Nephrotoxicity is one of the limiting factors for using doxorubicin (DOX). Honey, propolis, and royal jelly were evaluated for their ability to protect against nephrotoxicity caused by DOX. Forty-two adult albino rats were divided into control groups. The DOX group was injected i.p. with a weekly dose of 3 mg/kg of DOX for six weeks. The DOX plus honey treated group was injected with DOX and on the next day, received 500 mg/kg/day of honey orally for 21 days. The DOX plus royal jelly treated group was injected with DOX and on the following day, received 100 mg/kg/day of royal jelly orally for 21 days. The DOX plus propolis treated group received DOX and on the following day, was treated orally with 50 mg/kg/day of propolis for 21 days. The DOX plus combined treatment group received DOX and on the following day, was treated with a mix of honey, royal jelly, and propolis orally for 21 days. Results confirmed that DOX raised creatinine, urea, MDA, and TNF-α while decreasing GPX and SOD. Damages and elevated caspase-3 expression were discovered during renal tissue’s histopathological and immunohistochemical studies. Combined treatment with honey, royal jelly, and propolis improved biochemical, histological, and immunohistochemical studies in the renal tissue. qRT-PCR revealed increased expression of poly (ADP-Ribose) polymerase-1 (PARP-1) and a decline of Bcl-2 in the DOX group. However, combined treatment induced a significant decrease in the PARP-1 gene and increased Bcl-2 expression levels. In addition, the combined treatment led to significant improvement in the expression of both PARP-1 and Bcl-2 genes. In conclusion, the combined treatment effectively inhibited nephrotoxicity induced by DOX.

There is a huge demand for novel anticancer agents with fewer side effects compared to current therapies. Pitaya, or dragon fruit, is a reservoir of potent anticancer compounds. This research aimed to analyze the phytochemical components of Hylocereus undatus pulp and peel extracts using LC-MS and GC-MS, and to investigate the in vitro effects of both extracts against cancer (breast, MCF-7, and colon, Caco-2) and normal (lung; WI-38 and breast; MCF-10A) cell proliferation using the MTT assay. The apoptosis potential of the anticancer effects was also evaluated using flow cytometry, RT-PCR, and Western blot. The total phenolic and flavonoid contents in the peel extract were significantly higher than those in the pulp extract. Compared to the flavonoid and phenolic acid standards, the LC-MS analysis revealed the presence of nine compounds, which were represented as 84.32 and 5.29 µg/g of the flavonoids and 686.11 and 148.72 µg/g of the phenolic acids in the peel and pulp extracts, respectively. Among the identified compounds, chlorogenic acid, caffeic acid, ferulic acid, and rutin were found at the highest concentration in both plant extracts. Both extracts displayed cytotoxic activity against MCF-7 and Caco-2 cancer cells after 48 h of treatment at IC50 values ranging from 14 to 53 μg/mL with high selective indices against normal WI-38 and MCF-10A cell lines. The increase in apoptosis was revealed by the overexpression of p53, BAX, and caspase-9 and the downregulation of antiapoptotic Bcl-2 mRNA and protein expressions. The results indicate that H. undatus extracts can be a plant source for cancer therapy.

