Methotrexate (MTX) is widely used for the treatment of various types of cancer; however, it has drawbacks such as low solubility, lack of selectivity, premature degradation, and side effects. To solve these weaknesses, a hydrogel with the ability to contain and release MTX under physiological conditions without burst release was synthesized. The hydrogel was fabricated with a poly( ɛ -caprolactone)- b -poly(ethylene glycol)- b -poly( ɛ -caprolactone) (PCL–PEG–PCL) triblock copolymer, synthesized by ring-opening polymerization. The characterizations by proton nuclear magnetic resonance spectroscopy and Fourier-transform infrared spectrometry confirmed the copolymer assembly, whereas the molecular weight analysis validated the PCL 2000 –PEG 1000 –PCL 2000 structure. The copolymer aqueous solution exhibited sol–gel phase transition at 37°C and injection capacity. The hydrogel supported a load of 1,000 μg MTX·mL −1 , showing a gradual and sustained release profile of the drug for 14 days, with a delivery up to 92% at pH 6.7. The cytotoxicity of the MTX-loaded hydrogel was performed by the methyl thiazole tetrazolium assay, showing a mean inhibitory concentration of 50% of MCF-7 cells (IC 50 ) at 43 µg MTX·mL −1 .

The structural modification of biopolymers is a current strategy to develop materials with biomedical applications. Silk fibroin is a natural fiber derived from a protein produced by the silkworm (Bombyx mori) with biocompatible characteristics and excellent mechanical properties. This research reports the structural modification of silk fibroin by incorporating polyaniline chain grafts through a one-pot process (esterification reaction/oxidative polymerization). The structural characterization was achieved by 1H-NMR and FT-IR. The morphology was studied by scanning electron microscopy and complemented with thermogravimetric analysis to understand the effect of the thermal stability at each step of the modification. Different fibroin silk (Fib): polyaniline (PAni) mass ratios were evaluated. From this evaluation, it was found that a Fib to PAni ratio of at least 1 to 0.5 is required to produce electroactive polyaniline, as observed by UV-vis and CV. Notably, all the fibroin-g-PAni systems present low cytotoxicity, making them promising systems for developing biocompatible electrochemical sensors.

Inert polymeric implants must evolve to enhance their biological interactions with host tissue, triggering positive cellular responses and promoting tissue bonding and integration. Poly-ether-ether-ketone (PEEK) is widely used as an implant material; however, its inert nature results in limited biological interactions. Various surface modification techniques have been investigated to enhance its bioactivity and overall biological performance. In this study, the PEEK surface was bioactivated through a chemical treatment involving two steps: surface activation using low-pressure oxygen plasma, followed by biofunctionalization with phosphate and calcium ions. Comprehensive surface characterization by contact angle, scanning electron microscopy (SEM), X-ray photoelectron spectroscopy (XPS), and Fourier-transform infrared (FT-IR) confirmed the effect of plasma and the ionic surface incorporation. The biological response was evaluated through cell viability, adhesion, and proliferation in NIH/3T3 fibroblasts and HOS osteoblasts, and the results indicated the efficacy of the surface modifications. Therefore, the proposed treatments provide an efficient strategy to improve the biological performance of PEEK-based implants.
