Some glucagon-like peptide-1 receptor agonists (GLP-1 RAs), first used in the treatment of type 2 diabetes mellitus (T2DM), have been approved for the treatment of obesity in patients with or without T2DM (liraglutide—LIR, semaglutide—SEM, and tirzepatide—TIR). Social media had an important influence on the off-label use of GLP-1 RAs for obesity, especially for SEM. We analyzed the Google queries related to SEM to assess people’s interest in this drug. We also investigated the occurrence of adverse drug reactions (ADRs) by searching the EudraVigilance database (EV) for Individual Case Safety Reports (ICSRs) that reported SEM as the suspected drug and performed a descriptive and a disproportionality analysis. The data obtained for SEM were compared to other GLP-1 RAs. SEM had the highest proportions of searches on Google associated with the term “weight loss” and presented the lowest number of severe ADRs, but it also had the highest number of ICSRs reported in EV. Even though no unexpected safety issues have been reported for it until now, SEM has a hi3gh tendency for overdose reports. The most frequent off-label use was reported for SEM and TIR. In order to lower the risks of ADRs, the off-label use should be reduced and carefully monitored.

Glycyrrhiza glabra L., also known as licorice, belongs to the Fabaceae family and is one of the most commercially valuable plants worldwide, being used in the pharmaceutical, cosmetic, and food industries, both for its therapeutic benefits as well as for the sweetening properties of the extract. This study evaluates the phytochemical composition, the biological activities, and the safety profile of a methanolic extract of licorice root (LRE) obtained from Romania. Ten phytocompounds were quantified by the HPLC-DAD-ESI+, the most abundant being the triterpene glycyrrhizin (13.927 mg/g dry extract.), followed by these flavonoids: liquiritin, liquiritigenin-apiosyl-glucoside, and apigenin-rutinoside liquiritigenin. The total phenolic content of the LRE was found to be 169.83 mg gallic acid/g dry extract. (GAE/g d.e.), and the extract showed a maximum of 79.29% antioxidant activity in the 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay. Good antimicrobial activity of the LRE was observed for Gram-negative bacteria, especially for S. pneumoniae and S. pyogenes. The mineral content of the LRE was indicative of the lack of toxicity; heavy metals such as lead, cadmium, arsenic, nickel, and cobalt were below the detection limit. The safety profile of the licorice extract was assessed using the in vivo hen egg test-chorioallantoic membrane (HET-CAM protocol), indicating no irritability, good tolerability, and biocompatibility. The phytochemical and biological characterization of the Romanian licorice root extract reveals a good source of glycyrrhizin and polyphenols with antioxidant and antimicrobial potential, along with a safety profile that may be useful for future therapeutic applications.

Anthocyanins, a group of naturally occurring flavonoid compounds, have garnered increasing attention due to their wide-ranging biological activities that suggest their considerable potential to be utilized not only as natural food colorants but also as functional additives that can enhance food preservation and contribute to the development of health-promoting functional foods. Additionally, their sensitivity to environmental factors such as pH and temperature makes anthocyanins promising candidates for use in intelligent packaging systems, particularly as natural indicators for monitoring food freshness and quality throughout storage and distribution. Despite challenges related to their stability and regulatory acceptance, continued research into anthocyanins remains crucial for advancing sustainable, clean-label food technologies and reducing reliance on synthetic additives. To fully leverage their economic and health potential, it is essential to gain a comprehensive understanding of the various plant sources of anthocyanins, their chemical composition, extraction methods, and roles in different applications. Moreover, integrating anthocyanins into food and intelligent packaging systems presents various technical and regulatory challenges that are also summarized in this review.

Background/Objectives: In order to enhance the biological activity, novel complexes of N-(2-bromo-phenyl)-2-hydroxy-benzamide derivatives and β-cyclodextrin were obtained. Methods: The inclusion complexes were characterized using spectral and thermal analyses. The antimicrobial activity was determined using the disk diffusion agar method, and completed with the minimum inhibitory concentration (MIC) values obtained by the broth microdilution method. The in vitro anti-inflammatory activity was evaluated using the protease inhibition assay. Results: The computed supramolecular architectures of the inclusion complexes showed that the most stable molecular arrangements correspond to the models in which the N-(2-bromo-phenyl)-2-hydroxy-benzamide derivatives are partially included in the cyclodextrin cavity. The antimicrobial screening showed that the compounds were active against Gram-positive bacteria (MIC = 2.5–5.0 mg/mL). Also, the evaluation of the proteinase inhibitory activity showed that the IC50 values of the title compounds (0.04–0.07 mg/mL) were much lower than that of the acetylsalicylic acid (0.4051 ± 0.0026 mg/mL) used as positive control, proving their superior efficiency in inhibiting trypsin activity. Conclusions: The complexation proved to be beneficial for both antimicrobial and anti-inflammatory effects.