Five bacterial isolates (B-CP5- B-CP6 - B-CP7- B-CP8- B-CP9) were isolated from pesticides- contaminated soil in Egypt. The capability of these isolates to degrade chlorpyrifos was investigated using enrichment mineral salt (MS) medium containing chlorpyrifos. Two different PCR-based techniques, RAPD-PCR and PCR-RFLP for amplified16S rRNA fragment were used to conduct genetic fingerprinting and obtain specific molecular markers for the studied isolates. The isolates exhibited substantial growth in mineral salt medium supplemented with 100-300 mg/L chlorpyrifos as a sole source of carbon and energy. Based on their morphological, cultural and biochemical characters, the isolates have been identified as Pseudomonas stuzeri, Enterobacter aerogenes, Pseudomonas pseudoalcaligenes, Pseudomonas maltophila andPseudomonas vesicularis respectively. Pseudomonas stuzeri was the most potent degrader strain. Five specific markers for this strain were determined. The highest genetic similarity was observed between CP8 and CP7 (66%), while the lowest genetic similarity was detected between CP8 and CP6 (37%). All isolates had the same pattern after digestion of 16S rRNA amplified fragment with two restriction enzymes (EcoRI and AluI) except Enterobacter aerogenes, which generate two monomorphic bands at 420 and 130 bp, respectively. In conclusion, the strain Pseudomonas stuzeri could be used to clean up the areas contaminated with Chlorpyrifos. Obtained molecular markers might be used for identifying and tracking the most potent bacterial isolate. The used PCR techniques represent a powerful tool and could be used for rapid typing of this strain. Key words: Pesticide, chlorpyrifos, biodegradation, 16S rRNA, RAPD-PCR,Pseudomonas sp.

The present research was aiming to study In-Silico the effect of Glycyrrhizic Acid ammonium salt (GAS) and Salicylic acid (SA) on the coat protein of potato leafroll virus (PLRV). In addition, in-vitro studying the effect of (GAS NPs) and Salicylic acid (SA NPs) nanoparticles at concentrations 0.15, 0.30, 1.25 and 2.5 mM, respectively, to control, decline or reduce the presence of PLRV in potato plants Solanum tuberosum L. selena. (GAS NPs) and (SA NPs) were applied in the MS medium at concentrations 0.15, 0.30, 1.25 and 2.5 mM, respectively. Results revealed that, enhancement or decline the PLRV according to the initiation of specific pathways. The expression level of Kinase 3 gene increased significantly due to the two used concentrations of GAS NPs. While the expression of callose gene was upregulated significantly in response to treatment of PLRV infected plant with (GAS NPs) with concentration (0.30 mM). Treatment with (SA NPs) caused upregulation significance only of callose gene at (2.5 mM) concentration. The molecular modeling results of used compounds (glycyrrhizic acid ammonium salt and salicylic acid) showed highest score of binding and the best rms define value with a very good binding mode and perfect interactions with amino acids of the three subunits (A, B and C) forming the protein coat of leaf roll virus. Glycyrrhizic acid ammonium salt and salicylic acid nanoparticles could be perfect solution to produce potato plant free virus in-vitro. Further larger studies are needed to investigate the role of the studied compounds in vivo.

Premature ovarian failure (POF) is described as a loss of oocytes and the absence of folliculogenesis and is considered an adverse effect of chemotherapeutic drugs, which leads to infertility. Subsequently, the existing inquiry was achieved by exploring the potential suspicious influences of lemon peel extract (LPE), and resveratrol (RES) on cyclophosphamide (CPA) induced-POF. The results showed that CPA-induced POF significantly decreased serum estradiol (E2) and progesterone levels, along with a considerable rise in serum luteinizing hormone (LH) and follicle-stimulating hormone (FSH) levels. Moreover, CPA administration to rats significantly increased the serum level of Malondialdehyde (MDA) and significantly lowered the levels of reduced glutathione (GSH) and superoxide dismutase (SOD); in addition, it increased nuclear factor kappa B (NF-κB) levels, tumor necrosis factor-α (TNF-α), as well as cyclooxygenase 2 (COX-2) with the spread expression of inducible nitric oxide synthase (iNOS) mRNA levels and caspase-3 (Casp3) levels in ovarian tissues versus the control rats. However, treatment with LPE and RES suppressed the triggering of NF- κB pathways, evidenced by a considerable reduction in Casp3 & iNOS mRNA expression level and significant ameliorative effects in all evaluated parameters, as confirmed by the histological and immunohistochemical investigation when comparing the model group. In overall findings, both lemon peel extract and resveratrol can mitigate the adverse effects of CPA-induced POF. Most crucially, its combination therapy is a promising pharmacological agent for this disease.
