In the modern agricultural landscape, numerous challenges, such as climate change, diminishing arable lands, and the reduction of water resources, represent significant threats. The Mediterranean greenhouse farming model relies on low-input strategies to maximize both yield and quality. Its protected horticulture is essential for the year-round cultivation of high-value crops, ensuring efficient and sustainable production. In the realm of future agricultural strategies, leveraging internet-based approaches emerges as a pivotal factor for real-time and remote control of various agricultural parameters crucial for crop growth and development. This approach has the potential to significantly optimize agronomic inputs, thereby enhancing the efficiency of targeted vegetable production. The aim of the present review is to underscore the challenges related to the intensive greenhouse production systems emphasizing various strategies leading to low-input greenhouse vegetable production. The goal is to promote more sustainable and resource-efficient approaches in the cultivation of greenhouse vegetables. This review highlights several key strategies for optimizing the greenhouse environment, including efficient water management through conservation tillage, drainage water reuse, and selecting the most appropriate irrigation systems and timing. Additionally, light modulation and temperature control—using solar energy for heating and pad-and-fan systems for cooling—are crucial for enhancing both crop performance and resource efficiency. The review also explores low-input agronomical strategies, such as pest and disease control—including solarization and optimized integrated pest management (IPM)—as well as fertilization and advanced growing techniques. These approaches are essential for sustainable greenhouse farming.

Drought is one of the major challenges of global crop production, and its severity is increasing because of climate change. This abiotic stress is an important target for Brassica species, which are generally grown in arid and semi-arid climates. This study was conducted to investigate the effects of water deficit on a set of accessions belonging to the Brassica core collection of the EU H2020 BRESOV project, represented by Brassica oleracea L. crops and Brassica oleracea complex species (n = 9). In particular, the variation in the amount and profile of the glucosinolates (GLSs) compounds was analyzed on the root and the leaf tissues. The plant morphometric traits and GLSs amount and profile were detected for the plants grown in cold greenhouse in Catania (Sicily) during the autumn–winter season for ten weeks. The results showed a wide qualitative and quantitative variation among the Brassica accessions. The GLSs profile varied qualitatively and quantitively among both genotypes and portions of the plants (hypogenous-root and epigeous-leaf). Plants grown under drought stress, for the last two weeks of the growing cycle under consideration, showed a higher amount of GLS in their leaves (190.1 ± 8.9 µmol. g−1 d.w.) compared to their roots (17.3 ± 1.9 µmol. g−1 d.w.). Under water stress conditions, the highest increase in the glucosinolate amount was detected in broccoli (the accession BR1) with 85.4% and in cauliflower (the accession CV1) with 72.8% in the roots and leaves, respectively. Positive correlations were found between the major leaf and root GLSs identified. The selection of chemotypes allows for an important time reduction during the breeding programs after crossing accessions with the specific profiles of glucosinolates.

Next-Generation Sequencing (NGS) techniques have become a cornerstone of molecular diagnostics, enabling high-throughput, parallel analysis of multiple disease-associated genes. Their targeted design allows streamlined interpretation and optimised diagnostic yield, especially in disorders with known genetic heterogeneity. In this review, we provide a comprehensive overview of the clinical application of NGS techniques—targeted gene panels, whole exome sequencing (WES) and whole genome sequencing (WGS)—detailing the methodological workflow and the critical steps involved in their implementation. Particular emphasis is placed on the genes identified through NGS that are implicated in neurodevelopmental, neurodegenerative, psychiatric, neuromuscular, cardiovascular, and metabolic disorders. We also compare the advantages and limitations of panel-based diagnostics versus WES and WGS, and discuss future directions, including the integration of long-read sequencing technologies into multidisciplinary clinical practice. Finally, we consider how these advances may ultimately bridge biomedical research and clinical practise to improve the diagnosis and management of multifactorial diseases.

In the present study, we evaluated for the first time the variability of antioxidant traits of four Brassica wild species: B. incana, B. macrocarpa, B. villosa, and B. rupestris. The content of the main water-soluble antioxidants (phenolics, ascorbic acid, and total biothiols) and the in vitro antioxidant potential (1,1-diphenyl-2-picrylhydrazil (DPPH) and superoxide anion scavenging capacity) were investigated. A total of 28 polyphenolic compounds were identified by LC/MS and quantitated by HPLC/DAD analysis. Kaempferol and quercetin derivatives were the most abundant phenolics compared to hydroxycinnamoyl gentiobiosides. In the ten populations, phenolics ranged from 163.9 to 533.9 mg/100 g dry weight (d.w.), ascorbic acid from 7.6 to 375.8 mg/100 g d.w., and total biothiols from 0.59 to 5.13 mg/100 g d.w. The different classes of phytochemicals were separated using solid-phase extraction at increasing methanol concentrations, and the antioxidant power of fractionated extracts was evaluated. The superoxide anion scavenging activity was significantly correlated to phenolics, particularly to flavonol derivatives, while DPPH was mainly related to ascorbic acid content. The present findings improve the knowledge of the phytochemical composition of Italian Brassica wild species by showing the great diversity of phytochemicals among populations and highlighting their importance as a valuable genetic resource for developing new cultivars with improved bioactive content.

E3 ubiquitin protein ligase encoded by ARIH2 gene catalyses the ubiquitination of target proteins and plays a crucial role in posttranslational modifications across various cellular processes. As prior documented, mutations in genes involved in the ubiquitination process are often associated with autism spectrum disorder (ASD) and/or intellectual disability (ID). In the current study, a de novo heterozygous mutation was identified in the splicing intronic region adjacent to the last exon of the ARIH2 gene using whole exome sequencing (WES). We hypothesize that this mutation, found in an ASD/ID patient, disrupts the protein Ariadne domain which is involved in the autoinhibition of ARIH2 enzyme. Predictive analyses elucidated the implications of the novel mutation in the splicing process and confirmed its autosomal dominant inheritance model. Nevertheless, we cannot exclude the possibility that other genetic factors, undetectable by WES, such as mutations in non-coding regions and polygenic risk in inter-allelic complementation, may contribute to the patient's phenotype. This work aims to suggest potential relationship between the detected mutation in ARIH2 gene and both ASD and ID, even though functional studies combined with new sequencing approaches will be necessary to validate this hypothesis.

Drought tolerance of Brassica crops can be genetically improved by establishing plant ideotypes with improved yield responses associated with agronomic traits and biochemical markers. The objective of this study was to compare 20 Brassica oleracea L. accessions grown under two different water treatments (100% and 35% reintegration of evapotranspiration by irrigation) to select potential tolerant genotypes for organic cultivation based on several agronomic and biochemical parameters measured in response to drought stress. Significant differences were registered for the genotype and the irrigation regime and for their interaction (p < 0.0001 ***). A principal component analysis was performed to summarize the correlations among the analyzed phytochemicals and the stressed and not stressed genotypes and highlighted the importance of the antioxidant compounds as stress biomarkers. The present results showed that drought significantly reduces growth parameters and increases the amount of ascorbic acid and polyphenols compared to the irrigated control. Additionally, the results show that antioxidant metabolism increased by drought in some genotypes while others maintained a good biomass production by increasing the value of growth parameters considered. Based on the average sum of ranks (ASR) of morpho-physiological and biochemical parameters, the genotypes CR, CC, and BH were determined to be the most drought tolerant, whereas CI5, BU, and CV1 were determined to be the most susceptible. Due to the potential of these genotypes, further molecular and cellular research will be carried out to identify the genetic marker associated with the water stress response.
