In this study, we investigated the role and relationship between the cytokine profile and protective vitamin D by measuring their serum levels in COVID‐19 intensive care unit patients with severe illnesses. A total of 74 patients were included in our study. Patients were divided into two groups. Patients in the COVID‐19 group (n = 31) and individuals without a history of serious illness or infection were used as the control group (n = 43). The serum concentrations of interleukin‐1 (IL‐1), IL‐6, IL‐10, IL‐21, and tumor necrosis factor‐α (TNF‐α) were measured by enzyme‐linked immunosorbent assays. Levels of serum vitamin D were detected with Liquid chromatography–mass spectrometry methodologies. TNF‐α, IL‐1, IL‐6, IL‐10, IL‐21, and vitamin D levels were measured in all patients. The serum cytokine levels in the COVID‐19 patient group were significantly higher (151.59 ± 56.50, 140.37 ± 64.32, 249.02 ± 62.84, 129.04 ± 31.64, and 123.58 ± 24.49, respectively) than control groups. Serum vitamin D was also significantly low (6.82 ± 3.29) in patients in the COVID‐19 group than the controls (21.96 ± 5.39). Regarding the correlation of vitamin D with cytokine levels, it was significantly variable. Our study shows that COVID‐19 patients are associated with lower serum vitamin D and higher pro‐inflammatory cytokines associated with increased virus presence. Our data provide more evidence of the anti‐inflammatory effect of vitamin D on COVID‐19 patients and the protective effects of vitamin D on risk were demonstrated.
Purpose: This study aimed to investigate the effects of metabolic and bariatric surgery (MBS) on the corneal and anterior segment parameters of the eye. Materials and Methods: In this study, the anterior chamber depth, volume, angle and central corneal thickness of obese patients who underwent laparoscopic sleeve gastrectomy surgery were prospectively examined using Pentacam topography before and six months after surgery. Results: A total of 112 eyes of 56 individuals, including 26 males with a mean age of 38.88 ± 7.00 years and 30 females with a mean age of 40.94 ± 7.76 years, were evaluated (p = 0.310). Six months following MBS, the average body mass index (BMI) in men decreased from 45.5 kg/m2 to 34 kg/m2 (p < 0.001), and the BMI decreased from 47 kg/m2 to 33 kg/m2 (p < 0.001) in women. Central corneal thickness and intraocular pressure decreased significantly in both men and women. The anterior chamber depth, volume and angle significantly increased six months after surgery (p < 0.001 for all parameters). A statistically significant negative correlation was observed between postoperative BMI and anterior chamber depth, volume and angle in both sexes (p < 0.001 for all). Conclusion: This study revealed that anterior segment values, intraocular pressure, and corneal thickness may change after MBS. Caution should be exercised when evaluating for cataracts and refractive surgery after MBS. Graphical
Background/Objectives: Propofol and Thiopental are widely used anesthetic agents, yet their cumulative and high-dose effects on hepatic metabolism remain insufficiently characterized. This study aimed to evaluate the impact of supra-therapeutic concentrations of these agents on carnitine and amino acid metabolism in AML12 hepatocytes, with a focus on their toxicometabolic profiles. Methods: AML12 mouse hepatocytes were exposed to escalating concentrations (2.5–500 µg/mL) of Propofol and Thiopental to assess cytotoxicity. IC50 values (~255 µg/mL for both) were determined, and two high-dose concentrations (100 µg/mL and 200 µg/mL) were selected for metabolic profiling. Cell viability was assessed via the MTT assay. Intracellular carnitine and amino acid levels were quantified using LC-MS/MS. Statistical analyses included one-way ANOVA with post hoc tests, unpaired t-tests, and effect size estimations (Cohen’s d). Results: Propofol significantly suppressed carnitine metabolism in a dose-dependent manner, with a 79% reduction in free carnitine (C0), indicative of impaired mitochondrial β-oxidation. Thiopental, however, preserved or partially restored several acylcarnitines, including C16:1. While both agents reduced intracellular amino acid levels, 200 µg/mL Thiopental partially restored key metabolites such as glutamine, alanine, and histidine. Propofol exhibited broader metabolic suppression. Effect size analysis further confirmed the stronger inhibitory impact of Propofol. Conclusion: Although the concentrations used exceed typical clinical plasma levels, they may reflect prolonged or high-dose exposure scenarios observed in ICU settings. The findings highlight distinct toxicometabolic signatures for each agent and underscore the utility of metabolite profiling in modeling anesthetic-induced hepatic stress and guiding anesthetic selection in vulnerable populations.
Aim: We aimed to investigate the relationship between the presence of spondylolisthesis and Modic changes (MCs) by using lumbar magnetic resonance imaging (MRI). Methods: The study included 139 adult patients aged 18-65 years with lumbar spondylolisthesis detected on lumbar MRI. Demographic characteristics such as age, gender, and levels and grades of spondylolisthesis, and presence of lumbar MCs/types were recorded. The findings were compared between two groups including grade 1 and grade 2 spondylolisthesis. Results: Grade 1 and 2 spondylolisthesis groups were similar for age (p=0.787), sex (p=0.076), listhesis' level (p=0.268) and direction (p=0.280). The presences of pars spondylolysis, disc space narrowing, and MCs were significantly higher in patients with grade 2 spondylolisthesis than those with grade 1 (94.4% vs 64.1%), (97.2% vs 83.5%), and (100% vs 83.5%), respectively (all p
