This study describes the development of a new combined polysaccharide-matrix-based technology for the immobilization of Lactobacillus rhamnosus GG (LGG) bacteria in biofilm form. The new composition allows for delivering the bacteria to the digestive tract in a manner that improves their robustness compared with planktonic cells and released biofilm cells. Granules consisting of a polysaccharide matrix with probiotic biofilms (PMPB) with high cell density (>9 log CFU/g) were obtained by immobilization in the optimized nutrient medium. Successful probiotic loading was confirmed by fluorescence microscopy and scanning electron microscopy. The developed prebiotic polysaccharide matrix significantly enhanced LGG viability under acidic (pH 2.0) and bile salt (0.3%) stress conditions. Enzymatic extract of feces, mimicking colon fluid in terms of cellulase activity, was used to evaluate the intestinal release of probiotics. PMPB granules showed the ability to gradually release a large number of viable LGG cells in the model colon fluid. In vivo, the oral administration of PMPB granules in rats resulted in the successful release of probiotics in the colon environment. The biofilm-forming incubation method of immobilization on a complex polysaccharide matrix tested in this study has shown high efficacy and promising potential for the development of innovative biotechnologies.

In this study, we aimed to compare the performance of conventional PCR and real-time PCR assays as screening methods for identification of three frequent, clinically significant Salmonella serovars in Kazakhstan. We determined the diagnostic efficacy of three molecular methods for detection of S. enterica subsp. enterica and typing S. Typhimurium, S. Enteritidis, and S. Virchow. A total of 137 clinical samples and 883 food samples were obtained in Almaty in 2018–2019. All tests showed high analytical specificity for detecting S. enterica and its corresponding serovariants (100%). The sensitivity of real-time PCR for each of the tested targets was 1–10 microbial cells and in conventional PCR 10–100 microbial cells. The trials with conventional PCR and real-time PCR had a diagnostic efficacy (DE) of 100% and 99.71%, respectively. The DE of real-time PCR and conventional PCR for detecting S. Enteritidis and S. Typhimurium was 99.90%, while the DE of conventional PCR and real-time PCR for detecting S. Virchow was 99.31% and 99.80%, respectively. The RAPD-PCR analysis of the genomic DNA of Salmonella enterica showed the genetic kinship of S. Enteritidis isolates, and the genetic heterogeneity of S. Typhimurium and S. Virchow isolates. Thus, the developed methods can be considered as alternatives to classical serotyping using antisera.

Plant growth-promoting rhizobacteria (PGPRs) colonise the rhizosphere and root surfaces, enhancing crop development through a variety of mechanisms. This study evaluated microbial strains isolated from Triticum aestivum L. for key plant growth-promoting traits, including indole-3-acetic acid (IAA) production, phosphate and zinc (Zn) solubilisation, nitrogen (N2) fixation, and antifungal activity. Among 36 isolates, 3 (AS8, AS23, AS31) exhibited strong growth-promoting potential. IAA production, citrate assimilation, carbohydrate fermentation, and catalase activity were observed to a comparable extent among the selected strains. AS8 showed the highest protease, lipase, and amylolytic activity, while AS23 demonstrated superior phosphate and Zn solubilisation. Notably, AS31 emerged as the most promising multi-trait isolate, exhibiting the highest levels of IAA production, N2 fixation, antifungal activity against five phytopathogens (Fusarium graminearum, F. solani, F. oxysporum, Pythium aphanidermatum, and Alternaria alternata), potentially linked to its hydrogen sulphide (H2S) production, and cellulolytic activity. Molecular identification based on 16S rRNA gene sequencing revealed the isolates as Stenotrophomonas indicatrix AS8, Pantoea agglomerans AS23, and Bacillus thuringiensis AS31. Seed germination assays confirmed the plant growth-promoting efficacy of these PGPR strains, with vigour index increases of up to 43.4-fold. Given their positive impact on seed germination and significant Zn-solubilising abilities, the selected strains represent promising candidates for use as bio-inoculants, offering a sustainable and eco-friendly strategy to enhance agricultural productivity in nutrient-deficient soils. Future research should validate the efficacy of these PGPR strains under pot conditions to confirm their potential for practical agricultural applications.

Nanocomposites based on bacterial cellulose (BC), hydroxyapatite (HAp), and MXene represent a promising class of biomaterials for regenerative medicine and photothermal technologies due to their combination of biocompatibility, mechanical stability, electrical conductivity, and strong infrared absorption capability. However, the high sensitivity of MXene phases and the cellulose matrix to physicochemical impacts requires careful selection of sterilization approaches that provide sterility without substantial loss of key functional properties. This study presents a comparative evaluation of nine sterilization methods – autoclaving, dry heat, ethylene oxide treatment, γ-irradiation, electron beam (E-beam) irradiation, ultraviolet irradiation, ozonation, supercritical CO₂ treatment, and cold plasma – applied to BC/HAp/MXene nanocomposites. Sterility was assessed by direct plating on nutrient media; sterilization efficiency was calculated based on microbial growth. Complete sterility was achieved for all methods except UV (50%) and CO₂ treatment (90%). The effects of sterilization on material structure and properties were analyzed using SEM imaging, porosity measurement, mechanical testing, electrical conductivity assessment, and photothermal activity evaluation. Thermal treatment and ozonation caused the most pronounced structural damage and reduction in mechanical strength. Radiation-based methods induced moderate changes, whereas ethylene oxide, supercritical CO₂, and cold plasma treatment resulted in minimal morphological degradation. Cold plasma and EtO preserved up to 94–98% of the initial conductivity and photothermal response, while thermal treatment led to significant loss of functional performance due to thermo-oxidative degradation of MXene. Comprehensive analysis demonstrated that cold plasma and ethylene oxide provide the best balance between sterility and preservation of structural–functional properties of BC/HAp/MXene nanocomposites. These methods are the most promising for preparing such materials for biomedical applications.
