Cancer, which killed ten million people in 2020, is expected to become the world’s leading health problem and financial burden. Despite the development of effective therapeutic approaches, cancer-related deaths have increased by 25.4% in the last ten years. Current therapies promote apoptosis and oxidative stress DNA damage and inhibit inflammatory mediators and angiogenesis from providing temporary relief. Thioredoxin-binding protein (TXNIP) causes oxidative stress by inhibiting the function of the thioredoxin system. It is an important regulator of many redox-related signal transduction pathways in cells. In cancer cells, it functions as a tumor suppressor protein that inhibits cell proliferation. In addition, TXNIP levels in hemocytes increased after immune stimulation, suggesting that TXNIP plays an important role in immunity. Several studies have provided experimental evidence for the immune modulatory role of TXNIP in cancer impediments. TXNIP also has the potential to act against immune cells in cancer by mediating the JAK-STAT, MAPK, and PI3K/Akt pathways. To date, therapies targeting TXNIP in cancer are still under investigation. This review highlights the role of TXNIP in preventing cancer, as well as recent reports describing its functions in various immune cells, signaling pathways, and promoting action against cancer.

Liver diseases are responsible for global mortality and morbidity and are a significant cause of death worldwide. Consequently, the advancement of new liver disease targets is of great interest. Non-coding RNA (ncRNA), such as microRNA (miRNA) and long ncRNA (lncRNA), has been proven to play a significant role in the pathogenesis of virtually all acute and chronic liver disorders. Recent studies demonstrated the medical applications of miRNA in various phases of hepatic pathology. PPARs play a major role in regulating many signaling pathways involved in various metabolic disorders. Non-alcoholic fatty liver disease (NAFLD) is the most prevalent form of chronic liver disease in the world, encompassing a spectrum spanning from mild steatosis to severe non-alcoholic steatohepatitis (NASH). PPARs were found to be one of the major regulators in the progression of NAFLD. There is no recognized treatment for NAFLD, even though numerous clinical trials are now underway. NAFLD is a major risk factor for developing hepatocellular carcinoma (HCC), and its frequency increases as obesity and diabetes become more prevalent. Reprogramming anti-diabetic and anti-obesity drugs is an effective therapy option for NAFLD and NASH. Several studies have also focused on the role of ncRNAs in the pathophysiology of NAFLD. The regulatory effects of these ncRNAs make them a primary target for treatments and as early biomarkers. In this study, the main focus will be to understand the regulation of PPARs through ncRNAs and their role in NAFLD.

Arthritis is a chronic inflammation in joints, which mostly affects the bones. In the global prevalence of joint inflammation, it’s means to be taken it as potential issues. Current treatments mostly targeting the inflammatory cytokines or effector molecules and oxidative stress involved in arthritis as pain relief temporarily. This review has mechanistically demonstrated the type of joint inflammation insights in to arthritis, such as osteoarthritis, gouty arthritis and rheumatoid arthritis to understand the clear background. This review has also highlighted the prevalence, mechanism and mediations used for arthritis, in which various causative agents are used to induce arthritis in preclinical studies that has been collectively elucidated. This review is first to be reported the mechanism of various arthritis causative agents. It pointed out the side effects of clinical medicine used for arthritis and suggested the natural products involved in medication for above-listed conditions. As comparatively natural products are low cost, easily available and beneficial than modern medicines with minimal side effects.

Guar gum is an herbal drug that is used for treating the diabetes and cancer which is also used as a dietary supplement. It has properties as anti-microbial, anti-tumor, and anti-inflammatory. Hyperuricemia or Gout is an acute inflammation that can be found in joints with the indications of redness and swollen knees. The aim of our study is to know the potential effect of Cyamopsis tetragonoloba through in vitro and in silico analysis. The antioxidant activities like catalase, peroxidase, DPPH assay and total phenolic content were evaluated in the aqueous, ethanolic and methanolic extract of Cyamopsis tetragonoloba seed. Ligands of Cyamopsis tetragonoloba seed were obtained from the literature and the 3d structure of the ligands were retrieved from Pubchem online server followed by Corina molecular network. Receptors responsible for causing gouty arthritis were obtained from RCSB protein data bank in the format of 3d structure. Patch dock online serve was used to perform the docking analysis which was then visualized and analyzed in PyMOL. Extract of Cyamopsis tetragonoloba seed shown good antioxidant potential in which aqueous extract was observed to show better antioxidant activity. Docked complex with Hexopyranosyl hexopyranoside has shown better interaction with higher binding residues than the other ligands. Further, in vivo studies can be carried out to known the potential activity of this ligand against gouty arthritis.
