Our aim was to assess the efficacy of adjuvant programmed cell death protein-1 (PD-1) inhibitors and compare the other adjuvant treatments in patients with surgically resected stage III or IV acral melanoma. This study is a multicenter, retrospective analysis. We included 114 patients with stage III or IV acral malignant melanoma who underwent surgery within the past 10 years. We analyzed the effect of adjuvant programmed cell death protein-1 inhibitors on disease-free survival (DFS). The mean follow-up was 40 months, during which 69 (59.5%) patients experienced recurrence. Among the participants, 64 (56.1%) received systemic adjuvant therapy. Specifically, 48.4% received anti–PD-1 therapy, 29.7% received interferon, 14.1% received tezozolomide, and 7.8% received B-Raf proto-oncogene/mitogen-activated protein kinase inhibitors. Patients who received adjuvant therapy had a median DFS of 24 (10.9–37.2) months, whereas those who did not receive adjuvant therapy had a median DFS of 15 (9.8–20.2) months. Multivariate analysis for DFS revealed that the receipt of adjuvant therapy and lymph node metastasis stage were independent significant parameters (P = 0.021, P = 0.018, respectively). No statistically significant difference was observed for DFS between programmed cell death protein-1 inhibitor treatment and other adjuvant treatments. Regarding overall survival (OS), patients who received adjuvant treatment had a median OS of 71 (30.4–111.7) months, whereas those who did not receive adjuvant treatment had a median OS of 38 (16.7–59.3; P = 0.023) months. In addition, there were no significant differences in OS observed between various adjuvant treatment agents (P = 0.122). In our study, we have shown that adjuvant therapy had a positive effect on both DFS and OS in patients with stages III–IV acral melanoma who underwent curative intent surgery. Notably, we found no significant differences between anti–PD-1 therapy and other adjuvant therapies.

Objective Arum dioscoridis is a plant that has been shown to regulate biological activity on oxidation, immune system and inflammation grown in our country. We aimed to investigate the effects of arum dioscoridis in experimental colitis related to acetic acid in rats. Material and Methods In this study 56 Wistar Albino female rats, 6 weeks old, weighing 180- 210 g, were seperated into 7 equal groups with 8 animals in each group. Group 1 is the control one without any administration. An experimental colitis has been created on Group 2. Group 3 is treated with prophylactic oral arum dioscoridis before creating experimental colitis. The rats belongs to Group 4 has been received oral arum dioscoridis treatment with ethanolic solvent after creating experimental colitis. Group 5 was given oral treatment with solvent with DMSO (Dimethyl sulfoxide) after experimental colitis was created. Group 6 rectal treatment with solvent with DMSO after experimental colitis has been created. Group 7 was given a 2-fold oral treatment after experimental colitis. In tissue samples taken from mice, the effects of arum dioscoridis were evaluated macroscopically, histopathologically and biochemically. Results In the colitis group, significant weight loss occurred compared to the treatment groups. Statistically significant improvement was found in macroscopic and microscopic analysis with arum dioscoridis treatment and prophylaxis. In the colitis model which is created with acetic acid, total oxidant status, thiol / disulfide, malondialdehyde, myeloperoxidase levels increased and a decrease in total antioxidant capacity was detected. Conclusion This study showed that arum dioscoridis has positive effects on the colitis model created in rats, and it significantly improves intestinal inflammation especially in groups given by dissolving with ethanol and used for prophylaxis purposes

Purpose: The hemoglobin, albumin, lymphocyte, and platelet (HALP) score is a novel immune nutritional index associated with the prognosis of various malignancies. This study aimed to evaluate the clinical relevance of the HALP score in predicting recurrence, metastasis, and survival outcomes in patients with testicular cancer. Methods: This retrospective study included 131 patients with histologically confirmed testicular cancer who were treated between January 2010 and December 2024. HALP scores were calculated using the baseline laboratory parameters. An optimal cutoff value of 304.56 was identified using receiver operating characteristic (ROC) curve analysis (AUC: 0.820; 95% CI 0.708–0.902; p < 0.001). Patients were stratified into high and low HALP groups based on this threshold value. Survival outcomes were evaluated using Kaplan–Meier, and Cox proportional hazards regression analyses. Results: The median HALP score was 531.1 (IQR: 348.8–728.5). A low HALP score (≤ 304.56) was significantly associated with poorer 1-year overall survival (54.5% vs. 98.2%, p < 0.001) and a 15.6-fold increased risk of death (HR 15.604, 95% CI 2.718–89.645, p = 0.001) based on Cox regression analysis. In the multivariate analysis, a low HALP score (HR: 7.684, p = 0.016), the presence of comorbidity (HR 13.528, p = 0.002), and tunica albuginea invasion (HR 7.255, p = 0.030) were identified as independent predictors of recurrence or metastasis. In addition, the HALP score was significantly associated with disease stage ( p < 0.001), with lower scores more commonly observed in patients with advanced-stage disease. Conclusions: The HALP score was a simple, inexpensive, and effective prognostic biomarker in testicular cancer. A score ≤ 304.56 is independently associated with a higher risk of recurrence, metastasis, and mortality. Incorporating HALP into routine assessments may improve risk stratification and clinical decision-making in this patient population.

Background and Objectives: In early-stage HER2-positive breast cancer, ado-trastuzumab emtansine (T-DM1) has been adopted as the preferred adjuvant approach for patients left with residual invasive disease despite neoadjuvant therapy. The influence of different neoadjuvant regimens on subsequent outcomes in real-world settings remains uncertain. Materials and Methods: From 2019 to 2025, 102 patients treated with adjuvant T-DM1 following surgery after neoadjuvant chemotherapy were retrospectively assessed. Neoadjuvant regimens included doxorubicin plus cyclophosphamide followed by trastuzumab-paclitaxel, doxorubicin plus cyclophosphamide with pertuzumab–trastuzumab–docetaxel, or docetaxel–carboplatin–trastuzumab–pertuzumab. Clinical features, treatment response, survival, and toxicity were evaluated. Results: The mean age of the cohort was 49.7 years, and the majority of patients (80.4%) were aged 40 years or older. Hormone receptor positivity was 82.0%, and invasive ductal carcinoma accounted for 97.1% of cases. Regional responses included 39.2% with axillary pCR despite residual breast lesions, and 5.9% with breast pCR accompanied by axillary disease. Kaplan–Meier analysis demonstrated disease-free survival rates of 100%, 95.2%, and 92.2% at 1, 3, and 5 years, respectively. Adverse events were predominantly grade 1–2, while grade 3–4 toxicities occurred in under 5% of the cohort. Baseline characteristics varied across regimens, reflecting real-world treatment preferences, but survival outcomes remained comparable. Conclusions: Adjuvant T-DM1 was associated with high survival rates and manageable toxicity across different neoadjuvant regimens, underscoring its consistent benefit in routine clinical practice.
