The present study is focused on the availability of microbial sources capable of producing xylanase, a hemicelluloses-degrading enzyme with multiple modes of action along with specificity, and their real-world applications. For the accumulation of suitable data, article surfing was carried out using multiple search engines viz. Hinari and PubMed; irrelevant and duplicate articles were discarded and articles were summarized in a narrative way herein. This review article was written aiming to bridge the recent research activities with the commercial activities of xylanase going on around the globe. The readers would be able to acknowledge themselves with the basic idea of the hydrolytic enzyme xylanase, their classification representing their different families, presenting the affinity of different families at the structural level, the sources, and the commercial implications that have been going on alone and in combination. The major hemicellulose, Xylan is digested with the help of combination other enzymes such as alpha-amylase, subtilisin, protease, and endo-1,3(4)-β-glucanase along with xylanase. Xylanase has a diverse applications such as pharmaceutical, food and feed, bakery, paper and pulp, textile, and bio-refinery industries. The objective of this review article is to compile microbial sources of this enzyme and its application for betterment of human kind.

Background/Objectives: Propranolol HCl (PPH), a nonselective beta-adrenergic receptor blocker, is employed as an anti-hypertensive, anti-anginal, anti-arrhythmic, and anti-migraine agent. Given its utility in chronic conditions, developing a sustained-release dosage form becomes imperative to optimize therapeutic outcomes while enhancing patient adherence and minimizing side effects. In this study, we employed a widely adopted matrix-based system to develop PPH sustained-release (PPH-SR) matrix tablets, ensuring the uniform dispersion of the drug within the polymeric matrix to regulate its release rate. Methods: Utilizing cellulose-based polymers, specifically HPMC K100M and ethyl cellulose (EC), as matrix formers, nine different formulations were prepared at varying drug-to-polymer ratios. We employed a wet granulation method, followed by compression of the dried granules, to fabricate round-shaped biconvex PPH-SR tablets. Results: Among these different formulations, formulation 2 (F2), comprising 40 mg PPH and 50 mg HPMC K100M (along with other excipients), showed excellent flowability, as evidenced by Carr’s index and angle of repose values of 12.50 and 28.50, respectively. Additionally, the mechanical properties of F2 tablets showed a hardness of 12.34 ± 0.91 KP, an average weight of 200.45 ± 1.87 mg, with a friability of 0.20%, and a content uniformity of 98.36%. Moreover, in vitro release characteristics of F2 tablets demonstrated a sustained-release behavior, with 94.3 ± 10.2% drug release over 24 h. A comparative analysis with marketed tablets yielded similarity and dissimilarity factors of 64 and 8, respectively. Furthermore, the release profile of F2 exhibited a high degree of linearity with the Korsmeyer–Peppas model (R2 of 0.977), showcasing its reliability and predictability. Conclusions: In essence, this in-house developed PPH sustained-release formulation can improve patient adherence, reduce side effects, and improve therapeutic outcomes. These results align with our objective of enhancing the therapeutic efficacy of PPH and affirm the broader relevance of innovative formulation strategies in addressing the challenges of chronic disease management.

Orodispersible tablet is a patient-friendly alternative for delivering the drug to patients having problems with swallowing and sensitive to the bitterness of the drug. This study focuses on taste masking of cetirizine dihydrochloride by applying coating technique. Taste masked orodispersible tablet (ODT) was formulated using superdisintegrants and apply hydrophilic/hydrophopic polymer/lipid coating. Formulations were designed by 22 factorial central composite design, in which stearic acid and compritol 888 ATO were taken as independent variables. The taste masking was evaluated using the small-volume shake flask method, as an alternative to testing with a human panel. The concentration of stearic acid and compritol 888 ATO in the optimized formulation obtained from the Minitab software response optimization was nearly as same as that of formulation F3. Thus, formulation F3 was considered to be formulation of choice. The weight variation, disintegration time, friability, and dissolution were found to be satisfactory. The coating of API with compritol 888 ATO exhibited effective taste-masking properties while maintaining rapid disintegration and acceptable mechanical strength while masking the bitterness of the drug molecule. Taste masking of cetirizine dihydrochloride was successfully achieved using a lipid-based coating approach, with Compritol® 888 ATO and stearic acid as coating materials. Thus, the study successfully developed a taste-masked orodispersible tablet of cetirizine dihydrochloride using a lipid-based coating technique.
