A general report on the use of the Allium test as cytotoxicological and genotoxicological assay is proposed, with particular emphasis about the standardization of the test in several common applications. The intraspecific variation in Allium cepa has been overlooked, as in most investigations no mention is made about origin and denomination of the onion cultivar used. A standardization of the used material would allow a better generalization of the results, since we cannot be sure that all cultivars would give the same response. A more frequent use of transmission electron microscopy (TEM) investigation is proposed. Even if relatively time consuming and not available in all laboratories, it may help to better understand the mechanism of cytotoxicity, since many morphological characters may appear similar but be arisen from different processes observable only with TEM. About statistical testing, tests other than chi-squared may be used in case of a lower amount of data. The most commonly used statistical tests are the parametric tests ANOVA and Student’s t, and the non-parametric tests Kruskal–Wallis and Mann–Whitney U, for analysis of variance. Tests should be used also to assess the minimal sample dimension for obtaining significance, since data collection (microscope observation) appears to be one of the main bottle necks of the test. Also the use of the Allium test for testing liposomes and other nanovectors for drug delivery is proposed, in order to assess the cytotoxicity of these types of medium and the possible increase in cytotoxicity of the associated drug.

The present study aims to evaluate the potential for the pollution of the environment by two herbicides (quizalofop-p-ethyl and cycloxydim), using the Allium test. The species in question is Allium cepa (onion, 2n = 16), one of the most common plant indicators of environmental pollution. The working method consisted of obtaining the meristematic roots of Allium cepa and their treatment with herbicides at three different concentrations (0.5%, 1%, and 1.5%) for each herbicide for 24 h, for comparison with an untreated control. The results obtained from the cytological study indicated a strong cytotoxic and genotoxic effect for both herbicides, but especially for quizalofop-p-ethyl, where the mitotic index decreased from 30.2% (control) to 9.6% for the variant treated with 1.5% herbicide. In this case, a strong mitodepressive effect was shown by a highly significant percentage (35.4%) of chromosomal aberrations and nuclear alterations: stickiness, fragments, C-mitosis, lobulated nucleus, micronuclei, and nuclear erosion. The mitodepressive effect as well as the percentage of chromosomal aberrations increased with a higher herbicide concentration. The obtained results suggest the strong potential for pollution of the two herbicides, particularly at concentrations higher than 0.5%; therefore, we recommend caution in their use to avoid undesirable effects on the environment.

Nowadays, green synthesized nanoparticles (NPs) are extensively investigated to explore their biological potential. They are being explored to treat different infectious and cancerous diseases. Therefore, the current study was designed to evaluate the cytotoxic and genotoxic effects of biosynthesized silver nanoparticles (AgNPs) from the medicinal plant Moringa oleifera on breast cancer (MCF-7) and HUVEC (human umbilical vein endothelial cells) cell lines. M. oleifera-mediated AgNPs were synthesized from the M. oleifera extract (MOE) and then characterized through the use of a scanning electron microscope (SEM), X-ray diffraction (XRD) and UV–vis spectrophotometer. Biosynthesized AgNPs and MOE were employed on MCF-7 and HUVEC cell lines to evaluate their cytotoxic and genotoxic effects. More cytotoxic effects were observed by AgNPs and MOE on MCF-7 cell lines. The IC50 for biosynthesized AgNPs was found to be 5 μg/mL. DNA damage was also observed by the MOE and AgNPs on MCF-7 cell lines. However, non-significant DNA damage was observed by MOE and AgNPs on HUVEC cell lines. The findings of the current study revealed the cytotoxic and genotoxic effects of biosynthesized AgNPs on MCF-7 cell lines. However, these AgNPs were considered safe for normal HUVEC cell lines.

Flubendiamide (FLB) is an insecticide that is commonly employed to control pests on a variety of vegetables and fruits, with low toxicity for non-target organisms. However, due to its widespread use, the environmental risks and food safety have become major concerns. In this study, the toxicity potential of FLB was studied in the model organisms, Allium cepa and Drosophila melanogaster. The cyto-genotoxic effects of FLB on the root growth, mitotic index (MI), chromosomal aberrations (CAs) and deoxyribonucleic acid (DNA) damage in A. cepa root meristematic cells were investigated using the root growth inhibition Allium test and Comet assays. FLB caused CAs in the form of disturbed ana-telophase, chromosome laggards, stickiness, anaphase-bridge and polyploidy depending on the concentration and the exposure time. The toxicity and genotoxicity of FLB at various doses (0.001, 0.01, 0.1 and 1 mM) on D. melanogaster were investigated from the point of view of larval weight and movement, pupal formation success, pupal position, emergence success and DNA damage, respectively. FLB exposure led to a significant reduction of the locomotor activity at the highest concentration. While DNA damage increased significantly in the FLB-treated onions depending on the concentration and time, DNA damage in the FLB-treated D. melanogaster significantly increased only at the highest dose compared to that which occurred in the control group. Moreover, to provide a mechanistic insight into the genotoxic and locomotion-disrupting effects of FLB, molecular docking simulations of this pesticide were performed against the DNA and diamondback moth (DBM) ryanodine receptor (RyR) Repeat34 domain. The docking studies revealed that FLB binds strongly to a DNA region that is rich in cytosine-guanine-adenine bases (C-G-A) in the minor groove, and it displayed a remarkable binding affinity against the DBM RyR Repeat34 domain.

The aim of this work was to study how certain applied cropping systems (conventional systems differentiated by fertilization level or sowing season and subsistence farming) influence yield, quality, productivity elements, and morphological characters in a collection of Romanian and foreign wheat cultivars. The following indicators were evaluated: productive potential (yield), quality (test weight, protein content, wet gluten content, deformation index, sedimentation index, and gluten index), as well as other elements that determine yield (number of ears/square meter, thousand kernel weight, number of grains/ear, and weight of grains/ear) and plant height. The results show that the cropping systems influenced all the elements studied except the thousand-kernel weight. The only characteristics influenced by higher nitrogen fertilization were test weight, protein content, wet gluten content, deformation index, and gluten index. The superiority of a delayed conventional system was shown by the number of grains/wheat ear and the deformation index. Protein content was differentiated between the conventional and the subsistence system, but especially between the low-input and the conventional system. Nitrogen supply is the most important factor for determining wheat productivity and grain quality.
