven and eight hub genes in the fatty acid synthesis and fatty acid metabolism pathways, respectively. Further analysis of the co-expression network allowed us to identify several key transcription factors related to the volatile aroma, including AP2/ERF-ERF, C3H, MYB, NAC, C2H2, GRAS, and Trihelix, which may also be involved in the fatty acid synthesis. This study lays a theoretical foundation for studying volatile compounds in pear fruits and provides a theoretical basis for related research in other fruits.

Volatiles are essential substances that determine distinct fruit flavors and user preferences. However, the metabolic dynamic and molecular modulation models that regulate the overall flavor generation during fruit growth and ripening are still largely unclear for most fruit species. To comprehensively analyze the molecular mechanism and regulation mechanism of aroma accumulation and aroma component formation in Pyrus spp. ‘Panguxiang’ (‘Panguxiang’pear), this study compared pear phenotype, sugars, organic acid content, and the expression of related genes and metabolites amid pear growth and development in Pyrus spp. ‘Panguxiang’. A total of 417 VOCs (4 amines, 19 aromatics, 29 aldehydes, 31 alcohols, 38 ketones, 64 heterocyclic compounds, 89 terpenoids, 94 esters, and 49 others) were found. The potential gene expression patterns were explored by combining transcriptomics and metabolomics, and VOC-associated metabolism and transcriptome data from all samples were integrated during the growth and development period. On this basis, we constructed a colorful model depicting changes in the VOCs and genes throughout pear growth and development. Our findings reveal that terpenoid biosynthesis pathways are the main aroma production pathways during pear growth and development. In addition to providing novel insights into the metabolic control of fruit flavor during growth and development, this study also provides a new theoretical basis for studying aroma metabolites in pears.

ARs plays a crucial role in plant morphogenesis and development. The limited and inefficient rooting of scions poses a significant challenge to the efficiency and quality of clonal propagation of forest trees in silvicultural practices. Building on previous research conducted by our team, we found that applying IBA at a concentration of 1000 mg/L significantly enhanced mulberry rooting. This study aims to uncover the molecular mechanisms underlying this effect by analyzing RNA sequencing data from mulberry phloem before and after treatment with IBA over time intervals of 10, 20, 30, and 40 days. We identified 5226 DEGs, which were then classified into GO terms and KEGG pathways, showing significant enrichment in hormone signaling processes. Using WGCNA, we identified eight co-expression modules, two of which were significantly correlated with the IBA treatment. Additionally, 18 transcription factors that potentially facilitate ARs formation in mulberry were identified, and an exploratory analysis on the cis-regulatory elements associated with these transcription factors was conducted. The findings of this study provide a comprehensive understanding of the mechanisms of ARs in mulberry and offer theoretical support for the discovery and utilization of exceptional genetic resources within the species.
