The present study provides, for the first time, a physicochemical and biochemical characterization of the redox processes associated with the ripening of Solanum dulcamara L. (bittersweet) berries. Electron Paramagnetic Resonance Spectroscopy (EPRS) and Imaging (EPRI) measurements of reactive oxygen species (ROS) were performed in parallel with the tissue-specific metabolic profiling of major antioxidants and assessment of antioxidant enzymes activity. Fruit transition from the mature green (MG) to ripe red (RR) stage involved changes in the qualitative and quantitative content of antioxidants and the associated cellular oxidation and peroxidation processes. The skin of bittersweet berries, which was the major source of antioxidants, exhibited the highest antioxidant potential against DPPH radicals and nitroxyl spin probe 3CP. The efficient enzymatic antioxidant system played a critical protective role against the deleterious effects of progressive oxidative stress during ripening. Here, we present the EPRI methodology to assess the redox status of fruits and to discriminate between the redox states of different tissues. Interestingly, the intracellular reoxidation of cell-permeable nitroxide probe 3CP was observed for the first time in fruits or any other plant tissue, and its intensity is herein proposed as a reliable indicator of oxidative stress during ripening. The described noninvasive EPRI technique has the potential to have broader application in the study of redox processes associated with the development, senescence, and postharvest storage of fruits, as well as other circumstances in which oxidative stress is implicated.

As a functional extraction medium, natural deep eutectic solvents (NADESs) can dissolve various plant metabolites. Those solvents provide untapped potential for creating novel green extracts with distinctive phytochemical signatures and unique biological activities. This is particularly relevant given the rising need for eco-friendly and sustainable skin care products. The main aim of this work was to optimize the most efficient natural deep eutectic solvents (NADESs) for extracting bioactives from 18 medicinal herbs applicable to the cosmetic industry. Selection of the most potent herbs involved assessing their conventional extracts for tyrosinase inhibition, antioxidant activity, and keratinocyte cytotoxicity. Moreover, we analyzed the phenolic profile using ultra-high-pressure liquid chromatography/mass spectrometry (UHPLC/MS) and spectrophotometric assays such as total phenolic (TPC) and flavonoid content (TFC). Using the COSMO-RS method, we modeled the solubility of 12 phenolics in 64 virtual NADESs and selected the 7 most promising ones for further experimental validation. NADESs, including betaine-urea, betaine-proline, and betaine-lysine, were computationally chosen and demonstrated the highest levels of TPC and antioxidative capacity, as confirmed by in vitro assays. The proposed combination of NADES herbal extracts represents a promising natural constituent for the cosmetic industry.

Frankincense resin (Boswellia serrata), native to arid regions of India, the Middle East, and parts of Africa, has been highly valued for its medicinal properties. This study evaluated the antimicrobial potential of methanolic extracts of Boswellia serrata resin against Staphylococcus aureus, Pseudomonas aeruginosa, and Listeria monocytogenes. High-performance thin-layer chromatography (HPTLC) coupled with bioautography identified bioactive zones, while Liquid Chromatography–Mass Spectrometry (LC-MS) quantified the phenolic and terpenoid compounds. The cytotoxicity was assessed on HaCaT human keratinocyte cells to evaluate the safety for dermatological applications. The results demonstrated significant antibacterial activity, particularly against S. aureus and L. monocytogenes. The bioautograms revealed that samples from central and southern Serbia showed the highest antimicrobial effect against the tested bacterial strains. The active compounds included 11-keto-β-boswellic acid (up to 3733.96 μg/g), gallic acid (110.93 μg/g), and naringenin (53.13 μg/g). Cytotoxicity assays confirmed non-toxic effects at 10 µg/mL, with sample 6 enhancing the keratinocyte viability by 137%, while higher concentrations (50 µg/mL) showed variable cytotoxicity. These findings highlight the potential of B. serrata resin as a natural antimicrobial agent, particularly against antibiotic-resistant pathogens. Its therapeutic applicability in pharmaceutical and cosmetic formulations is promising provided that dosing ensures a balance between efficacy and safety.
