Bats are natural hosts of a wide variety of viruses, including adenoviruses. European bats are known to carry mastadenoviruses categorized as species B (widespread in European Vespertilionidae bats) and whose taxonomy has not been clarified. We examined fecal samples from Vespertilionidae bats (five species) captured in central Russia and found that 2/12 (16%) were positive for mastadenoviruses. The partial genome of the mastadenovirus was assembled from Pipistrellus nathusii, representing the bat adenovirus species B. The complete genome (37,915 nt) of a novel mastadenovirus was assembled from Nyctalus noctula and named BatAdV/MOW15-Nn19/Quixote. Comparative studies showed significant divergence of the Quixote genome sequence from European bat mastadenoviruses, while the only known virus showing low similarity was the isolate WA3301 from an Australian bat, and together they formed a subclade that separated from other BatAdVs. Phylogenetic and comparative analysis of the protein-coding genes provided evidence that Quixote is related to a novel species within the genus Mastadenovirus, provisionally named “K” (as the next available letter for the species). Phylogenetic analyses revealed that some earlier viruses from Western European bats, for which only partial DNA polymerase genes are known, are most likely members of the tentatively named species “K”. Thus, at least two species of mastadenovirus are circulating in bats throughout Europe, from western to eastern areas.

Manufacturing adulteration is the major cause of discrepancies between the declared and actual composition of food products. While high-throughput sequencing (HTS) of DNA barcodes is a promising method to identify adulterants, its practical application is hampered by technical challenges. Food pre-processing and differences in GC composition can lead to unequal amplification or complete loss of DNA barcode components. Consequently, HTS results require independent confirmation using an orthogonal method based on very different physical principles than DNA sequencing. To address this, we evaluated the suitability of a multi-omic approach that coupled DNA barcode HTS analysis with proteomic analysis, to enhance the detection of food fraud in herbal beverages. To resolve discrepancies between genomic and proteomic findings, we employed traditional botanical morphology as an arbiter. Among the samples studied, the combined approach revealed two main adulterations of Epilobium with Lythrum — a substitution potentially hazardous to consumers — as well as several minor substitutions, all confirmed by orthogonal methods. Our findings demonstrate that proteomic analysis provides enhanced confidence for verifying the presence or absence of plant components identified by HTS. However, its effective application is guided by prior sequencing to define specific targets for subsequent proteomic verification. This study established that a multimodal analytical approach is not only beneficial, but essential for the reliable and comprehensive characterization of components in complex plant mixtures.

Bats are natural hosts for a wide variety of viruses and could therefore be the reservoir, source and spillover for several mammalian viruses, including adenoviruses. We examined faecal samples from 12 bats (five species of the family Vespertilionidae) from central Russia (Moscow Region) and found that two of them (16%) were positive for mastadenoviruses. One of the identified viruses was isolated from faeces of Nyctalis noctule and named BatAdV/MOW15-Nn19. The genome of the BatAdV/MOW15-Nn19 was 37,915 bp in length. Comparative studies have shown significant divergence of the nucleotide sequence of the BatAdV/MOW15-Nn19 genome from European mastadenoviruses, the highest genome sequence similarity was to isolate WA3301 from an Australian bat. Phylogenetic analysis, as well as a comparative analysis of the protein coding genes to homologous available from public database, showed evidence of novel species within Mastadenovirus genus (preliminary named «K»). Analysis of the short part of the DNA polymerase gene of BatAdV/MOW15-Nn19 reveals few NCBI records from Europe with high identity to the new virus, i.e. mastadenoviruses related to the new species are circulating in European bats. The partial genome of another mastadenovirus from Pipistrellus nathusii was identified, it represented bat adenovirus species B, which is widely distributed in European Vespertilionidae bats. New data on the genetic diversity of bat adenoviruses may help to better understand how they evolved in mammals and spread across Europe.

A clinical strain of the opportunistic pathogen Serratia rubidaea, a known contaminant of healthcare environments and an emerging cause of invasive infections, is described. The studied isolate, recovered from a nurse’s hand skin swab during routine screening, exhibits a broad profile of antibiotic resistance combined with reduced susceptibility to several disinfectants. Phenotypic susceptibility testing using a tablet-based microdilution and disk diffusion method was employed to determine the minimum inhibitory concentrations (MICs) of antimicrobial agents from different classes, while broth microdilution assays with disinfectants revealed high-level tolerance to widely used agents, including 70% C2H5OH, 3% H2O2, 0.05% polyhexamethylene guanidine (PHMG) and others. Whole-genome sequencing identified multiple resistance-associated determinants, such as chromosome-encoded class C β-lactamase (ampC), several efflux systems (sdeXY, macAB, and emrAB) combined with multicopy tolC, and specific transferases (fos and arnT). Shotgun bottom-up HPLC-MS/MS proteomics confirmed baseline expression of these and other stress-tolerance-related proteins under non-inducing conditions. Taken together, these data underscore the importance of surveillance for Serratia spp. in healthcare facilities to detect strains that combine intrinsic or acquired multidrug resistance with robust survival traits such as disinfectant tolerance and biofilm formation. The present study provides a reference-level phenotypic, genomic, and proteomic characterization of a S. rubidaea clinical isolate, contributing to the understanding of the adaptive potential of this resilient opportunistic pathogen in clinical environments.
