Boron (B)-deficiency is a widespread problem in many crops, including Citrus. MicroRNAs (miRNAs) play important roles in nutrient deficiencies. However, little is known on B-deficiency-responsive miRNAs in plants. In this study, we first identified miRNAs and their expression pattern in B-deficient Citrus sinensis roots by Illumina sequencing in order to identify miRNAs that might be involved in the tolerance of plants to B-deficiency.

MicroRNAs play important roles in the adaptive responses of plants to nutrient deficiencies. Most research, however, has focused on nitrogen (N), phosphorus (P), sulfur (S), copper (Cu) and iron (Fe) deficiencies, limited data are available on the differential expression of miRNAs and their target genes in response to deficiencies of other nutrient elements. In this study, we identified the known and novel miRNAs as well as the boron (B)-deficiency-responsive miRNAs from citrus leaves in order to obtain the potential miRNAs related to the tolerance of citrus to B-deficiency. Seedlings of ‘Xuegan’ [Citrus sinensis (L.) Osbeck] were supplied every other day with B-deficient (0 μM H3BO3) or -sufficient (10 μM H3BO3) nutrient solution for 15 weeks. Thereafter, we sequenced two small RNA libraries from B-deficient and -sufficient (control) citrus leaves, respectively, using Illumina sequencing. Ninety one (83 known and 8 novel) up- and 81 (75 known and 6 novel) down-regulated miRNAs were isolated from B-deficient leaves. The great alteration of miRNA expression might contribute to the tolerance of citrus to B-deficiency. The adaptive responses of miRNAs to B-deficiency might related to several aspects: (a) attenuation of plant growth and development by repressing auxin signaling due to decreased TIR1 level and ARF-mediated gene expression by altering the expression of miR393, miR160 and miR3946; (b) maintaining leaf phenotype and enhancing the stress tolerance by up-regulating NACs targeted by miR159, miR782, miR3946 and miR7539; (c) activation of the stress responses and antioxidant system through down-regulating the expression of miR164, miR6260, miR5929, miR6214, miR3946 and miR3446; (d) decreasing the expression of major facilitator superfamily protein genes targeted by miR5037, thus lowering B export from plants. Also, B-deficiency-induced down-regulation of miR408 might play a role in plant tolerance to B-deficiency by regulating Cu homeostasis and enhancing superoxide dismutase activity. Our study reveals some novel responses of citrus to B-deficiency, which increase our understanding of the adaptive mechanisms of citrus to B-deficiency at the miRNA (post-transcriptional) level.

Tobacco (Nicotiana tabacum L.) is an economically important crop in China. The risk of cadmium (Cd) pollution is increasing, making the mitigation of Cd stress critical for the safe production of tobacco. Silicon (Si) has been demonstrated to enhance plant growth under heavy metal stress. To elucidate the physiological and biochemical mechanisms by which Si alleviates Cd stress in tobacco, this study conducted a hydroponic experiment with three replicates using tobacco variety Yunyan 87 and employed one-way analysis of variance (ANOVA) for statistical analysis. The effects of Si on tobacco growth and the antioxidant defense system under Cd stress were investigated. The results showed that Cd stress significantly inhibited tobacco growth, increased Cd accumulation, and induced oxidative damage. Si treatment alleviated Cd stress in tobacco, increased biomass, reduced Cd concentration in different plant organs, and decreased the Cd translocation factor and bioconcentration factor. Meanwhile, the activities of superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) in tobacco roots and leaves were significantly enhanced, while the activities of ascorbate peroxidase (APX) and glutathione reductase (GR) were also elevated. The accumulation of ascorbic acid (AsA) and glutathione (GSH) increased, and malondialdehyde (MDA) concentration decreased. Overall, these results demonstrate that Si mitigates Cd stress in tobacco by limiting Cd accumulation and transport and by coordinately activating both enzymatic and non-enzymatic antioxidant systems.

The ponkan (Citrus reticulata Blanco cv. Ponkan), an important citrus crop, is increasingly threatened by soil acidification. This study evaluated the efficacy of various soil amendments, including lime alone (L), lime with gypsum and organic fertilizer (LGOF), lime plus K2CO3 (LK), and lime with chicken manure ash (LCMA), in mitigating soil acidification and improving ponkan seedling growth. Surface-applied lime raised topsoil pH and acid buffering capacity while reducing exchangeable Al. However, combined amendments (LGOF, LK, LCMA) more effectively alleviated acidity throughout the soil profile. They significantly increased pH and buffering capacity, decreased exchangeable H and Al in the 20–40 cm layer, and elevated exchangeable base cations (K+, Ca2+, Mg2+). These changes reduced Al content in roots, stems, and leaves, promoted deeper root growth, and increased biomass and nutrient uptake (N, P, K). Physiologically, combined amendments enhanced photosynthetic performance (chlorophyll, Pn, ΦPSII) and increased activity of key metabolic enzymes (Rubisco, SS, SPS, NR, GS), promoting sucrose, starch, and protein accumulation. LK rapidly raised subsoil pH and potassium levels, ideal for K-deficient orchards. LGOF and LCMA improved overall fertility by supplying Ca and Mg, with LGOF additionally enhancing soil structure in poorly structured acidic soils.
