Bisphenol A (BPA) is a ubiquitous plasticizer which is absorbed by ingestion and dermal contact; propyl paraben (PPB) inhibits the microbiome and extends the shelf life of many personal care products, whereas triclosan (TCS) is commonly found in antiseptics, disinfectants, or additives. In this work, Caenorhabditis elegans was used as a biological model to assess the toxic effects of BPA, PPB, and TCS. The wild type strain, Bristol N2, was used in bioassays with the endpoints of lethality, growth, and reproduction; green fluorescent protein (GFP) transgenic strains with the hsp-3, hsp-4, hsp-16.2, hsp-70, sod-1, sod-4, cyp-35A4, cyp-29A2, and skn-1 genes were evaluated for their mRNA expression through fluorescence measurement; and quick Oil Red O (q ORO) was utilized to stain lipid deposits. Lethality was concentration-dependent, while TCS and PPB showed more toxicity than BPA. BPA augmented worm length, while PPB reduced it. All toxicants moderately increased the width and the width–length ratio. BPA and PPB promoted reproduction, in contrast to TCS, which diminished it. All toxicants affected the mRNA expression of genes related to cellular stress, control of reactive oxygen species, and nuclear receptor activation. Lipid accumulation occurred in exposed worms. In conclusion, BPA, PPB, and TCS alter the physiology of growth, lipid accumulation, and reproduction in C. elegans, most likely through oxidative stress mechanisms.

Lepidium meyenii (maca) is a Peruvian nutraceutical plant, whose hypocotyl has a variety of colors ranging from black to white. The black and red varieties of maca have been the most studied since their extracts are associated with effects such as increased sperm count, decreased glucose levels, reversal of prostatic hyperplasia, among others. However, the properties related to reduction of oxidative stress, metabolic diseases and anti-aging have not yet been confirmed. The aim was to evaluate the effects of aqueous and ethanolic extracts, obtained from spray-dried hydroalcoholic extract of hypocotyl of black and red maca (Lepidium meyenii), on mortality, growth, reproduction, lipid accumulation and the expression levels of genes related to oxidative stress and heat shock, in the in-vivo model, Caenorhabditis elegans (C. elegans), using different concentrations. The results showed that maca extracts were not toxic to the model at concentrations below 100 mg L-1. However, higher concentrations caused high mortality, growth disturbances, oxidative stress and lipid accumulation. Black maca extracts increased the reproduction of C. elegans by increasing the number of offspring in C. elegans, both in aqueous and ethanolic extracts. On the other hand, ethanolic extracts produced an increase in the expression of genes related to oxidative stress, indicating a mild stressor behavior of the same. C. elegans represents an established model for evaluating the biological properties of nutraceutical plants of biological interest and can be used in the search for antioxidant activity of L. meyenii (hypocotyl), as well as it can be used in future studies to identify some metabolites involved in each biological property and to understand the biochemical and molecular mechanisms involved in these properties.

Background: One of the pathways involved in liver regeneration processes is TWEAK/Fn14 (tumor necrosis factor-related weak inducer of apoptosis/fibroblast growth factor-inducible 14), which has been proposed to act directly and selectively on hepatic progenitor cells; however, its role in the regeneration of steatotic liver metabolic dysfunction associated fatty liver disease has not been fully elucidated. Objective: To evaluate the behavior of Fn14 and its ligand TWEAK, as well as cellular stress signals as biochemical cues for possible liver regeneration in MAFLD. Materials and methods: A prospective study was carried out where the behavior of Fn14 and its ligand TWEAK, as well as cellular stress signals were observed as biochemical indications of a possible liver regeneration in a condition of tissue damage caused by excessive lipid accumulation. The expression of TWEAK, Fn14 and heat shock proteins in hepatic steatosis of non-alcoholic origin was assessed using immunohistochemistry and western blotting. Results: The histological classification of the tissues under study corresponded to microvesicular steatosis. We report a high level of expression of heat shock proteins in the cytoplasm. The expression of TWEAK and Fn14 in liver tissue affected by lipid accumulation was localized in the cytoplasm of hepatocytes, showing a higher intensity of reactivity for Fn14 compared to its ligand TWEAK. Conclusion: The expression of TWEAK/Fn14 axis was positive suggesting reactivity of the signaling pathway in metabolic dysfunction associated fatty liver disease.
