In wheat, stem water soluble carbohydrates (WSC), composed mainly of fructans, are the major carbon sources for grain filling during periods of decreasing photosynthesis or under drought stress after anthesis. Here, in a field drought experiment, WSC levels and associated enzyme activities were followed in different stem segments (peduncle, penultimate internode, lower parts of stem, and sheath) during grain filling. The focus was on two double haploid (DH) lines, DH 307 and DH 338, derived from a Westonia/Kauz cross, two drought-tolerant wheat varieties that follow different drought adaptation strategies during grain filling. The results showed that in irrigated plants, in the period between 20 to 30 days after anthesis (DAA), 70-80% of WSC were fructans. Before and after this period, the fructan proportion varied from 10 to 60%, depending on the location along the stem. Under drought, the fructan proportion changed, depending on genotype and developmental stages. After anthesis, stem fructans accumulation occurred mainly in the peduncle and penultimate internode until 14 DAA in both DH lines, with clear genotypic variation in subsequent fructan degradation under drought. In DH 307 a significant reduction of fructans with a concomitant increase in fructose levels occurred earlier in the lower parts of the stem and the sheath, as compared to DH 338 or other stem segments in both lines. This was associated with an earlier increase of grain weight and thousand grain weight in DH 307. Spatiotemporal analysis of fructan dynamics and enzymatic activities in fructan metabolism revealed that several types of FEHs are involved in fructan remobilization to the grain under drought.

Background: Glycinebetaine, whose biosynthesis could be catalyzed by choline oxidase (COD), is an extremely efficient compatible solute for scavenging oxidative stress-inducing molecules and protecting the photosynthetic system in plants. To study the effects of the codA transgene for choline oxidase on the drought resistance and recovery, a transgenic potato cultivar (SC) bearing codA gene and a non-transgenic (NT) control cultivar were raised in pots under moderate and severe drought stress. The experiment was constituted by a two-day-pretreatment with 20% PEG and a four-day-water stress combined with two-day-recovery treatment. Results: Under the four-day-water stress, plants were provided with normal water condition, 10% or 20% polyethylene glycol. The results of pretreatment showed an expression of codA gene in transgenic potato and an accumulation of glycine betaine (GB); leaf water potential was higher in SC than in NT. In the stress-recovery-treatment, SC showed stronger antioxidant ability, more efficient photosynthetic system, higher chlorophyll content, lower malondialdehyde content and better recovery from water deficit stress than NT. Conclusion: Although this work concentrated on the short-term water stress and recover treatments on transgenic potato plants with the over-expression of CodA gene and its control line. The datas shows that the exogenous codA gene provided potato a stronger drought resistance and recovery ability.

The replacement of winter wheat varieties has greatly contributed to yield improvement worldwide and the progress in enhancing yield has been enormously achieved in China. Two sets of data, production yield from the National Bureau of Statistics of China (http://www.stats.gov.cn/english/) and experimental yield from literature, were used in this study for (1) displaying the increasing patterns of production yield among different provinces from 1978 to 2018 in China (2) exploring the genetic gain in yield and yield relevant traits through the varieties replacement based on experimental yield from 1937 to 2016 in China (3) comparing the yield gap between experimental yield and production yield. The results showed that both the production and experimental yield showed significantly increase trends along with the varieties replacement. For the production yield, the annual yield increase ratio was 1.67% at national scale from 1978 to 2018, varying from 0.96% in Sichuan Province to 2.78% in Hebei Province. In term of experimental yield, there was an annual yield increase ratio at 1.13% from 1937 to 2016 in this study. The yield gap between experimental yield and production yield showed decreasing trends from 1970s to 2010s. Regarding the yield components, the significant increased trend appeared in thousand grain weight, kernel number per spike, and grain number per square meter, while no change showed in spike number per square meter along with the variety replacement. The biomass and harvest index showed consistently significant increased, whereas the plant height was significantly decreased.

Understanding the changes in phenotype resulting from the selection pressure and agronomic adaptation of grain yield provide an indication of the pathways for future increases in grain yield. Six dry land representative winter wheat cultivars (Triticum aestivum L.) released from 1942 to 2004 in the Loess Plateau of China were investigated to determine how the yield components of winter wheat were associated with grain yield at the Changwu Agricultural Research Station during the 2011–2012 and 2012–2013 seasons, using a completely randomized block design with three replicates. Plant height, aboveground biomass, grain yield, and yield components were measured, together with the traits of superior and inferior grains and the pre-anthesis stored dry matter remobilized to the grain was determined. In the relatively wet 2011–2012 season, there was a significant increase in grain yield and aboveground biomass with the year of release, but not in the dry 2012–2013 season. The harvest index (HI) and average grain weight (AGW) increased significantly with the year of release in both cropping seasons. HI and AGW are likely potential traits for improving grain yield of winter wheat in the Loess Plateau. The increase in HI mainly resulted from the decrease in plant height, and the increase in the use of pre-anthesis stored assimilates for grain filling. The increase in AGW mainly resulted from the increase in the proportion of superior grain (SG) and the decrease in the proportion of inferior grain (IG) in the whole spike in both cropping seasons. Depending on the climatic conditions, the different winter wheat cultivars showed different ability to use pre-anthesis stored assimilates. Modern wheat cultivars had higher yield under different rainfall conditions, and high ability to use pre-anthesis stored assimilates to fill the grain than earlier released cultivars. Both, the increase in sink capacity and source availability, should be considered as a strategy for increasing future grain yield in Loess Plateau of China.
